Although diverse studies about deer and their velvet antler and constant researches about chemical component of velvet antler have been done, there is no research for evaluation of quality of velvet antler. Also, there is no criteria for appreciating the quality of velvet antler throughout the world. In the present situation that diverse velvet antlers are produced and used in various nations, absence of criteria means that it is impossible to make a difference in velvet antler. Thus, this study is conducted to gain basic data which is beneficial for evaluation and improvement of quality of velvet antlers by analyzing and investigating the components of velvet antlers of elk and spotted deer in Korea.
The purpose of these studies are to develop the technique for quality standardization of domestic velvet antler from 2006 to 2008 year. In order to investigate the physiological properties of velvet antler in the Korean elk velvet antlers (n=378) and spotted deer (n=93) were collected in 8 areas including Gyeonggi-do, Gangwon-do, Chungcheongbuk-do, Chungcheongnam-do, Jeollabuk-do, Jeollanam-do, Gyeongsangbuk-do, and Gyeongsangnam -do. Velvet antlers were measured the characteristics of exterior shape (yields, length, girth, and beacon). Mean yields of elk and spotted deer were significantly differences according to the areas. Mean number of point of Korean elk and spotted deer were 3.0±0.5 and 1.6±0.5, respectively. As the girth of velvet antler, top was markedly thicker compared to the middle, which has a narrow cylinder shape in the middle. As the length of point, 1 point was significantly longer compared to the 2 point and 3 point.
Based on the above results, elk (n=18) and spotted deer (n=11) velvet antler samples were randomly selected for measurement of the chemical characteristics. The dry matter content and the crude ash content increase from the top to the base in elk and spotted deer. Crude protein, crude lipid, and sugar component contents tend to decrease from the top to the base. Thus, the upper the part is, the more organic matters such as proteins, lipids and sugar components it contains. In contrast, the lower the part is, the more inorganic matters such as ashes it contains. There is a certain tendency that physiologically active components such as GAGs, uronic acid, sialic acid and collagen contents are higher in the upper parts of the velvet antlers and are lower in the lower part of them. Especially, the collagen content decreases from the base to the top. Also, the Ca and the P contents of the top are markedly higher compared to the base, but there is no accurate difference between the side points in elk and spotted deer, indicating the lowest base markedly developed integumental thickening by mineralization process compared to the top of velvet antler. Moreover, the top tends to have more free amino acids than the base, though glutamic acid, glycine, arginine and alanine contents are higher in the base. On the other hand, the there are more serine, glutamine, threonine, proline, valine, isoleucine, leucine and lysine in the base than in the middle.
As the correlation between exterior and interior characteristics, the crude protein, sugar and GAGs have close correlation with top length, yield, and depth of beacon, respectively.
Uronic acid has positive correlation with yield, depth of beacon and total girth of velvet antler. Also, sialic acid and collagen contents have negative correlation with point length in Korean elk and spotted deer. On the other hand, free amino acid content was closely correlated with capacity.
In order to develop the technique for quality standardization of domestic velvet antler by growth period and parts, the correlation analysis was conducted between physical and chemical characteristics in Korean elk (n=16) and spotted deer (n=15). Velvet antlers of elk were cut intervals from 60 day to 95 day (65, 80, and 95 day), and velvet antler of spotted deer were cut in 40, 50, and 60 day. In velvet antler of Korean elk, average velvet antler yield, total length and girth, top length and length of 3 point at 95 day were markedly higher compared to those of 65 and 80 day. As the velvet antler cut off at 95 day, crude protein, free amino acid and fatty acid contents show the close correlation with total length. Moreover, sugar and GAGs contents has close correlation with top length and girth, respectively in velvet antler cut off at 65 day.
In velvet antler of spotted deer, average velvet antler yield at 60 day was markedly higher compared to those of 40 and 50 day. Ash, GAGs and uronic acid contents tend to increase by increasing the cutting time contrary to the crude protein, sialic acid and collagen contents. Yield, total length, top length and depth of beacon in spotted deer velvet antler was positive correlation with dry matter, ash, GAGs, uronic acid and sugar contents but negative correlation with crude fat, sialic acid and collagen contents.
Based on the above results, we established the quality standardization of domestic velvet antler and graded A (33-40), B (23-32), C (17-22), and D (9-16) according to synthetic judgment including yield, total length, top length and girth of top.
