알츠하이머병(Alzheimer‘s Disease, AD)의 환자에서 인지기능과 관련된 해마와 대뇌피질 신경세포의 심각한 소실메 발견되며, amyloid plaque, neurofibrillary tangle이라는 병리적인 현상을 특징으로 한다. 알츠하이머병의 발병 요인으로는 amyloid precursor protein, apolipoprotein E, presenilin 및 α2-macroglobulin 등의 위험유전자가 발현되었으며, amyloid plaque의 주성분인 beta-amyloid peptide
알츠하이머병(Alzheimer‘s Disease, AD)의 환자에서 인지기능과 관련된 해마와 대뇌피질 신경세포의 심각한 소실메 발견되며, amyloid plaque, neurofibrillary tangle이라는 병리적인 현상을 특징으로 한다. 알츠하이머병의 발병 요인으로는 amyloid precursor protein, apolipoprotein E, presenilin 및 α2-macroglobulin 등의 위험유전자가 발현되었으며, amyloid plaque의 주성분인 beta-amyloid peptide (Aβ)의 축적이 알츠하이머병의 발병과 긴밀한 관계가 있는 것으로 알려져 있으나 아직까지 그 확실한 병인기전은 밝혀져 있지 않다. 따라서 본 연구과제에서는 신경세포의 퇴행과 사멸을 방지하고 재생을 촉진시키기 위하여, Aβ에 의한 신경독성을 방지하는 효과가 보고된 신경성장인자, estrogen, 항염증제, 천연물등의 신경 보호 기전을 규명하고저 하였다.
제 1 세부과제 : 해마의 배양된 1차 신경세포, explant culture, 신경세포주등을 이용하여 약 10종의 신경 성장인자가 신경세포의 생존, outgrowth, Aβ에 의한 세포독성억제에 미치는 효과를 평가하였다. 그중 bFGF, NT3, BDNF가 해마신경세포의 생존뿐만 아니라 neurite outgrowth에도 가장 탁월한 효과가 있음을 확인하였다. Aβ 에 의한 신경세포 독성이 NO, peroxynitrite등을 생성함으로써 세포내 산화적 스트레스를 유발할 수 있는지 조사한 결과 NO의 생성이 증가하지 않으며 Aβ에 의한 세포사멸이 NOS 저해제에 의해 변화하지 않는 것으로 미루어 Aβ에 의해 독성을 유발할 정도의 NO가 생성되지 않는 것으로 보이고 nNOS 의 발현에도 변화가 없었다. 한편, Aβ 와 NO donor인 SIN을 동시에 처리시, 저농도의 SIN(<50 uM)에서는 Aβ에 대한 신경세포보호효과가 있었으나 고농도 (>100 uM)에서는 보호효과가 없었다. 이상의 결과는 Aβ가 신경세포독성을 유발하는 농도에서 신경세포 내 NO생성을 직접적으로 변화시키지 못하나 rnicroenvironment로부터 신경세포가 NO에 노출시 NO의 농도에 따라 Aβ에 의한 신경독성이 조절되는데, 저농도의 NO는 Aβ에 의한 신경독성을 보호하나, 고농도의 NO는 Aβ에 의한 신경독성을 보호하지 못함을 제시한다. Aβ에 의한 세포독성 보호효과가 관찰된 신경성장인자 중 bFGF는 astrocyte에서 A+IFN-r에 의한 NO생성을 촉진함을 발견하였다. 이러한 작용에는 MAP kinase pathway가 관련되어 있고 cAMP에 의해 저해되고 PI3 kinase에 의해 repression되어 있음이 확인되었다.
결론적으로 아밀로이드 fibril의 주성분인 Aβ의 신경독성은 신경세포에 직접적으로 NO를 생성시키기 보다는 astrocyte에 작용하여 IFN-r와 같은 immunocytokine과 작용시 iNOS를 유도함으로서 NO를 과다생성하여 신경세포에 독성을 유발할 가능성이 큰 것으로 사료된다. bFGF와 같은 산경성장인자는 신경세포보호 효과가 있는 반면 astroglia cell을 활성화시켜 NO를 증대시키므로 신경성장인자에 대한 뇌질환에서의 역할이 재조명되어야 할 것으로 생각된다.
