The present invention relates to methods of modulating mammalian stem cell and progenitor cell differentiation. The methods of the invention can be employed to regulate and control the differentiation and maturation of mammalian, particularly human stem cells along specific cell and tissue lineages. The methods of the invention relate to the use of certain small organic molecules to modulate the differentiation of stem or progenitor cell populations along specific cell and tissue lineages, and in particular, to the differentiation of embryonic-like stem cells originating from a postpartum placenta or for the differentiation of early progenitor cells to a granulocytic lineage. Finally, the invention relates to the use of such differentiated stem or progenitor cells in transplantation and other medical treatments.

1. A method of treating an individual in need of granulocytes comprising: contacting mammalian CD34+ or CD133+ hematopoietic stem cells with 4-(amino)-2-(2,6-dioxo(3-piperidyl))-isoindoline-1,3-dione or 3-(4-amino-1-oxo-1,3-dihydro-isoindol-2-yl)-piperidine-2,6-dione, under conditions under which said hematopoietic stem cells differentiate, wherein said contacting increases the number of CFU-GM and decreases the number of BFU-E differentiated from said stem cells relative to the numbers of CFU-GM and BFU-E differentiated from said hematopoietic stem cells not contacted with said compound; andadministering said differentiated cells to said individual. 2. The method of claim 1, wherein said individual is human. 3. The method of claim 1, wherein said individual has neutropenia. 4. The method of claim 1, wherein said individual has chronic granulomatous disease. 5. The method of claim 1, wherein said administering causes an increase in the number of CD15+ granulocyte progenitors in said individual. 6. The method of claim 1, wherein said individual has an infection. 7. The method of claim 6, wherein said individual has a chronic infection. 8. The method of claim 6, wherein said infection is bacterial neonatal sepsis. 9. The method of claim 6, wherein said infection is neutropenia-associated infection. 10. The method of claim 9, wherein said individual is a cancer patient suffering a neutropenia-associated infection. 11. The method of claim 6, wherein said individual is a bone marrow transplant recipient.
