인간 PTP는 세포 내 신호전달경로에 관여하고 있으며, 서로 잘못된 상호작용은 신호전달의 오작동을 초래하여 암, 대사성 질환, 면역질환, 신경세포 손상 등 거의 모든 질병들에 관여한다고 알려졌다. 본 연구에서는 인간 PTP의 subfamily인 Dual specific phosphatase(DUSP)들의 활성화 도메인을 대용량 발현, 정제, 결정화 및 구조분석을 통하여 생체내의 탈인산화 활성 메카니즘과 신호전달조절에 관해 이해하고, 구조적인 차이점에 따른 기질 특이성질을 예측하여 정확한 PTP 조절기능을 분석할 수 있는 기술을 개발
인간 PTP는 세포 내 신호전달경로에 관여하고 있으며, 서로 잘못된 상호작용은 신호전달의 오작동을 초래하여 암, 대사성 질환, 면역질환, 신경세포 손상 등 거의 모든 질병들에 관여한다고 알려졌다. 본 연구에서는 인간 PTP의 subfamily인 Dual specific phosphatase(DUSP)들의 활성화 도메인을 대용량 발현, 정제, 결정화 및 구조분석을 통하여 생체내의 탈인산화 활성 메카니즘과 신호전달조절에 관해 이해하고, 구조적인 차이점에 따른 기질 특이성질을 예측하여 정확한 PTP 조절기능을 분석할 수 있는 기술을 개발하였다.
(1) PTP 100종의 유전자를 human cDNA library를 통해 확보 하였으며, 이중 56종을 융합단백질을 이용하지 않고, 대장균에서 활성형태로 발현 및 정제함. 이중 20종의 구조를 규명하였고, 효소 활성을 고효율로 탐색할 수 있는 기술을 개발하여 선택적 저해제 개발의 기초 자료를 확보하였음
(2) MKP1(DUSP1), MKP7(DUSP16), Pyst2(DUSP7), hVH5(DUSP8) 의 활성화도메인을 대장균에서 대량발현 하여 결정화를 시도하여 MKP7, hVH5의 결정을 확보하고 hVH5의 결정구조규명 하고 MKP1, MKP7, Pyst2는 homology modelling 에 의해 삼차구조를 규명
(3) 기존에 규명된 DUSP 단백질들과 전체적으로 비교한 결과 여러 가지 secondary structure element 가 차이를 보이는 것을 발견하였고 DUSP 단백질들의 개별적 차이를 바탕으로 subgroup 을 형성할 수 있음을 발견하였고 3가지 subgroup 과 7가지 세부그룹으로 구분하여 분석하였음
(4) 14종의 MTMR 중에서 MTMR4의 GRAM-PH 도메인과 MTMR8 의 활성화 도메인 결정을 확보하였음고 MTMR4는 MAD 방법으로 구조규명을 준비중이며 MTMR8은 분자치환법으로 삼차구조를 규명하였음
(5) HYVH1(DUSP12), BEDP(DUSP13a), VHP(DUSP28), PIR1(DUSP11), MTRM3, MTRM4, MTRM8 활성화도메인을 10 mg 이상 대량 정제하고 HYVH1(DUSP12), PIR1(DUSP11), PTPsigma, MTRM3, MTRM4 를 결정화함

Human have 107 PTPs, which catalyze dephosphorylation of target molecules. PTPs are involved in diverse cellular signaling pathways. Defects in PTP pathways were often associated with many human diseases, including cancer, metabolic disease, immunological disease, neuronal disease. Structural elucidations of the catalytic domains of Dual specific phosphatases (DUSPs) are critical in understanding the molecular mechanisms and the functions of PTPs. We aim at a family-wide structure determination of the DUSP famiy enzymes and the development of the technologies to specifically regulate PTPs.
The phase I study was focused on the structure determination and structure-based functional analysis of DUSP in a family level. The catalytic domain of each individual DUSP has characteristic features in the active site and in surface-charge distribution, indicating substrate-interaction specificity. The active-site loop-to-strand switch occurs in a subtype-specific manner, indicating that the switch process is necessary for characteristic substrate interactions in the corresponding DUSPs. A comprehensive analysis of the activity–inhibition profile and active-site geometry of DUSPs revealed a novel role of the active-pocket structure in the substrate specificity of DUSPs. A structure-based analysis of redox responses indicated that the additional cysteine residues are important for the protection of enzyme activity. The family-wide structures of DUSPs form a basis for the understanding of phosphorylation-mediated signal transduction and the development of therapeutics.