제2세부 : 학습과 기억을 담당하는 해마의 CA1 부위와 Entorhinal cortex는 알츠하이머 환자에서 신경세포의 퇴행이 가장 심각한 부위이며 BDNF는 CA1 부위에서 가장 많이 발현되고 해마의 pyramidal cell과 basal forebrain의 cholinergic neuron의 생존인자이다. 본 연구에서는 신경간세포의 분화과정에서 신경성장인자 BDNF와 그 receptor TrkB의 발현과 인산화를 통해 신경세포의 생존과 분화가 증진되는 것을 연구하였으며 recombinant BDNF-adenovirus를 제조하여 흰쥐 의 entorhinal cortex에 ibotenic acid를 주입한 치매모델에서 recombinant BDNF-adenovirus가 신경재생을 촉진하는 것을 관찰하였다. 따라서 본연구에서 제조된 recombinant BDNF-adenovirus가 신경재생을 촉진하는 치매 치료제로 개발될 가능성을 제시한다.
제3세부 : 알츠하이머 병(Alzheimer's disease. AD)의 뇌병변을 구성하는 주요물질은 세포독성을 나타내는 베타 아밀로이드 펠티드 (Aß, ß-amyloid peptide) 이다. Aβ의 전구단백의 대사물중 Aβ를 포함하는 카르복시말단 105아미노산 조각 (carboxy terminal 105 arnino acid fragment, CTF105)도 역시 세포독성을 나타내는 것으로 알려져 있는데, 본 연구에서는 천연물로부터 뇌보호 또는 뇌기능개선기능이 있는 물질을 검색하고자 하였다.
PC12 cell에 대한 CTF105의 세포독성에 대한 진세노사이드(Rb1, Ro, Rg1) 와 Egb761의 억제효과를 살펴보았다. 세포독성은 MTS assay를 이용하여 ELISA reader로 흡광도를 측정하였다. PC12 cell에 0.1 - 1.0 mg/ml의 농도로 진세노사이드와 Egb761을 전처리하고 나서, CTF105노출후 72시간 뒤에 관찰할 결과, Egb761은 CTF105의 세포독성을 억제하였으나, 진세노사이드는 억제하지 못하였다. Egb761 은 CTF105에 노출후 2시간뒤에 처리하여도 CTF105의 세포독성을 억제하였다. 항산화효과를 가진 진세노사이드-Rb1 및 Rg1 이 CTF105의 독성을 억제하지 못하는 것으로 보아, Egb761의 CTF105의 세포독성억제화과의 기전은 항산화효과보다는 PAF효과의 길항과 관련이 있을 것으로 추정되었다.
제4세부 : 에스트로겐의 신경세포 보호 기전은 free radical 생성을 저해하는 antioxidant로서의 역할과 에스트로겐 수용체(ER)를 매개로 하여 신경세포 보호와 관련되는 유전인자의 발현에 영향을 미쳐 일어나는 것을 확인하였다. 특히 antiapoptotic한 특성을 나타내는데 이것은 caspase-3의 활성을 막아서 신경세포사멸을 방지한다는 것을 규명하였다. 이 과정에서 에스트로겐에 의하여 PKC 활성화가 일어나고 ER 중 ERα가 에스트로겐에 의한 신경보호 효과를 매개하는 것으로 나타났다.
이러한 결과로부터 알츠하이머병 환자들을 대상으로 한 임상실험 결과들의 작용기전은 에스트로겐의 antioxidant로서의 역할과 더불어 ERα 를 매개로 하여 antiapoptotic한 단백질의 발현을 활성화시켜 신경세포 사멸을 방지하며 시냅스의 재생을 유도할 것이라 예측할 수 있다.
