Catecholamine의 생합성 조절작용 및 monoamine 대사활성에 관한 연구는 각종 중추신경계 질환(우울증, 정신분열증, 파킨슨씨병등), 순환기 질환(고혈압, 심장병)과 밀접한 관련이 있다. 본 연구에서는 중추신경계 질환에 주로 사용되는 생약을 포함한 200여종의 천연물을 대상으로, catecholamine 생합성 및 대사조절작용을 가진 생리활성 물질을 검색하고 그 작용기전을 규명하고자 하였다. 이를 달성하기 위하여 천연 생리활성 물질의 catecholamine 생합성 조절작용에 대한 연구, PC12 cells 중의 dopamine 함량, TH 및 AADC 활성에 미치는 영향연구, 천연 생리활성 물질의 .대사 효소 MAO 활성에 대한 연구, 천연 생리활성 물질의 catecholamine 생합성 조절작용-작용기전에 대한 연구 (Dopamine 함량, Cyclic-AMP 함량, Intracellular Ca++ 농도, Blotting analysis), 천연 생리활성 물질의 catecholamine 생합성 효소 활성에 대한 연구 (TH 활성, DBH 및 PNMT 활성에 대한 효소 화학적 연구), 천연 생리활성 물질의 대사효소 MAO 활성 및 그의 작용기전에 대한 연구 (MAO 활성 작용기전,항우울 작용 검색, 체온강하 억제작용) 등을 수행하였다. 천연물로부터 MAO 저해활성물질로는 산초나무 유래의 lacinartin (IC50 2.4 ug/ml, 침향 유래의 psolaren (IC50 5.33 uM), 계피의 coumarin (IC50 : 7.3 ug/ml), 3,3-dimethyl-1-propenyl benzene (IC50 : 19.7 ug/ml, cinnamic acid (IC50 : > 100 ug/ml, 2-methoxycinnamaldehyde (IC50 13.5 ug/ml, 갯기름나물의 praeruptorin A, xanthotoxin, psoralen, bergapten, Basidiomycetes sp.8082 로부터 화합물 5-methylmellein, 전동싸리의 4종 화합물, Kaempferia galanga로부터 5종 화합물, 벌사상자로부터 분리된 coumarin 화합물 23 종에 대한 MAO 저해활성을 측정하고 구조와 활성관계를 연구하였다. catecholamine 생합성 조절작용 연구결과 200 여종의 천연물중에서 황련, 히드라스티스근, 구기자, 한방기, 오수유 등이었으며 dopamine 생합성을 저하시켰으며, 현호색, 백굴채, 방기 등은 dopamine 함량 증가작용을 나타내었다. Catecholamine 생합성 조절작용의 작용기전을 알기 위하여 alkaloid 화합물 isocorydine, hydrastine, perlolyrine, protoberberine 등을 이용하여 dopamine 함량·에 대한 영향과 작용기전을 조사하였다. Catecholamine 생합성 조절작용 연구결과 200 여종의 천연물중에서 황련, 히드라스티스근, 구기자, 한방기, 오수유 등은 dopamine 생합성을 저하시켰으며, 현호색, 백굴채,방기 등은 dopamine 함량 증가작용을 나타내었다. Catecholamines are neurotransmitters anφOr hormones having diverse biological activities in the periphery and in the central nervous system. In order to find out the modulators of catecholamine biosynthesis from natural products, we tested the inhibitory activities on monoamine oxidase and effect of dopamine contents in PC 12 cells with with two hundred medicinal plants. Some MeOH extract of medicinal plants showed MAO inhibitory activities and others showed the regulating effects of catecholamine biosynthesis. A number of MAO inhibitors have been isolated from natural products lacinartin (IC50 2.4 ug/ml from Zanthoxylum schinifolium, psolaren (IC50 5.33 uM) from Aquilaria agalocha, coumarin (IC50 7.3 ug/ml, 3,3-dimethyl-1-propenyl benzene (IC50 : 19.7 ug/ml, cinnamic acid (IC50 : >100 ug/mI) , 2-methoxycinnamaldehyde (IC50 13.5 ug/ml from Cinnamomi Cortex, praeruptorin A, xanthotoxin, psoralen, bergapten from Peucedanum japonicum, 5-methylmellein from Basidiomycetes sp.8082, four pterocarpane compounds from Mallotus alba, five cinnamic aldehyde derivatives form Kaempferia galanga and 23 compounds from Cnidium monnieri. Several naturally occurring coumarin and cinnamic acid derivatives showed potent inhibitory .activities against MAO. Also we investigated the regulating activities of catecholamine biosynthesis with 200 medicinal plants. Seven plants showed the lowering effects of dopamine contents in PC12 celIs, whereas three plants showed the increse of dopamine contents. In order to investigate the mechanism of regulating catecholamine biosynthesis, we tested the effects of dopamine contents with several alkaloids such as isocorydine, hydrastine, perlolyrine and protoberberine. From these research, we obtained several inhibitors and modulators from natural products. These compounds can be used as lead compounds for the treatment of depressionin. Further investigations are required to establish structure-activity relationship and in vivo studies of naturally occurring these catecholamine synthesis modulators.
