Ⅳ. 연구개발 결과
1. 종묘생산 시스템 구축
가) 모충 관리 시스템
모충관리를 위해 유수식 및 반순환식 시스템을 구축하였다. 모충사육을 위한 사육수조는 6 m x 1 m x 0.7 m의 FRP와 콘크리트 수조를 사용하였다.
해수는 10 L/min 이상 공급하였고, 조도는 3,000 lux 내외로 조절하고 에어레이션을 실시하였다. 사육수조 저면에 자갈을 10 cm 높이로 깔고 마사토, 굴패각, 황토 등을 4.5:4.5:1의 비율로 조성하였다. 기질 위 수위는 25 cm 이하로 유지하였다. 부경대학교 수산과학기술
Ⅳ. 연구개발 결과
1. 종묘생산 시스템 구축
가) 모충 관리 시스템
모충관리를 위해 유수식 및 반순환식 시스템을 구축하였다. 모충사육을 위한 사육수조는 6 m x 1 m x 0.7 m의 FRP와 콘크리트 수조를 사용하였다.
해수는 10 L/min 이상 공급하였고, 조도는 3,000 lux 내외로 조절하고 에어레이션을 실시하였다. 사육수조 저면에 자갈을 10 cm 높이로 깔고 마사토, 굴패각, 황토 등을 4.5:4.5:1의 비율로 조성하였다. 기질 위 수위는 25 cm 이하로 유지하였다. 부경대학교 수산과학기술센터 내에 기조성되어 있던 유수식 시스템은 5월~8월 발생하는 자연종묘 채집을 위해 활용하였으며, 반순환식은 겨울철(11월~3월 사이)에 수온조절을 통한 조기종묘를 생산하기 위하여 콘크리트 수조를 새롭게 구축하였다.
나) 유생 사육 시스템
초기 폐사율이 높은 유생의 생존율을 향상시키기 위해 별개의 유생 사육시스템을 구축하였다. 초기 유생은 20℃의 항온항습실에서 사육하였다. 사육상자는 45 cm x 30 cm x 12 cm 의 플라스틱 상자를 사용하였고 멸균된 해수 6 L를 넣고 2만미의 유생을 입식하였다. 입식 전 해적생물 분리 작업을 통해 유생만을 선별하였다. 유생의 착저를 돕기 위해 2일간 지수상태를 유지하고 3일째 부분환수(50% 이상)를 실시하였다. 7일까지 항온항습실에서 사육한 다음 후기 사육(nectochaete 시기)을 위해 다단식 프레임으로 조성된 유충 사육동으로 옮겨 사육하였다. 후기 사육은 유생의 생산시기에 따라 유수식 및 반순환식 시스템을 적용하였다. 반순환식 시스템에는 비닐하우스 및 전기히터 그리고 여과시스템이 추가로 구성되었다. 사육수조는 40 cm x 70 cm x 20 cm로 초기 유생 사육상자보다 큰 것을 사용하였으며 3 cm 이하의 마사토를 사육기질로 사용하였다.
다) 치충 사육 시스템
후기 유생 사육시스템의 연장선으로서 유수량 및 사료량을 늘려 유충의 빠른 성장을 유도하기 위하여 사육시스템을 설계하였다. 먼저, 10 um의 필터(bag type filter)를 여러 개 설치하여 여과해수를 원활하게 공급하였다. 유수식 사육시스템은 별도의 변경 없이 분양까지 수량 및 사료량을 증가시켰다. 반순환식 사육시스템은 겨울철 저수온기를 지나 해수 수온이 상승하는 4~5월 이후 천천히 자연해수의 유입량을 증가시켜 유수식으로 전환시켰다. 사료량의 증가에 따른 수질 오염을 방지하기 위해 주 2회 수질 분석과 수조청소를 실시하였고, 스키머장치를 활용하여 유기배출물을 제거하였다.
2. 인공종묘생산을 위한 번식생물학적 연구
가) 번식생리 주기 규명
바위털갯지렁이의 성성숙, 산란주기 등의 생리적 특징을 규명하고, 이를 기반으로 인공종묘생산 기술을 개발하고자 하였다. 생식소 발달과정을 조직학적으로 관찰한 결과 분열증식, 성장기, 성숙기, 산란기 및 휴지기의 연속적인 5단계로 구분할 수 있었다. 1월에서 4월까지 암수 생식소가 성장·성숙하여 5~8월 사이 방란·방정하는 것을 확인하였다.