제5세부 : 연구개발사업의 목표는 치매 위험인자인 뇌 염증반응의 기전을 이해하기 위하여, 1) beta-amyloid에 의한 microglia 활성화의 신호전달체계를 이해하고, 2) microglia의 활성화를 유도할 수 있는 뇌 내의 물질을 확인하기 위함이다. 이를 위해 사용된 연구방법은 1) Microglia를 쥐의 뇌로부터 순수 배양하거나 BV -2 mouse microglia 세포주 사용하며, 2) Microglia의 활성화를 nitric oxide 분비와 iNOS, COX-2, TNF-alpha와 IL-1beta 등의 발현으로 확인하고 3) 활성화에 관여하는 신호전달물질로 PKC, MAPK, NF-kB, 활성산소종의 관련성을 확인한다. 본 연구의 결과, 1) Ceramide를 생성하는 sphingomyelinase (SMase)는 microglia의 활성 화를 유도하나 ceramide 자체 의 microglia 활성화 효과는 미약한 반면 ceramide와 SMase 모두 beta-amyloid와 LPS에 의한 microglia의 활성화를 촉진시키는 효과는 없었다. 2) beta-amyloid에 의한 microglia의 활성화에 활성산소종의 관여하였다. 3) Microglia의 활성화물질로 ganglioside와 thrombin의 가능성 확인하였다. 4)beta-amyloid와 LPS와 유사하게, ganglioside와 thrombin에 의한 microglia의 활성화에 MAPK, NF-kB, PKC가 관여함 등을 알아내었다.
제6세부 : Chrysamine G의 amide 유도체 6가지를 합성하였다. 이들은 Chrysamine G와 같이 Aβ fibril 에 대하여 affinity를 가지며 Aβ fibril로부터 세포 독성 방어 효과를 보여준다. 이들의 구조를 고찰함으로써 Carboxylic acid의 적당한 배향보다는 전체 분자의 hydrophobocity가 중요하다는 결과를 알 수 있었다.
제7세부 : human placental alkaline phosphatase (SEAP)의 ectodomain과 APP 의 β-secretase deavage site 및 cytoplasmic region 이 연결된 융합단백질인 SEAP-APPswe를 제조하였으며, SEAP-APPswe의 발현이 APP 및 BACEl 의 subcel1ular localization에 미치는 영향을 조사하였다. SEAP-APPswe는 APP의 localization에는 영향을 미치지 않았으나 BACE1 의 localization에는 큰 영향을 미쳐 ER, Golgi-apparatus, plasma membrane, endosome에 분포하던 BACE1이 plasma membrane에서만 발견되었다.
이 결과는 endomembrane system의 lumen에 protein phosphorylation에 의한 protein trafficking 조절기작이 존재함을 의미하며, 특히 이 조절기작을 조작하여 BACE1 과 y-secretase의 localization을 괴리시킴으로서 APP로부터의 Aβ생성을 억제하는 치매 치료제 개발의 가능성을 확인하였다.
제8세부 : 본 연구에서는 알츠하이머 치매의 병인 기전으로 알려진 베타 아밀로이드에 의한 세포손상에 지질 과산화물인 HNE가 관여하며 HNE가 세포손상을 일으킨다는 것과 이의 기전을 MAP kinase pathway를 중심으로 밝히고자 하였다. 베타 아밀로이드가 처리된 세포에서 4시간에 HNE의 생성이 최고에 이르렀다. HNE는 MAP kinase중에서 JNK를 선택적으로 활성화하였으며 ERK나 p38 kinase에는 영향을 주지 못했다. SEK와 ASK의 wild type과 dominant negative mutant cDNA를 transfection시킨 실험결과는 HNE가 JNK의 upstream에 있는 SEK1과 ASK1을 경유하여 신호를 전달했으며 이 경로가 막히면 HNE의 JNK에 대한 신호도 차단되고 세포손상도 초래하지 않았음을 암시하였다. 또한 JNK와 p38 kinase의 억제재인 CPT-cAMP와 SB203580을 이용한 실험을 통해 HNE에 의한 세포손상이 ERK 혹은 p38 kinase가 아닌 JNK의 pathway를 통한다는 것을 확인하였다.

