This invention pertains to solubility enhanced polymers and methods, kits and compositions which enhance the aqueous solubility of said polymers. One set of preferred methods, kits and compositions embody or utilize phosphorous containing synthons and are most useful for modulating the solubility of synthetic nucleic acids and synthetic nucleic acid analogs. A second set of preferred methods, kits and compositions are most useful for modulating the aqueous solubility of peptides, other polyamides and most preferably peptide nucleic acid (PNA) polymers.

A method for improving the solubility of synthetic polymers selected from the group consisting of a nucleic acid, a nucleic acid analog, a peptide, a polyamide, a chimera and a linked polymer; the method comprising reacting a polymer, a functional group of a subunit of a polymer or a synthesis support upon which a synthetic polymer is assembled, with one or more simple branched compositions or synthons which when linked to the polymer improve the solubility of the polymer or minimize or eliminate polymer self-aggregation, wherein the simple branched composition links to the polymer to form a modifying moiety that is achiral. 2. The method of claim 1, wherein the simple branched composition links to the polymer to form a modifying moiety that is uncharged and non-nucleophilic. 3. The method of claim 1, wherein the simple branched composition has the formula: 4.