나) 인공종묘생산기술개발
모충의 성숙을 위해 반순환식 시스템을 활용하였다. 100일간의 사육수온 조절을 통해 성숙한 모충의 정자와 난을 여과해수에서 10분간 정치, 혼합하여 인공수정 및 부화를 시도하였다. 수정난의 크기는 230~270 ㎛였고 1~3일사이에 분할이 시작되었다. 4~5일째에 한쪽 부분이 돌출되면서 미부로 발달하였고, 반대편에는 안점이 형성되었다. 6일째부터 대부분의 유생이 2쌍의 강모(chaetae; 우족, parapodia)를 가지고 타원형의 담륜자 유생(trochophore stage)으로 완성되었으며, 최저 수온(7℃)부터 이때까지의 누적수온은 530℃였다. 발생된 유생을 관찰한 결과 자연환경조건에서 발생한 유생과 동일한 형태를 가지고 있었다. 따라서 인위적인 유생 발생의 유도에 의한 인공종묘생산이 가능하게 되었다(특허등록).
다) 유생의 초기성장과 적응 특성
분출된 유생은 후기 담륜자(metatrochophore) 시기의 유생으로 난황과 섬모를 가지고 있었다. 유생은 2~3일간 착저와 유영을 반복하였다. 난황과 섬모가 사라지면서 3번째 우족(third parapodia)을 가지며 nectochaete기로 접어들었다. 7일 경과 후 장이 생성되었고 먹이 및 퇴적물을 섭취하기 시작하였다.
또한 점액질을 분비하여 서식공을 만들었다. 관상의 서식공은 표층에 ‘ㅡ’, ‘ㅅ’ 모양으로 생성되었다. 15일 경과 후 진짜 눈이 나타났으며, 치충(juvenile) 단계로 접어들었다.
라) 유생의 발생 환경 특징 규명
최근 4년간의 유생 발생 환경(수온, 일조량 및 조석)을 조사하였다. 유생의 발생 수온 범위는 18~24℃였다. 따라서 매년 5월 초·중순에 유생이 발생하였고 25℃ 이상 수온이 형성되는 8월 중순에 발생이 정지하였다. 유생발생은 새벽 시간에 주로 분출되며, 일조량이 하락하는 시기에 대량 발생하였다. 조석의 영향을 살펴본 결과 조금과 사리 때의 유생 발생량은 낮은 반면, 3~5물, 10~12물 사이에 유생 발생량이 매우 높았다. 또한 최대 발생 시점에 모두 박무 상태를 나타내었다. 이것은 흐리고 박무가 형성된 새벽녘 잔잔한 시간대에 유생이 부상하여 적당한 물의 흐름을 따라 안전하게 이동·확산되어 착저할 수 있도록 인지된 발생생태학적 전략인 것으로 사료된다. 이러한 유생발생 환경과 물때로 유생 발생 시기를 유추할 수 있기 때문에 앞으로 양식 현장에서 손쉽게 활용할 수 있고, 생물의 생식주기와 발생환경과의 상관관계에 관한 기초연구에 중요한 자료로 활용될 수 있을 것으로 기대된다.
마) 연령에 따른 산란 능력 조사
바위털갯지렁이의 연령별 체중, 체절수, 포란수, 포란한 난의 크기별 구성비 등을 조사하여 각각의 산란 능력을 조사하였다. 바위털갯지렁이의 연령별 평균체중은 평균 4.31 g(2006년), 2.27 g(2007년), 1.80 g(2008년), 0.9 g(2009년), 0.25 g(2010년)이었다. 체절수 증가율은 1년 20%, 2년 27.8%, 3년 2.3%, 4년 12.5%였다. 연령별 포란수는 2006년산 평균 31,902 개, 2007년산은 평균 15,037 개, 2008년산은 평균 9,750 개, 2009년산은 3,000~6,000 개의 난을 포란하였다. 난 크기는 2006년산이 275 ㎛ 이상의 난이 43%로 조사대상의 연령 중에서 가장 높은 구성비를 보였고, 2007년산은 15%, 2008년산은 7%였고, 2009년산은 모두 245 ㎛ 이하였다. 2년 이상 사육된 개체는 모두 체절 200개 이상을 보유하였고 산란능력이 있음을 확인하였다.