Alzheimer's disease(AD) is neurodegenerative disorder characterized by progressive cognitive decline resulting from loss of neurons. Senile plaques, deposit of beta-amyloid peptide (Aβ) are known as the hallmark of neuropathological feature of AD, but the role of Aβ in AD pathogenesis is not well understood. In this study, we studied the mechanism of Aβ-mediatd neurotoxicity and brain inflammation in order to understand the ways of blocking cell death. In addition, we investigated molecular mechanism of protection mechanism of several neuroprotective agents, such as neurotrophic factors, estrogen, natural products, chemical compounds against Aβ-mediatd neurotoxicity.
The hit project: In this project we investigated to understand whether Aβ-induced neuronl cell toxicity is mediatd through NO generation and which neurotrophic factors are most effectively protects Aβ-mediated cell death. In addition we examined the neurotrophic factors and other agents to find the factors that regulate gene expressIOn of low-density lipoprotein receptor related protein (LRP) which is a multi-ligand receptor and has been considered as a key molecule for Aβ clearance.
Treatment of SHSY5Y, human neuronal cell lines. with A /3 showed no effect on NO synthesis and expression of nNOS protein and mRNA levels. Incubation of NOS inhibitor together with Aβ also did not protect neuronal cells from Aβ toxicity. In contrast, NO donor, SIN showed dual effects on A /3 toxicity: SIN showed neuroprotective activity at the lower concentration than 50 f-1 M but no protective effect at the higher concentration than 100 uM. Thus, treatment of Aβ, at the concentrations which cause neuronal cell death did not induce NO production and no changes in nNOS expression in the neuronal cells, suggesting NO is not involved in Aβ-mediated neuronal cell death. In constrast, NO generated in the astroglial cells modulates neuronal cell death and affects differentaially depending on the concentration of NO donor. We also evaluated the neuroprotective activities of neurotrophic factors. Of ten neurotrophic factors tested, BDNF, NT-3 and bFGF protected neurons most effectively against Aβ toxicity. However, bFGF potentiated IFN-r/Aβ-induced NO production in astrocyte via MAP kinase pathway. NO production by bFGF was blocked by cAMP and potentiated by PI-3 kinase inhibitors. Therefore, it is necessary to reconsider the role of neurotrophic factor in neurons and astroglial cells.
The 2nd project: In Alzheimer's disease patients, neuronal degeneration is very serious in CAl subfield of hippocampus and entorhinal cortex and BDNF is a survival factor for pyramidal cells in hippocampus and cholinergic neurons in basal forebrain. This study shows that neurotrophic factor, BDNF facillitated neuronal survival and differentiation by the expression and phosphorylation of the BDNF receptor TrkB during neuronal stem cell differentiation. We generated recombinant BDNF-adenovirus and showed that it promoted neuronal regeneration in AD animal model which was , injected by ibotenic acid into entorhinal cortex.
The 3rd project: Amyloid beta-peptide (Aβ) is the main component of the brain pathology of Alzheimer's disease. C-terminal 105 amino acid fragment (CTF105) of amyloid peptide precursor is harboring Aβ domain and is known to be also cytotoxic. The protective effects of ginsenosides and Egb76l was examined on the cytotoxicity of CTF105 to PCl2 cells. Cytotoxicity was measured by MTS assay (Promega) kit with absorbance reading with ELISA reader. Ginsenosides (RbI, Ro, Rgl) and Egb76l was pretreated at the final concentration of 0.1 - 1.0 mg/m!, and PCI2 cells were examined for 72 hours after CTF105 exposure.
Ginsenoside could not protected PC12 cells, however, Egb761 could protect PC12 cells even when it is added to the cells 2 hours after CTF105 exposure. As ginsenoside RbI and Rgl showing antioxidant effect could not protect PC12 cells from CTF105 cytotoxicity, the mechanism of the protective effects of Egb761 was expected to be related not with its antioxidant effect but with anti-PAF activity.
