. 1차년도는 계란을 섭취 후 경정맥 캐뉼라가 부착된 쥐의 간에서 새롭게 합성․분비되는 콜레스테롤과 중성지방에 관한 glycerolipids의 분할기작을 동위원소(cholesteryl [1-14C]oleate)표식에 의한 생체모니터링 기법으로 규명하였다. 실험 Ⅰ[계란 섭취 쥐에서 생체 지질대사의 생화학적 기작]과 실험 Ⅱ[계란 섭취후 경정맥캐뉼라 부착 쥐에서 생체 지질의 분할기작 모니터링]로 구분하여 수행하였다. SDS 랫드에게 6주동안 실험사료를 급여하면서 난황을 경구투여 하여 실험 Ⅰ을 수행하였고 실험 Ⅰ

SDS 랫드에게 6주동안 실험사료를 급여하면서 난황을 경구투여 하여 실험 Ⅰ을 수행하였고 실험 Ⅰ 종료 후 경정맥캐뉼라를 설치한 후 생체 지질분할의 대사기작 모니터링으로 생체 지질대사의 분할기작을 규명하였다. 그 결과, 난황을 경구투여한 동물에서 혈액 지질이 감소하였고 HMG CoA reductase 활성억제 및 스테로이드 배설 촉진과 관련한 지질의 흡수억제 대사경로에 의한 기작으로써 계란의 섭취가 혈액 내 유해지질을 낮춤과 동시에 간 기능관련 효소 수준을 정상으로 유지해준다는 새로운 사실을 확인하였다. 본 연구에서 밝혀낸 또 다른 새로운 사실은 계란의 섭취가 지질의 흡수를 억제함과 동시에 산화를 가속화하여 줌으로써 생체조직 내 지질축적을 낮추고, 간에서 새롭게 합성된 Glycerolipid 분할에서 중성지방 분비를 낮춤과 동시에인지질 분비를 증가시키고 14CO2 발산을 증가시키기 때문에 혈액 지질을 낮출 수 있으며, 간에서 합성 분비되는 중성지방과 인지질의 대사적 분할능력을 조절하는 기작에 의해 혈액 지질을 낮춘다는 점이었다. 2차년도는 흰쥐에게 임시 전 암컷과 숫컷으로부터 분만 시 까지, 분만 후부터 이유 시 까지, 그리고 이유후부터 6주 동안 장기적으로 계란을 함유하는 동일한 실험사료를 급여하여 수행하였다. 실험Ⅰ[차세대 정상 쥐와 고지혈증 유도 쥐에서 생체 지질대사의 생화학적 기작], 실험Ⅱ[차세대 정상 쥐와 고지혈증 유도쥐에서 생체 지질의 분할기작 모니터링], 실험 Ⅲ[비만 유도 쥐에서 생체 지질대사의 생화학적 기작], 실험 Ⅳ[성인에 대한 계란 섭취 전, 후의 혈액지질 변화 임상실험]를 수행하였다. 그 결과, 장기적으로 계란을 섭취한 동물에서 HMG CoA reductase 활성억제 및 스테로이드 배설촉진과 관련한 지질의 흡수억제 대사경로로써 혈액 지질감소를 확인하였고, 계란의 섭취가 지질의 흡수를 억제함과 동시에 산화를 가속화하여 줌으로써 생체조직 내 지질축적을 낮추며, 간에서 새롭게 합성된 Glycerolipid 분할에서 중성지방 분비를 낮춤과 동시에 인지질 분비를 증가시키고 14CO2 발산을 증가시키기 때문에 혈액 지질을 낮출 수 있다는 점, 특히 계란의 섭취 시 간에서 합성 분비되는 중성지방과 인지질의 대사적 분할능력을 조절하는 기작에 의해 혈액 지질을 낮춘다는 새로운 사실을 확인하였다. 또한, 비만 쥐에서 계란의 섭취가 혈액 내유해지질을 낮춤과 동시에 간 기능관련 효소 수준을 낮춤과 동시에 정상인에서 계란의 섭취가 혈액지질 변화에 있어서 유의한 영향을 주지 않는다는 사실을 확인하였다. 3차년도는 교미 1개월 전-교미-임신-분만
-출생 후 이유 6주령까지 장기간에 걸쳐서 계란을 섭취한 차세대 쥐에서 수행하였다. 실험Ⅰ[계란 섭취 차세대 쥐에서 성장능력, 혈액 지질 profiles, 인지질 변화], 실험 Ⅱ[계란 섭취 차세대 쥐에서 지질대사 관련한 생체유전자 발현], 실험 Ⅲ[계란 섭취 차세대 쥐와 스코폴라민 유도 치매쥐에서 수영 테스트], 실험 Ⅳ[계란 섭취 차세대 쥐에서 뇌해마 신경세포 활성능력]를 수행하였다. 그 결과, 계란의 섭취가 흰쥐의 혈액중 유해한 지질을 낮춤과 동시에 유익한 HDL-C와 함께 인지질 함량을 높인다는 점, 흰쥐의 간에서 콜레스테롤 합성 관련 유전자인 SREPB-1c의 발현을 하향조절 함과 동시에 지질분해와 관련한 간 PPARα, 지방조직의 LPL의 발현을 상향조절 해줌으로써 혈액지질을 낮춘다는 점, 난황을 경구투여 군에서 수영 도피대를 찾아가는 시간이 유의하게 가장 빨랐으며 스코폴라민 을 통하여 단기 기억손상을 유도한 후 수영에서 가장 빨라서 계란의 섭치가 흰쥐의 기억학습 능력 증진 및 치매예방에 도움을 준다는 점, 난황을 경구투여한 쥐에서 동일한 현미경 시야를 합체해 본 결과 면역염색된 세포는 세포돌기가 뚜렷하게 관찰되는 신경세포 생성이 촉진되었고, 계란 섭취가 신경세포활성에 도움이 된다는 새로운 사실을 확인하였다.

Controversial opinions have existed regarding the fact that egg intake elevates blood LDL cholesterol levels. It has been newly reported, however, that egg intake maintains normal blood LDL cholesterol levels and it also enhances brain functions. Accordingly, this attempts has clarified the mechanisms by which the above phenomena occur. The object of current study, alongside with the developmental growth of next-generation rats which had an intake of the eggs during pregnancy and the mechanism of lipid metabolism was to clarify using an in vivo monitoring technique with isotope labelling. With the activation of brain function, human study has performed to demonstrate the effectiveness of egg intake on maintaining the levels of normal blood cholesterol.