3. 대량 종묘생산 및 생산성 향상 기술개발
가) 유수식 자연 종묘 생산
자연 해수를 이용한 모충사육수조에서 연간 유생의 생산량 변화를 조사한 결과, 최근 4년간(2010~2013년) 유생 생산량은 2010년 대비 5.8배 증가하였다. 유수식 종묘생산에서 유생발생을 위한 누적수온은 400℃ 이상이었다. 유생 발생시기에 모충장 사료 급이 제한은 모충의 영양상태를 악화시켜 유생발생과 생존에 큰 영향을 미치는 것으로 판단되었다. 실제 2013년 종묘생산기간 동안 꾸준한 모충 관리를 통해 많은 양의 유생을 5월 초에 확보할 수 있었으며, 초기 생존율을 높일 수 있었다. 따라서 모충의 영양 관리에 따른 성성숙, 발생 및 유생의 생존율에 대한 추가 연구가 필요하다.
나) 반순환식 인공 종묘 생산
수온조절을 통해 겨울철 조기 종묘 생산을 시도한 결과 유생의 발생은 성공적으로 이루어졌으며, 이를 통해 연간 2회 종묘를 생산할 수 있었다. 최근 3년간의 인공종묘생산 결과 유생 발생까지의 적산수온은 530℃ 이상으로 관찰되었다. 동일 사육수조에서 유생 생산량은 2011년 273만미였으나, 2013년 1,634만미로 6배 증가하였다. 반순환식에서 성공적인 대량종묘생산을 위해서는 안정적인 수온 및 수질관리, 해적생물의 대량증식방지와 지속적인 모충장 관리가 필요한 것으로 판단되었다.
다) 유생의 생존율 향상 조건 규명
유생의 생존율을 향상시킬 수 있는 사육 조건을 실험한 결과, 최적 사육수온은 20℃, 사육수는 여과해수 및 멸균해수, 기질은 A기질(특허출원중)로 높은 성장 및 생존율(92%)을 나타내었다. 특히 기질에 따른 유생의 착저시간은 성장과 반비례하였다. 먹이는 배합사료와 탈각알테미아가 비교적 효과적이었으며, 그리고 적절한 사육밀도는 10~40 마리/cm2로 나타났다. 상기 결과를 토대로 초기 유생의 성장과 사육기법, 그리고 초기 사육단계별 먹이 프로그램을 확립하였다. 따라서 유생의 발생 초기 대량 폐사를 방지할 수 있었으며, 기존에 보고된 바위털갯지렁이 유생의 형태적 특성보다 더 빠른 발달을 관찰할 수 있었다.
라) 초기 사육기법을 적용한 대량생산 종묘의 양성
초기 사육기법을 적용하여 유생 발생으로부터 12개월간 성장 상황을 월별로 조사하였다. 육안으로 치충을 확인 가능한 시기는 3개월이 지난 후부터였으며, 이때의 평균 무게는 0.0037 g이었다. 5개월 차에 급격한 성장을 보였으나 이후 저수온기에 접어 들면서 성장이 크게 둔화되었다. 자연수온과 가온수에서 양성한 치충의 평균 체중 변화에서 30% 이상의 차이가 발생했고 생존율도 20% 이상 차이가 났다. 따라서 월동 시기에는 가온 사육이 필요한 것으로 판단되었다.
마) 대량생산 종묘의 생산성 향상 기술 개발
종묘생산 기간의 가장 심각한 문제점은 해적생물 발생에 의한 포식과 생존율 저하로 확인되었다. 따라서 유생 및 치충의 생존율 향상을 위해 포식성 요각류와 원생동물의 제어 실험을 실시하였다. 포식성 요각류 제어를 위한 Trichlorfon(일반명; 마조텐)의 적정 농도는 3 ppm 이하, 0.1 ppm 이상이었으며, 처리 시간은 48시간이었다. 이외 유생으로부터 분리·정제한 추출물에 의한 요각류의 제어방법을 모색하였다. 멸균해수 사용 및 광촉매 처리법 등의 수처리 방법을 통해 발생시기 및 양을 조절할 수는 있었으나, 아직까지 원생동물을 원천 차단할 수 있는 방법은 숙제로 남아 있다. 한편 치충 양성 수조를 변경, 재배치하여 사육시스템을 개선하였고, 부착규조류의 대량 배양 시스템(특허출원)을 구축하여 먹이공급을 원활하게 하여 생산성 향상 효율성을 높였다.
