연구의 목적 및 내용: ∙ 극초단 적외선 이차원 분광법 개발 ∙ 시분해 진동 pump-probe 측정방법을 개발하여 극초단 화학 및 생물학적 현상에 대한 실험 연구 ∙ 키랄성 분자의 적외선 분광 스펙트럼, 진동 광학 활성 스펙트럼, 이차원 적외선 진동 스펙트럼의 양자 및 분자 동력학 시뮬레이션 방법 개발 연구결과: ∙기초연구: (1) 펨토초 적외선 레이저 펄스를 이용하여 이차원 적외선 분광기기를 개발하엿다. 특히 광-에코 측정 방식을 도입하여 각각의 펄스들의 편광 상태를 개별적으로 조절하는 것이 가능했고, 이 방식을 이용해 Polarization-Angle-Scanning 이차원 분광 실험이 가능함을 최초로 제안 검증하였다. (2) 생체분자 또는 다양한 유기화학 분자들의 경우 키랄성 구조의 특이성을 분석 측정하는 것은 매우 중요하다. 본 연구실에서는 광학 활성을 펨토초의 시간 분해능으로 측정할 수 있는 장비를 개발하였다. 특히 광 간섭 현상을 이용하여 광학 활성 시그널을 증폭하는 방식을 이론적으로 제안한 바 있으며, 그 가능성을 검증하는 실험을 처음으로 검증하였다. 특히 단 하나의 펨토초레이저 펄스만을 이용해서도 광학활성 스펙트럼을 측정하는 것이 가능하다는 것을 보여줌으로써 이분야의 측정기술 발전에 중요한 기여를 하였다. (3) 새로운 분광법의 개발과 함께 관련 장비를 구축하는 기초 연구와 함께, 적외선 탐침 화학 작용기를 아미노산에 도입하여 작은 펩타이드의 구조적 동력학을 피코초 단위에서 분석하는 연구를 수행하였다. ∙응용연구: (1) 본 연구실에서 개발한 이차원 분광장비를 이용하여 응축상 분자의 구조를 규명하였다. 특히 용액상 이온들간의 이온쌍 결합-해리 시간을 측정하였고, 응축상의 인공 아미노산의 구조, 분자내 진동 에너지 이동, 초고농도 전해질 용액에서의 이온 동력학 현상을 규명하였다. (2) 광학 이성질성 분자의 구조 규명 및 구조적 전이 과정을 측정할 수 있는 극초단 시분해능 장비를 이용하여 DNA-drug 복합체의 펨토초 원편광이색성 스펙트럼을 측정하였고, 다양한 올리고펩타이드의 구조와 구성 아미노산간의 관계를 규명하였다. (3) 적외선 탐침은 형광 또는 전자스핀 탐침 분자에 비해 그 크기가 작기 때문에 생체분자의 구조적 변형을 크게 유발시키지 않는다는 장점이 있다. 예를 들어 아지도기를 포함하는 인공아미노산에 대한 분광학적 연구를 통해 분자내 및 분자간 상호작용에 대한 원리를 규명하는 것이 가능하였고 이 방법을 통해 생체 분자의 특정 영역 주변에서의 국소적 전기장을 선택적으로 측정하는 것이 가능함을 증명하였다. 연구결과의 활용계획: ∙ 새로운 연구 방법의 개발은 다양한 분야의 연구에 지대한 영향을 미친다는 점에서 중요하다. 특히 본 연구실에서 개발한 이차원 적외선 분광학 및 펨토초 키랄광학 측정법은 다양한 화학 및 생물학적 현상의 메카니즘을 연구하는데 폭넓게 사용될 것으로 보인다. 현재 이 실험 장비 및 방법을 지속적으로 발전 개발시켜 생체 이미징 기술 및 기능성 재료의 분석 기술로의 확대 발전을 계획하고 있다. ∙ 적외선 탐침을 이용한 측정 방법은 효소 및 생체 단백질의 활성 자리 주변에서 특히 중요한 작용을 하는 국소적 전기장을 정량적으로 측정는데 사용될 수 있기 때문에 이 방법을 이용하여 세포막 단백질의 기능 및 메카니즘 연구를 추진하고 있다. 또한 세포질 내부의 전기적 환경 및 물분자 구조-동력학 연구에도 응용할 예정이다.

Purpose & contents: ∙Coherent multidimensional spectroscopy of biomolecules such as proteins, nucleic acids, and artificial polypeptides ∙Experimental investigations of chiral molecules and biomolecules ∙Developments of novel computational methods for calculating linear and nonlinear (2D) optical spectra of complex biomolecules ∙2D spectroscopic studies of biomolecular and chemical reaction dynamics in condensed phases Result: ∙ Fundamental: Coherent multidimensional spectroscopy, which is an optical analog of multidimensional NMR, has been paid a great deal of attentions due to its capability of studying molecular structure and dynamics in femtosecond time scale. In addition, time-resolved vibrational circular dichroism, which has been experimentally demonstrated in this year 2009, is considered to be an incisive tool for extracing chiral information on biomolecules. Thus, the present proposal consists of three parts: (1) coherent multidimensional optical and vibratrional optical activity measurement studies of chiral biomolecules, (2) novel development of computational methods combining quantum chemistry and molecular dynamics methods for direct simulations of the corresponding 2D and vibrational CD spectra, and (3) design, synthesis, and characterization of novel biomolecules containing IR probes for the above experimental and theoretical studies. It is emphasized that the present work requires concerted efforts including novel spectroscopic studies, computaton and theoretical modeling, and artificially-made biomolecules. ∙ Applications: (1) The current multidimensional spectroscopic studies of chiral biomolecules will help us to understand structural and dynamics aspects of biomolecules in real time. This will trigger further studies on protein folding-unfolding, protein-nucleic acid binding, and so on. (2) The time-resolved vibrational optical activity measurement technique developed here will again be used to elucidate the underlying mechanism of protein folding and to develop a fast drug-screening technology in the future. Finally, (3) Novel IR probes that can be easily incorporated into proteins and nucleic acids will be of critical use in studying local electrostatic environment around a probe, which in turn will provide information on the changes of local electric field in and out of enzyme active sites. This will give us a critical clue on detailed mechanism. Expected Contribution: ∙ Academic: Multidimensional optical and vibrational CD spectroscopies will play a critical role in studying ultrafast chemical and biological phenomena such as protein folding and their function. Novel tool such as these will stimulate a development of new scientific research fields. ∙ Industrial: These novel spectrometers will be eventually commercialized in the future so that the present work will be the first and fundamental step toward such an industrial development.