With the target of demonstrating the effectiveness of egg lecithin which can enhance the brain health as well as the safety of egg cholesterol which has been reported to be unrelated to the etiology of cardiovascular diseases, in rats’ liver to which the jugula canula mechanisms of glycerolipids about triglyceride performed using the in vivo monitoring technique of selective labelling with cholesteryl [1-14C]oleate. Thus, the study has elucidate the mechanism that the egg intake maintains the normal level of blood cholesterol levels and triglyceride(the first year). In human disease model animals(obese rats, hyperlipidemic rats) and the next-generation rats which made a delivery following the egg intake during pregnancy, the biochemical mechanisms by which the developmental growth, blood lipid metabolism and liver protection occur determined. Thus, this study has elucidated the mechanism that the egg intake is not associated with the elevated level of blood cholesterol levels as well as the improving the developmental growth in next-generation rats, obese rats and hyperlipidemic rats. In male and female adult college students with a normal lipid profile, changes of blood lipids due to egg intake measured(the second year). In normal rats which had an intake of eggs and those with dementia from which the memory impairment was induced by scopolamine, following an gene expression and analysis of maze test about the activation of brain function, it has clarified mechanism that the egg intake contribute to enhancing the memory(the third year).
The facts which were disclosed through the current study would be applied as the promotional data for the promotion of egg consumption as well as new knowledge about the nutritional value of egg.
Moreover, the clarification of mechanisms by which egg cholesterol does not elevate blood lipid would greatly contribute to transferring the knowledge about a lack of the relationship between the disease occurrence and egg intake in those who have a fear against egg intake, i.e., those who are worried about cerebrovascular and cardiovascular diseases. Based on the fact that egg intake could also be of help for promoting the developmental growth of next-generation children and the learning ability, and preventing senile dementia, egg will be of great interest as a Brain food. A synchronous analysis of the index factor of nutrient and metabolism which is greatly associated with cerebrovascular and cardiovascular diseases and an inference of changes in the metabolic pathway would be a baseline data for the functional metabolomics. An in vivo monitoring technique would be applied to studies about lipid such as diabetes, hypertension and obesity.