4. 산업폐기물 재활용 양식 및 친환경 육상 산업생산 실용화 기술개발
가) 친환경 고밀도 중간양성 기술개발
마사토, 모래, 굴패각을 이용해 최적 중간양성을 위한 기질을 선정하였다. 실험결과 굴패각 100% 구간에서 성장이 가장 우수(굴패각 단독 구간 200%)하였고, 배수율 또한 90% 이상으로 모래, 마사토 구간보다 높게 나타났다. 기존 갯지렁이 양식기질로 사용하고 있는 모래 및 마사토를 굴 양식 폐기물인 굴패각으로 대처 또는 혼합하여 활용하는 효과적인 산업폐기물 재활용법이 제시됨(특허출원·등록)으로써 친환경 저비용 양식 적용이 기대된다. 적정 밀도에 관한 실험은 세가지 다른 그룹(0.5 g 이하, 0.6~1.5 g 및 1.6~2.5 g/개체)의 바위털갯지렁이를 이용하였으며, 실험결과 0.5 g 이하의 그룹에서는 1,000 마리/㎡의 밀도, 나머지 두 그룹에서는 2,000 마리/㎡의 밀도에서 가장 높은 일간 생산력(daily biomass production)을 나타내었으나, 생존율과 개체 성장은 저밀도에서 높았다. 그룹 내에서도 밀도에 따라 생산력의 차이가 컸기 때문에 분양 및 분조 시에는 입식밀도에 주의해야 할 것으로 판단된다.
나) 바위털갯지렁이 육상 산업생산 실용화 기술개발
육상 산업생산 실용화를 위해서 성장에 우수했던 굴패각 기질을 활용한 친환경 양식(특허출원) 및 넙치와 바위털갯지렁이의 연계양식(특허등록)을 시도하였다. 먼저 기존 기질을 친환경 소재인 굴패각으로 대체하여 실제 현장에 적용할 수 있도록 각 성장 단계별 생존 및 성장율을 조사하였다. 유충(0.003 g 이하) 단계에서는 마사토와 굴패각을 75%, 25% 혼합한 구간에서 성장률이 가장 높았으며, 치충(0.007 g 이하) 단계와 성충단계에서는 굴패각 100%가 성장률이 가장 높았다. 성장 단계별 굴패각 비율이 조금씩 다르게 나타났지만 본 연구를 통해 각 단계별 적정 비율을 알 수 있었다. 넙치-바위털갯지렁이의 연계양식 실험 결과 별도의 사료 공급 없이 넙치 사육배출수로 바위털갯지렁이 양식이 가능함을 보여주었다. 낮은 입식 밀도에서 높은 성장과 생존율을 나타내었으나, 적절한 사료를 추가로 공급해 준다면 고밀도에서도 높은 생존율을 나타낼 수 있을 것으로 판단된다.
다) 친환경 양식기술을 적용한 오염제어 효과 및 경제성 평가
넙치와 바위털갯지렁이의 연계양식 실험에서 유수식과 반순환사육시스템의 배출수의 오염제어 효과를 검토하였다. 유수식 사육시스템에서 가장 높은 제어효과는 그룹 2(0.6~1.5 g)에서 TSS(총부유고형물; 86%) 및 총질소(56%), 그룹 1(0.5 g 이하)에서 총인(59%), 그룹 3(1.6~2.5 g)에서 COD(30%)로 나타났다. 반순환식 시스템에서는 그룹 2에서 총질소(63%) 및 COD(21%), 그룹 3에서 총인(53%)과 TSS(총부유고형물; 80%) 제어 효과가 가장 높게 나타났다. 또한 바위털갯지렁이 사육을 통해 로티퍼 배양 배출수 내의 찌거기 및 TN 그리고 COD가 낮아짐을 알 수 있었다. 이를 통해 기존 육상 양식시스템을 활용하여 양식장의 유기오염 배출물을 제거하고 수질환경 또한 개선할 수 있는 것을 증명하였다.