The 4th project : Although estrogen is known to exert beneficial effects on Alzheimers disease (AD), its underlying cellular mechanisms have not been clear. In this study we investigated whether or not neuroprotective effects of estrogen are mediated by estrogen receptors (ERs). Treatment of estrogen reduced beta-amyloid (Aβ)-induced death of ER-expressing cells. This effect of estrogen was blocked by a specific ER blocker ICI 182,780. When estrogen was treated to HT22 cells, which lack functional ERs, Aβ-induced cell death was not affected. Transfection of HT22 cells with human ER α, but not ER β, restored protective action of estrogen against Aβ. Hoechst staining revealed that estrogen protected ER α-expressing cells by bocking Aβ-induced apoptosis. These results indicate that estrogen blocks Aβ-induced cell death via ER α-dependent pathways.
The 5th project : The specific aims of this study is to understand intracellular signaling mechanisms and endogenous activators of brain inflammation, a risk factor of Alzheimer's disease. For this, 1) we examined intracellular signaling mechanisms that mediate beta-amyloid-induced microglial activation. 2) We investigated endogenous microglial activators and examined intracellular signaling mechanisms involved in their action. The methods used in the experiments as followings: 1) microglia were prepared from neonatal rat brain. 2) NO release, and expression of iNOS, COX -2, TNF-alpha and IL-1beta were detected by Griess's method, Western blot, and RT-PCFt. 3) The involvement of MAPK, PKC, NF-kB, and reactive oxygen species mOS) in microglial activation were studied by Western blot, gel shift mobility assay, and enzyme inhibitors, etc. The results obtained were 1) sphingomye1inase (SMase) but not ceramide induced microglial NO release. However, neither SMase nor cerami de enhanced the effect of beta-amyloid and LPS on microglial activation. 2) ROS mediated beta-amyloid-induced microglial activation. 3) Ganglioside and thrombin could be endogenous microglial activators. 4) MAPK, NF-kB, and PKC mediate microglial activation induced by gnaglioside and thrombin.
The 6th project : Six derivatives of the Chrysamine G are synthesized. They show comparable affinities to Aβ fibril as Chrysamine G and protect neuronal cells from the toxic effects of the Aβ fibril. It seems that hydrophobicity of the molecule is the most important factor for the affinity of the molecule to The Aβ fibril.
The 7th project: SEAP-APPswe containing human placental alkaline phosphatase (SEAP) fused with the β-secretase cleavage site and the cytoplasmic region of APPswe was generated. We examined the effect of SEAP-APPswe on the subcellular localizations of APP and BACEL SEAP-APPswe did not show any effect on the subcellular localization of APP. On the other hand, the localization of SEAP-APPswe was largely changed by SEAP-APPswe. Without SEAP-APPswe, BACE1 is distributed in ER, Golgi-apparatus, plasma membrane, and endosome. However, BACE1 was found mainly in plasma membrane in the presence of SEAP-APPswe. These results suggest the existence of a regulation mechanism on protein trafficking by protein phosphorylation within the lumen of endomembrane system. Furthermore, the separation of BACE1 and r-secretase by manipulating this regulation mechanism may result in the repression of Aβ production from APP.
The 8th project : We tried to investigate the role of HNE in β-amyloid-induced PC12 cell damage and its molecular mechanism. Specifically, we determined the effect of HNE exposure on the activities of MAP kinases involved in early signal transduction. HNE was detected in the highest level at 4h after β-amyloid was treated. Within 15 to 30 min after HNE treatment, JNK was maximally activated before it returned to control level at 1h post-treatment. In contrast, the activities of ERK and p38 kinase remained unchanged. The pathway of HNE-induced JNK activation through SEK1, an upstream kinase of JNK, and ASK1, an upstream kinase of SEK1, was demonstrated by the transient transfection of expression constructs for SEK1 or ASK1 wild type along with JNK into COS-7 cells. The data with specific inhibitors such as CPT-cAMP, a survival promoting agent, or SB203580, a specific inhibitor of p38 kinase suggest that the selective JNK activation by HNE is essential for the apoptosis of PC12 cells and that the HNE-induced apoptosis is likely to be medjated through a selective activation of the ASK -SEK1-JNK pathway without activation of ERK or p38 kinase.
