Aloesin은 일반행동, 중추신경계, 심장순환계, 호흡기계, 소화기계, 운동신경계, 면역계, 체온에 미치는 영향 등을 관찰한 일반약리 시험 및 급성, 아급성, 유전독성 시험 결과에서 매우 안전한 화합물로 규명됨. Aloesin 계열 후속물질로 도출된 10종의 화합물들 중, 7-O-methylaloesin은 aloesin과 비교할 때 항산화 작용 및 rat primary cortical cell culture를 이용한 산화성 세포독성으로부터의 신경보호 효능이 우수하고, 국소 허혈성 뇌손상 동물모델에서도 신경보호 효능을 나타냄. 한편 손바닥선인장 및 치자를 연구한 결과, 21종의 화합물이 순수하게 분리되었으며, 그 중 6종의 화합물들은 자연계에서 처음 분리되었음. 분리 화합물들의 항산화 작용 및 배양 신경세포에서의 신경보호 효능을 검색한 결과, KYS 50182(quercetin 3-methyl ether)는 1) 다양한 항산화 검색시스템에서 모두 강력한 항산화 효능을 나타내었으며 2) 배양 신경세포를 사용한 검색에서도 신경세포 보호효과가 우수하며, 3) 일시적 뇌허혈 백서모델에서도 허혈유발 30분 후 10 mg/kg 용량으로 단회 정맥 투여한 결과, 경색용적 및 부종율 모두 대조군에 비해 약 50%의 감소를 나타냄으로서 뚜렷한 조직학적 신경보호 효과는 물론 신경행동학적 회복 효과를 나타내었음. 또한, 4)KYS 50182의 정맥투여 급성독성 실험결과 LD50는 150 mg/kg 이상으로 나타나 적어도 15배 이상의 safety margin을 가지는 것으로 예측됨. 따라서 KYS 50182를 활성성분으로 포함하고 있는 손바닥선인장의 추출물은 후속 개발연구를 진행하면 뇌졸중 치료제나 신경보호용 식품의약으로의 개발 가능성이 매우 높은 것으로 사료됨.