한편 바위털갯지렁이 종묘생산과 양성을 통한 이윤 창출을 위해서는 각 과정의 면밀한 경제성 비교·검토를 실시하였다. 동계 저수온기를 거쳐야 하는 지역적 여건을 감안하여 조기 종묘생산과 중간양성 단계별 가온의 필요성이 제기되었다. 전복생산과 비교했을 때 갯지렁이 양식의 발전 가능성은 생산기간 감소에 따른 경비절감의 높은 경제성 효과를 보였다. 또한 사육기법에 따라 면적대비 생산량을 증대시킬 수 있고, 현재 생존율 10% 기술수준을 향후 50% 이상으로 높일 수 있기 때문에 앞으로 더 높은 이익 창출을 기대할 수 있을 것이다. 바위털갯지렁이 양식기술 보급이 아직 미흡한 상태이고, 어류양식 등 수산생물 생산시스템에서 발생하는 해양 오염원 제어를 위한 기술은 거의 반영되지 못하고 있는 실정이다. 고부가 친환경 소재인 바위털갯지렁이의 대량생산기술의 실용화 확대에 따른 어업인 소득 증대는 물론, 이에 대한 친환경 현장 활용기술을 갯벌 복원 및 연안오염 방지를 위한 중장기 정책과제로 반영하여 그 성과를 조속히 거둘 수 있기를 기대한다.

1. Establishment of seed production system for polychaete Marphysa sanguinea
(1) Brood stock management system
It had been constructed two different kinds of stock management tanks for mass seed production, such as (a) flow-through system, and (b) semi-recirculation system. Culture vessels made of fiberglass reinforced plastic or concrete (L 6 m × W 1 m × H 0.7 m), that were constructed for adult polychaete. Sea water was supplied at 10 L/min. Illumination (ca. 3,000 Lux) and aeration were also supplied. The bottom of the tank covered with gravel (10 cm in height) and mixer of sand, oyster shell and ocher (4.5:4.5:1). Water level was maintained at 25 cm. The flow-water rearing system was utilized to collect larvae produced from rearing tanks during May to August. The semi-recirculation system was utilized for early seed production during November to March.
(2) Larvae rearing system
In order to an effective management for larval growth with high survival, it was constructed the control room for larvae breeding. The larvae from the brood stock tanks were collected and transported to the control room with temperate and humid chamber (20℃ maintained). Rearing cages made of polyethylene (L 45 cm × W 30 cm × H 12 cm) and filled with sterile seawater (6 L). The densities of larvae (20,000 inds/cage) were strictly maintained. After separating harmful organisms, larvae had been cultured in the sterile seawater for 7 days. After settling down completely, larvae grown up to nectochaete stage were moved to flow or semi-recirculation culture system. Semi-recirculation culture system was consisting of vinyl house, electric heaters and filtration system. Cages for nectochaete (L 40 cm × W 70 cm× H 20 cm) were bigger than that of larval stage. Sand substrate was also supplied.
(3) Juvenile culture system
This is an extension of larvae rearing system. This rearing system had been developed by increasing feed and water supply for the purpose of high growth performance of juvenile worms. Additionally, several bag filters (10 μm) were initially installed for supplying filtered sea water. In flow-through system, food and water quality were only increased without any other system changes. In semi-recirculation system, it had been supplied natural seawater during April to May, instead of heated seawater. Finally, water quality was regularly monitored to prevent pollution and cages were also removed organic substances including suspended solids by using skimmer device.
2. Reproductive biological research for seed production of polychaete Marphysa sanguinea
(1) Investigation of the reproductive physiology cycle
Based on the conceptual knowledge of sexual maturation, spawning cycle and physiological characteristics of M. sanguniea, we had been tried to develop artificial seed production techniques. M. sanguniea have multiplication stage, growing stage, mature and spawning stage, and resting and degenerative stage during gonadal development processes. On the natural seawater flow-through system, gonads of female and male were matured from January to April.
Spawning and spermatogenic spurt had been observed from May to August. After then, they had resting stage from September to November. Finally, gonadal development would be observed in the following year.
(2) Development of the artificial seed production techniques
In order to mature of mother worms through semi-recirculation system, water temperature had been controlled for 100 days. After maturation, collected sperm and egg had been tried to mix for 10 minutes. Finally, size of fertilized eggs was 230~270 μm. Cell division began between 1 to 3 days. After 5 days, one part of larvae was developed to the tail and eye sport was appeared in other side. Each larva has two chaete. Since 6 days, it had been started the trochophore stage inside the burrow where larvae spurted out to swim toward surface water, and daily cumulative temperature until larvae production from bottom temperature (7℃) was 530℃. Morphological characteristics of larvae was the same in both of artificial and natural system.
