본 연구의 목적은 식물 뿌리 주위의 설탕으로부터 새로운 구조의 당을 합성하고 다당을 분해하는 토양 유래 미생물의 개발과 효소 당전이 특성을 확인하는 것임. 식물 뿌리 주위에는 잎에서 광합성으로 생산된 설탕의 10% 이상이 분비되고 있어, 이를 이용하는 토양 미생물이 다양하게 분포 되고 있음. 이 중에는 특히 설탕을 이용하여 다양한 구조의 다당을 합성하는 균과 합성된 다당을 분해하는 특성의 균들이 있으며, 합성 및 분해를 하는 효소들 중에는 당전이 물질을 합성하는 특성의 효소가 여러 가지 존재 함. 따라서 Nucleotide-sugar를 단당의 제공체로 사용하는 대신 값이 저렴한 설탕만으로 새로운 구조의 당을 만들고, 신규 다당을 분해하고 산물을 전이하는 간단하고 효율적인 새로운 효소의 개발에 대한 요구가 절실하며, 본 연구에서는 식물 뿌리 부근의 토양 미생물 중설탕을 이용하여 새로운 구조의 다당을 합성하고, 분해하는 특성의 새로운 당전이 효소를 개발하고, 효소 특성을 연구하고자 진행함. 설탕 배지를 이용하여 다당을 합성하는 균들을 여러 가지 식물들의 뿌리 주위 토양에서 선별하였고, 생산 된 효소들은 설탕의 글루코오스를 전이할 수 있는 당전이 효소임을 확인함. 이 효소는 천연물 유래 항산화 소재의 배당체를 합성하는 연구에 활용하기 위한 연구를 진행 중임. 덱스트란을 분해하는 효소를 생산하는 균을 선별하였고, 선별을 위해 사용한 배지는 불루-덱스트란으로 가수분해 효소의 분해로 투명한 존을 형성하는 정도로 활성이 높은 균을 확인 할 수 있음. 이 균의 효소를 포함하여 세가지 효소를 이용한 덱스트란 가지결합을 확인하는 효소적 방법을 개발하였음. Paenibacillus sp.endo-dextranase는 덱스트란을 선형과 가지결합 이소말토올리고당으로 분해하고, Streptococcus mutan의 dextran glucosidase는 선형 또는 가지결합된 이소말토올리고당을 글루코스와 가지결합된 이소말토4탄당으로 분해가 되며, 가지결합된 이소말토4당은 Bacteroides thetaimicron alpha-glucosidase로 분해를 할 수 있어, 이 효소들의 시너지 반응을 통하여 가지 결합을 가지는 덱스트란을 모두 글루코오스로 전환 할 수 있었으며, 512F 덱스트란이 가지결합을 5.42% 가지고 있음을 확인함. 가지결합을 더 잘 분해 할 수 있는 효소를 대량 생산 할 수 있는 균을 개발하며, 글루칸의 구조 결정과 이를 이용한 새로운 당전이 산물을 개발하고, 생산하는데 활용할 수 있겠다.

Plant photosynthesis produces starch, which is converted to sucrose (Suc). Suc moves to storage organ, like seeds (rice). It was well known that plants secret Suc from their roots to soil, and use Suc to cross-talk to soil-microorganisms (SM). Very recently, plants were discovered to release the huge amount of Suc, corresponding to 10% of one day's photosynthesis: We call this system as a "Sucrose-World". Purpose of this joint-project was the analysis of sucrose-world members [SM and their enzymes (SM-E)] and their application to industrial fields. More precisely, i) Isolation of SM and SME from soil near plant roots to produce polysaccharides (PSs) from Suc; ii) Isolation of SM and SM-E to convert PS to oligosaccharides (OSs; linear or cyclic form) or glycosides. We isolated various microorganisms synthesizing viscous polysaccharides using sucrose, such as dextran. We collected soil samples near plant roots and plated on sucrose containing medium. Then, mucous colonies were isolated and the enzyme produced in liquid culture was tested for glucansucrase activities. In addition to glucansucrases, we also isolated strains which produced dextranase using blue-dextran. Those enzymes can also transfer of glucose to other compounds which have free – OH group. Thus, those enzymes can be used for the synthesis of various natural polyphenol compounds. Including those dextranse, a novel quantitative method for the determination of degree of branching in Leuconostoc mesenteroides B-512F dextran was developed by using the combination of three dextran-degrading enzymes. First, Paenibacillus sp. endo-dextranase was randomly degraded B-512F dextran into linear or branched isomalto-oligosaccharides (2-8) with various degree of polymerization. Second, Streptococcus mutans dextran glucosidase hydrolyzed linear or branched isomalto-oligosaccharides into glucose and branched isomalto-penta-saccharides. Third, the branched isomalto-penta-saccharide was degraded into glucose by using Bacteroides thetaimicron α-glucosidase. The number of branching points in B-512F dextran (5.42%) was determined by the difference in the amount of glucose in the reaction digest between BTGase-PDex and DGase-PDex treatments. The degree of branching of highly branched dextrans, such as B-742CB or B-1299C dextran could not be determined due to limited PDex hydrolysis activity. The development of stronger hydrolytic enzymes to hydrolyze B-742CB or B-1299C dextran is in progress.
