화장품 원료 중 자외선 차단성분 옥틸메톡시신나메이트와 아보벤존, 살균보존제 페녹시에탄올과 클로로아세트아마이드)의 노출도 평가를 통한 위해도 평가를 실시하여 옥틸메톡시 신나메이트, 아보벤존, 페녹시에탄올 그리고 클로로아세트아마이드의 리스크프로파일링을 작성하고자 하였다. 생체 시료 내에서 옥틸메톡시신나메이트, 아보벤존, 페녹시에탄올 그리고 클로로아세트아마이드의 분석을 위하여 LC/MS/MS를 이용한 생체분석법도 확립하였다. 자외선 차단성분 연구결과, 옥칠메톡시신나메이트를 정맥 내 주입 후 혈중 농도를 200ng/ml로 유지한 후, 각 조직 장기에서의 분포를 분석한 결과, 표적장기로 여겨지는 고환에서 가장 낮은 분포를 보였다. 아보벤존의 경우, 랫드에서 정맥 주입후 표적장기인 고환에서 가장 적은 분포를 나타내었다. 랫드에서 옥틸메톡시신나메이트와 아보벤존의 피부를 통한 전신 노출도 평가 결과, 12시간까지 혈장내에서 검출되지 않았으며, 피부에서 미량이 검출되었다. 또한, 랫드의 피부에서 분해율 실험 결과 24시간에 각각 93, 63%가 분해 되었다. 이를 통해 두 물질의 경피 투여 시 미량이 피부로 흡수되나 혈액을 통한 전신흡수가 일어나지 않는다고 사료된다. 옥틸메톡시신나메이트와 아보벤존의 안전역은 각각 5882와 173으로 확보되어 옥틸메톡시신나메이트와 아보벤존의 허용 최대 배합한도 7.5%와 5%에서는 안전하다고 사료된다. 살균보존제 연구결과, 패녹시에탄올을 정맥 내 주입 후 정상상태 (steady-state)에서 페녹시에탄올은 신장으로 상대적으로 높게 분포하였으나 타 장기에서의 농도는 혈장농도와 유사하거나 낮은 것으로 보아 조직분포는 대체로 잘 일어나지 않는 것으로 보인다. 경피투여 후 페녹시에탄올의 흡수율은 에멀젼과 로션제제에서 약 76%로 산출되었다. 위해도평가 결과 페녹시에탄올의 안전역 (MoS)은 166.7로 산출되어 페녹시에탄올의 배합한도 1%는 안전하다고 사료된다. 체내로 투여된 클로로아세트아마이드는 주요 장기인 비장, 신장, 고환에서 낮은 조직분포율을 나타내었으며 (Kp < 0.75), 간에서는 검출되지 않았다. 경피투여 후 클로로아세트아마이드의 흡수율은 46.5-51.5%로 측정되었으며, 이를 기반으로 산출된 안전역 (MoS)은 23.5로 배합한도에 대한 검토가 필요할 것으로 사료된다. 이러한 결과는 자외선 차단성분 중 배합한도 성분인 자외선 차단성분 (옥틸메톡시신나메이트와 아보벤존)과 살균보존제 (페녹시에탄올과 클로로아세트아마이드)의 위해평가를 통해 정책 결정 기초자료를 제공할 수 있다. 화장품 원료에 대한 네거티브 시스템의 과학적 근거마련의 근거를 제공한다. 위해요소에 대한 자체 교육 자료로 활용하여 위해성 평가자들의 기술적 자질 향상을 통하여 위해성 평가 및 위해 관리에 보다 능동적이고 과학적인 대응이 가능하다.

For basic pharmacokinetic (PK) profile, iv study was performed and PK parameters were determined for octyl methoxycinnamate (OMC) and butylmethoxydibenzoylmethane (avobenzone). At the two UV filtering agents by iv infusion, OMC and avobenzone concentration in various tissues and organs were determined for tissue distribution. Interestingly, testes were the lowest concentration of the two agents implying existence of barrier system, even though testes are assumed to be target for OMC and avobenzone. In vivo dermal absorption study showed that MOSs (margin of safety) of 7.5% OMC and 5% avobenzone emulsion and lotion formulation were estimated to be 5882 and 173, respectively. These results suggested that the up to 7.5% OMC and 5% avobenzone might be safe for UV-filtering agents. Phenoxyethanol and chloroacetamide, which are used as preserving agents, were studied for risk assessment using exposure assessment and risk profiles are described. For basic pharmacokinetic profile, iv study was performed and PK parameters were determined for phenoxyethanol and chloroacetamide. At the two preserving agents by iv infusion, phenoxyethanol and chloroacetamide concentrations in various tissues and organs were determined for tissue distribution. Phenoxyethanol concentration in kidney was the hight among various tissue and other tissue concentations were lower than plasma comcentration. Chloroacetamide concentration in liver was not detected and most of tissue concentrations were lower than plasma concentrations. In vivo dermal absorption study of phenoxyethanol showed MOSs (margin of safety) of emulsion and lotion formulation were estimated to be 166.7. These results suggested that the up to 1% phenoxyethanol might be safe for UV-filtering agents. In vivo dermal absorption study of chloroacetamide showed MOSs (margin of safety) of emulsion and lotion formulation were estimated to be 23.5. These results required that should be reconsidered the up to 0.3% chloroaceamide might be sdjusted for safety.