(3) Early development and adaptation characteristics of the larvae
Released larvae were metatrochophore that had yolk sac and cilia. Larvae were repeated swim and settle down for 3 days. After disappear of yolk sac, they have 3rd chaetes (parapodia) in nectochaete stage. The larvae had started to feed at seven days and secreted mucus that was used as the adhesive to particles for their tube housing. It constructed two types (‛-’, ‛ㅅ’) of tube. During juvenile stage, true eyes had been appeared after 15 days.
(4) Environmental characteristics on larvae production
We had been examined the relationship between larvae production of M. sanguinea and environmental factors (such as temperature, sunshine duration and tides) to provide information for seed production. During seed production period, water temperature had been maintained from 18℃ to 25℃. Therefore, the optimal larval production period was from May to mid-August. Larvae had largely produced at dawn time and lower sunshine duration. Also it was shown that there were strong positive relationship between larval production and tidal cycles. A huge amount of large had been collected on 3~5 and 10~12 tide flows. Whenever larvae produced at peaks with time-lapsed tidal cycles, it was almost cloudy and mist.
(5) Investigation of the yearly egg production
This study was provided basic information for artificial seed production and investigated some parameters (i.e. weight, number of segments, absolute fecundity and egg size of M. sanguinea). The approximate weight per individual of M. sanguinea was 4.31 g (production in 2006), 2.27 g (in 2007), 1.80 g (in 2008), 0.9 g (in 2009), 0.25 g (in 2010). The rate of segmentation had been increased by 20% (1 year), 27.8% (2year), 2.3% (3 year) and 12.5% (4 year). Fecundity (egg formation) of M. sanguinea was 31,902 (in 2006), 15,037 (in 2007), 9,750 (in 2008), and 3,000-6,000 (in 2009).
Egg size (275 μm) had been increased by 43% (in 2006), 15% (in 2007) and 7% (in 2008). In 2009, all of the egg was smaller than 245 μm. The worms which kept rearing for more than 2 years, had more than 200 chaetes. They had still ability for egg production.
3. Development of mass seed production and productivity enhancing techniques
(1) Natural seed production in the flow-through system
It was investigated the factors that enhanced larval production in the flow-through rearing system with natural seawater temperature. During last four years, larval production had been increased by 5.8%. In the flow-through system, daily cumulative temperature was maintained at above 400℃. In 2013, we tried to manage brood-stock for seed production. So, larval production had been increased and the highest production was recorded in early of May. Therefore, future researchs are needed in case of maturation, nutritional level, larval occurrence and larval survival rate.
(2) Artificial seed production in the semi-recirculation system
During winter season, early seed production was possible through controlling water temperature. As a result, larvae could be produced twice a year. During last three years, larval production had been increased by 6%. In the semi-recirculation system, daily cumulative temperature for artificial seed production was over 53 0℃. Therefore, some management strategies were needed such as stable temperature and water quality management, predator prevention and brood-stock management.
(3) Improving survival rate of the larvae
In the seed rearing experiment, the optimum temperature for the survival of larvae was found to be 20℃. There was no significant difference between the filtered seawater and sterilized seawater by the autoclave processes. Substrate A (which a patent was filed for) was the most effective among the rearing substrates. It showed higher growth and survival rate (92%). It had also time-consuming effect on the settling-down in the trochophore stage. The appropriate densities for settlement were 10-40 inds/c㎡. Early-rearing techniques were applied for the enhancement of survival rate on larval mass culture. When juveniles grew on optical confirming size in naked eye after 3 months, average weight was 0.0037 g. They had been shown rapid growth since 5 months, but they could not grow in winter season when their survival rate had also been decreased by 20%. So, it should be needed temperate control by heating for the survival and growth in winter season.
