- Investigated the anti-hepatosteatotic effect of new lead compounds in vitro in hepaticsteatosis model systems.
- Established a NASH mouse model, fed a 60% high-fat diet (HFD) to demonstrate the anti-fibrotic effect of new lead compounds, which has anti-oxidant and anti-inflammatory properties i
- Investigated the anti-hepatosteatotic effect of new lead compounds in vitro in hepaticsteatosis model systems.
- Established a NASH mouse model, fed a 60% high-fat diet (HFD) to demonstrate the anti-fibrotic effect of new lead compounds, which has anti-oxidant and anti-inflammatory properties in vivo.
- Investigated gene expression in HFD induced NAFLD mice by using affymetrix genechips. We selected candidate genes, which have gene expression change in HFD-induced NASH mouse model.
_ We confirmed our selected new marker genes of NASH in the mouse model by treatment with new lead compounds.
New marker genes demonstrated change in mRNA expression revel. These genes will play a role as new NASH markers.
- We investigated the anti-hepatosteatotic effect of new lead compounds in NAFLD in vitro model.
- We investigated cell signal pathways of new lead compounds. The mRNA and protein expression was reduced in NAFLD by treating new lead compounds. However, the result exhibited preventive effect of NASH to fibrosis progression in NASH mouse model by new lead compounds treatment.
- Camphor and oleuropein were significantly enhanced biomarkers of NAFLD by reducing the levels of hepatic lipid profiles and plasma biomarkers in liver injury.
- Oleuropein and piperine both were significantly reduced in levels of hepatic and plasma lipid and were significantly enhanced by relation with NAFLD and metabolic diseases.
- Therapeutic effects of oleuropein or piperine against hepatic steatosis were exhibited through up-regulated expressions of molecules involved in adiponectin-AMPK signaling pathways which lead to decrease lipogenesis and enhance fatty acid oxidation.
- Improvement of hepatic steatosis by these compounds was also associated with increased expression of molecules involved with insulin signaling cascades in the liver.
- We confirmed that therapeutic effects of piperine were exhibited by direct SIRT1 activator in hepatic steatosis.
- ASXL1 and ASXL2 are opposite to each other, which has the anti-hepatosteatotic effect.
- We established assay methods for activity of PPARα , LXRα , AMPK and SIRT1, which are important for regulation of lipid accumulation. In addition, activity of those factors was prevented by lead compounds.
- We analyzed changes of proteins expression related with ASXL1/2 in NASH mouse model.
- We have developed a liver specific ASKL1 knockout mice and investigated accumulation of lipid at variable dietary condition.
