The present invention provides molecular constructs and methods for use thereof, including constructs including heterologous miRNA recognition sites, constructs for gene suppression including a gene suppression element embedded within an intron flanked on one or on both sides by non-protein-coding sequence, constructs containing engineered miRNA or miRNA precursors, and constructs for suppression of production of mature microRNA in a cell. Also provided are transgenic plant cells, plants, and seeds containing such constructs, and methods for their use. The invention further provides transgenic plant cells, plants, and seeds containing recombinant DNA for the ligand-controlled expression of a target sequence, which may be endogenous or exogenous. Also disclosed are novel miRNAs and miRNA precursors from crop plants including maize and soy.

A method of providing a 22-nucleotide engineered mature miRNA to a plant cell, comprising transcribing, in a plant cell, a DNA encoding an engineered miRNA precursor, wherein said DNA encoding an engineered miRNA precursor comprises a nucleotide sequence derived from the sequence of a native miR167 precursor selected from the group consisting of SEQ ID NOs:6, 7, 8, and 9 by: replacing one or more nucleotides at the location of said mature miRNA in said native miR167 precursor with nucleotides complementary to a portion of a target sequence, andmaintaining the position and number of mismatches in the stem portion of the fold-back structure of said native miR167 precursor by altering, as needed, one or more nucleotides at the location of the miRNA*, andleaving unchanged the nucleotides other than those at positions at the location of the mature miRNA and miRNA*;whereby a 22-nucleotide engineered mature miRNA is provided to said plant cell. 2. The method of claim 1, wherein said target sequence is an endogenous plant gene, or a transgene in a plant, or an endogenous gene of a pest or pathogen of a plant. 3. The method of claim 1, wherein said target sequence is coding sequence, non-coding sequence, or both coding and non-coding sequences. 4. The method of claim 1, wherein said target sequence is selected from the group consisting of a microRNA (MIR) gene, a fatty acid desaturase or acyl-ACP thioesterase gene, and a zein gene. 5. The method of claim 1, wherein said native miR167 precursor encodes a mature miRNA with the RNA sequence corresponding to SEQ ID NO:4. 6. The method of claim 1, wherein said DNA encoding an engineered miRNA precursor is provided to said plant cell by stably transforming said plant cell with a recombinant construct comprising said DNA encoding an engineered miRNA precursor. 7. The method of claim 1, wherein said DNA encoding an engineered miRNA precursor is provided to said plant cell by transient expression in said plant cell of said DNA encoding an engineered miRNA precursor. 8. A plant cell in which a 22-nucleotide RNA molecule is provided by the method of claim 1, wherein the plant cell comprises the DNA encoding an engineered miRNA precursor of claim 1. 9. The plant cell of claim 8, wherein said DNA encoding an engineered miRNA precursor is provided to said plant cell by stably transforming said plant cell with a recombinant construct comprising said DNA encoding an engineered miRNA precursor. 10. The plant cell of claim 8, wherein said plant cell is a cell of a crop plant. 11. The plant cell of claim 8, wherein said plant cell is in a seed. 12. The plant cell of claim 8, wherein said 22-nucleotide engineered mature miRNA directs in-phase production of trans-acting siRNAs.