(5) Development of productivity-enhancing techniques
The most important problem on the mass culture processes was a serious deterioration of survival rate by predation of pirates organisms. In order to improve productivity during larva to juvenile stages, we had been conducted experiments. In the remedy method, predatory copepods were effectively responded in the trichlorfon (0.1-3 ppm) for 48 hrs. In addition, the activity of copepods, especially harpacticoid, was lowered by treatment of larval extracts. It was necessary to manage seawater to protect from protozoan harassing during breeding. The sterile seawater was used to delay the occurrence of protozoan in the initial breeding time. So, we had been improved the juvenile breeding container and aquarium in order to increase productivity. We built a mass culture system of adhesive diatom (Patent pending) to supply food during larval production.
4. Development of industrial waste recycling aquaculture and eco-friendly industrial application in land-based aquaculture
(1) Development of eco-friendly aquaculture techniques for intermediate rearing stage
We had been investigated the growth rate of juveniles by using different type of substrates for intermediate rearing stage. Substrates with oyster shells showed higher survival of worms and did water flow rate (90%). Substrates without oyster shells showed lower growth, survival and discharge rate (83.3%). Substrates mixed with oyster and sand showed the highest weight gain (443.9%). It was conducted to find optimal density for intermediate rearing stage. For the study of effect of density on growth of M. sanguinea with commercial feed, worms in group 1 (less than 0.5 g, 1,000 inds/㎡) had the highest pure production by feeding on 3% body weight. In group 2 (0.6-1.5 g), worms (2,000 inds/㎡) had the highest pure production by feeding on 3.5% of body weight. However survival and individual worm growth in high density was not better than low density but daily biomass production in all groups belonged to medium and high density. This study demonstrated that M. sanguinea can be one of the most suitable species to commercially exploit in marine worm farming system.
(2) Development of eco-friendly industrial application in land-based aquaculture
Waste shells after harvesting of oyster culture was used for the substrate, instead of sand grains. The highest and lowest value in the growth rate and weight gain of juveniles were shown when they were reared in 100% and 20% of oyster shell, respectively. This results demonstrated that the growth of M.sanguinea was correlated with sediment features. It is expected that the oyster shell on 50% substrate will give an effective growth of M. sanguinea (Patent pending). However, the integrated culture system with oyster shell and finfish culture outlets into polychaete culture tanks were developed for eliminating of organic pollutants, caused by fishery farm (e.g. a black porgy and flatfish). This system (Patented) was also very efficient both in economic and environmental points because there was no need/ or a little to supply additional food for polychaete. So, it is expected to increase profits for farmers and revitalize the business.
(3) Effects of water quality improvement applying eco-friendly aquaculture techniques and economic evaluation
Aquaculture is growing rapidly all over the world and the increasing number of fish culture industries has begun to create serious environmental problems due to the impact caused by fish farming wastes. Based on the information on great potential of the polychaete worms for bioremediation and the successful seed production of the polychaete rockworm Marphysa sanguinea at Fisheries Science and Technology Center, PKNU, three trials have been conducted to test the water quality improvement and growth performance in the rockworm settlement tanks receiving wastes from olive flounder rearing tanks, in the integrated culture systems with flow-through and semi-recirculation. In comparison between culture systems, the pure production of the worm was 1.5 times higher in flow-through system than in semi-recirculating system. Different effects on the removal efficiency were appeared both in culture systems and among treatments. In flow-through system, the highest removal efficiency showed in TN (56 %) in G2 (0.6~1.5 g/initial weight), TP (59%) in G1 (<0.5 g), TSS (86%) in G2, and COD (30%) in G3 (1.6~2.5 g). In semi-recirculating system, the highest removal efficiency was in TN (63%) in G2, TP (53%) in G3, TSS (80%) in G3, and COD (21%) in G2. These results suggest that the rockworm M. sanguinea is an excellent candidate for integrated aquaculture and nutrient recycling including the removal of organic wastes in land-based systems.
Overall, higher larval survival rate is possible by developing rearing techniques. On the base of seed production, we compared between polychaete and abalone. To achieve higher profits in polychaete culture systems, it is needed temperate control by heating during winter season and should be adapted to early seed production. As a result, polychaete rockworm culture is still now ongoing development of commercial production level, but the production of this high-value polychaete must be competitive in terms of price, quality and product diversity when compared to the available sources. The need to achieve a sufficient degree of competitiveness requires a highly efficient production system. For this reason, there is a need for collaboration between the scientific sector and the production sector. The results of this research, if made available in the public and governmental sectors, may lead to important changes and applications in the understanding of polychaetes at the most fundamental level.
