Provided are methods of applying biological compositions, that is, autologous bioadhesive sealant compositions containing one or more biological agents, to an individual, wherein all the blood components used in preparing the composition are derived from the patient who is to receive the biological composition. In one embodiment, the method comprises obtaining a whole blood sample from an individual; forming an inactive platelet rich plasma from the whole blood sample; mixing a biological agent into the inactive platelet rich plasma; obtaining thrombin from the whole blood sample; mixing the thrombin into the inactive platelet rich plasma to form a biological composition; and applying the biological composition to the individual.

The invention claimed is: 1. A method of producing an autologous biological composition consisting of: forming an autologous platelet rich plasma; forming an autologous platelet poor plasma; separating each of the autologous platelet rich plasma and the autologous platelet poor plasma into a respective first portion and a respective second portion; subsequent to the separating, obtaining autologous thrombin from one of the first and second portions of the autologous platelet poor plasma; and combining a defined volume of the obtained autologous thrombin with one of the first and second portions of the autologous platelet rich plasma; producing the autologous biological composition as a result of the combining; wherein time for producing the autologous biological composition is dependent on the defined volume of the obtained thrombin combined with one of the first and second portions of the autologous platelet rich plasma such that the more defined volume of the obtained thrombin is added the quicker the time for producing the autologous biological composition. 2. The method of claim 1 wherein the autologous platelet poor plasma contains a calcium chelating agent. 3. The method of claim 2 wherein the calcium chelating agent is sodium citrate. 4. The method of claim 1, wherein obtaining the autologous thrombin includes adding calcium ions to the autologous platelet poor plasma. 5. The method of claim 1, wherein the autologous platelet poor plasma contains heparin. 6. The method of claim 1, wherein the autologous platelet poor plasma is formed by centrifuging a sample of whole blood. 7. The method of claim 1, wherein the autologous platelet rich plasma is formed by centrifuging a sample of whole blood. 8. The method of claim 1, wherein obtaining autologous thrombin comprises activation of at least a portion of the coagulation pathway. 9. The method of claim 1, wherein the autologous platelet rich plasma and the autologous platelet poor plasma are formed from a single sample of whole blood. 10. The method of claim 1, wherein obtaining autologous thrombin includes placing the autologous platelet poor plasma in a container comprising an activator that activates blood coagulation. 11. The method of claim 1, wherein obtaining autologous thrombin includes placing the autologous platelet poor plasma in a container comprising a surface that activates blood coagulation. 12. The method of claim 1, wherein obtaining autologous thrombin includes placing the autologous platelet poor plasma in a container comprising glass. 13. A method of applying a biological composition to an individual, consisting of: forming a platelet rich plasma from blood taken from the individual; forming a platelet poor plasma from blood taken from the individual; separating each of the platelet rich plasma and the platelet poor plasma into a respective first portion and a respective second portion; subsequent to the separating, obtaining thrombin from one of the first and second portions of the platelet poor plasma; combining a defined volume of the thrombin obtained from the platelet poor plasma into one of the first and second portions of the platelet rich plasma; forming the biological composition as a result of the combining; and applying the biological composition to the individual; wherein formation time of the biological composition is dependent on the defined volume of the obtained thrombin combined with one of the first and second portions of the platelet rich plasma such that the more defined volume of the obtained thrombin is added the quicker the formation time. 14. The method of claim 13, wherein applying the composition results in a barrier against blood loss from a vessel. 15. The method of claim 13, wherein the composition is applied to a needle hole. 16. The method of claim 13, wherein obtaining the thrombin includes adding calcium ions to the platelet poor plasma. 17. The method of claim 13, wherein the platelet poor plasma is formed by centrifuging the blood. 18. The method of claim 13, wherein the platelet rich plasma is formed by centrifuging the blood. 19. The method of claim 13, wherein obtaining thrombin includes activating at least a portion of the coagulation pathway. 20. The method of claim 13, wherein the platelet rich plasma and the platelet poor plasma are formed from a single sample of blood taken from the individual. 21. The method of claim 13, wherein obtaining thrombin includes placing the autologous platelet poor plasma in a container comprising an activator that activates blood coagulation. 22. The method of claim 13, wherein obtaining thrombin includes placing the autologous platelet poor plasma in a container comprising a surface that activates blood coagulation. 23. The method of claim 13, wherein obtaining thrombin includes placing the autologous platelet poor plasma in a container comprising glass. 24. The method of claim 20, wherein the single sample of blood comprises whole blood. 25. An autologous biological composition prepared by a method consisting Of: forming a platelet rich plasma from blood taken from an individual to whom the composition is to be applied; forming a platelet poor plasma from blood taken from the individual; separating each one of the platelet rich plasma and the platelet poor plasma into a respective first portion and a respective second portion; subsequent to the separating, obtaining thrombin from one of the first and second portions of the platelet poor plasma; and combining a defined volume of the thrombin with one of the first and second portions of the platelet rich plasma; preparing the autologous biological composition as a result of combining; wherein preparation time of the autologous biological composition is dependent on the defined volume of the obtained thrombin combined with one of the first and second portions of the platelet rich plasma such that the more defined volume of the obtained thrombin is added the quicker the preparation time. 26. The composition of claim 25 wherein the forming of platelet poor plasma includes adding an anticoagulation agent to the blood taken from the individual. 27. The composition of claim 26 wherein the anticoagulation agent is a calcium chelating agent. 28. The composition of claim 25 wherein obtaining thrombin includes adding calcium ions to the platelet poor plasma. 29. The composition of claim 25 wherein the platelet poor plasma is formed by centrifugation. 30. The composition of claim 25 wherein the platelet rich plasma is formed by centrifugation. 31. The composition of claim 25, wherein obtaining thrombin includes activation of at least a portion of the coagulation pathway. 32. The composition of claim 25, wherein obtaining thrombin includes placing the platelet poor plasma in a container comprising an activator that activates blood coagulation. 33. The composition of claim 25, wherein obtaining thrombin includes placing the platelet poor plasma in a container comprising a surface that activates blood coagulation. 34. The composition of claim 25, wherein obtaining thrombin includes placing the platelet poor plasma in a container comprising glass. 35. A method of producing an autologous biological composition consisting of: centrifuging blood removed from an individual to whom the composition is to be applied to form a platelet rich plasma and a platelet poor plasma; separating each one of the platelet rich plasma and the platelet poor plasma into a respective first portion and a respective second portion; adding calcium ions to one of the first and second portion of the platelet poor plasma; subsequent to the separating, obtaining thrombin from one of the first and second portions of the platelet poor plasma containing calcium ions, wherein thrombin is formed via activation of at least a portion of the coagulation pathway; and combining a defined volume of the thrombin with one of the first and second portions of the platelet rich plasma; producing the composition as a result of the combining; wherein time for producing the composition is dependent on the defined volume of the obtained thrombin combined with one of the first and second portions of the platelet rich plasma such that more defined volume of the obtained thrombin is added the quicker the time for producing the composition; and adding an agent to the platelet rich plasma prior to producing the composition wherein the agent is selected from the group consisting of: analgesic compounds, antibacterial compounds, antibiotics, antifungal compounds, anti-inflammatories, antiparasitic compounds, antiviral compounds, enzymes, enzyme inhibitors, glycoproteins, growth factors, hormones, steroids, glucocorticosteroids, immunomodulators, immunoglobulins, minerals, neuroleptics, proteins, lipoproteins, tumoricidal compounds, tumorstatic compounds, toxins and vitamins.
