연구의 목적 및 내용
본 연구는 줄기세포와 탄소나노섬유가 결합된 스캐폴드, 향신경성 인자 및 재활치료의 복합치료를 통해 척수손상에 가장 효과적인 치료방법을 개발하는데 목표가 있음. 이를 위해서 초기에 계획했던 분야 중에서 줄기세포와 스캐폴드, 전달체, 동물실험의 네분야를 나누어 줄기세포 분야에서는 유도만능 줄기세포의 수립 및 신경전구세포로의 분화, 스캐폴드 분야에서는 척수손상의 재생에 효과적인 스캐폴드의 개발, 전달체 분야에서는 신경재생관련 약물을 효과적으로 전달하는 다공성 나노입자 (MSN) 이용, 동물실험 분야에서는 척수손

본 연구는 줄기세포와 탄소나노섬유가 결합된 스캐폴드, 향신경성 인자 및 재활치료의 복합치료를 통해 척수손상에 가장 효과적인 치료방법을 개발하는데 목표가 있음. 이를 위해서 초기에 계획했던 분야 중에서 줄기세포와 스캐폴드, 전달체, 동물실험의 네분야를 나누어 줄기세포 분야에서는 유도만능 줄기세포의 수립 및 신경전구세포로의 분화, 스캐폴드 분야에서는 척수손상의 재생에 효과적인 스캐폴드의 개발, 전달체 분야에서는 신경재생관련 약물을 효과적으로 전달하는 다공성 나노입자 (MSN) 이용, 동물실험 분야에서는 척수손상모델의 정립과 다양한 평가분야 개발 및 확인에 중점을 두어 연구를 진행함
연구결과
이번 연구를 통해서 나타난 결과는 다음과 같음
-줄기세포 분야는 척수손상의 재생에 효과적일 것으로 기대되는 신경능선 세포로의 분화방법을 개발하여 특허를 출원하였음. 그리고 기존에 계획했던 유도만능 줄기세포의 단점을 극복하는 새로운 직접 역분화 신경줄기세포 및 희소돌기아교전구세포를 공급받아 동물실험 연구를 진행함
-스캐폴드 분야에서는 탄소나노섬유를 코팅한 인산유리섬유로 만든 삼차원 스캐폴드를 척수손상모델에 이식하여 신경재생 및 행동학적 기능 개선에 효과가 있음을 확인하였고 이에 대한 실험 및 리뷰 논문이 각각 1편씩 출판되고 관련 특허 3건이 출원됨. 다음 단계로 삼차원 스캐폴드 내에 미세섬유를 채우지 않고 미세도관이 형성되는 더욱 진보된 스캐폴드를 제작하여 현재 동물실험을 진행 중임
-전달체 분야에서는 PTEN 억제제를 함유할 수 있는 MSN을 신경세포 및 동물모델에 투여하여 신경재생에 효과가 있음을 확인함
-동물실험 분야는 줄기세포 및 스캐폴드를 이식하고 이에 대한 효과를 조직학적, 행동학적, 요역동학적, 전기진단학적으로 분석하고 이에 대한 결과가 논문 및 특허로 발표됨
-줄기세포와 향신경성인자, 스캐폴드가 결합된 형태의 복합 치료는 아직 진행되지 못하였으며 후속 연구를 통해 계속 진행될 예정임
연구결과의 활용계획
-신경능선 세포의 척수손상모델의 효과는 아직 밝혀진 바 없어 이에 대한 동물실험 연구를 통해 새로운 줄기세포 효과를 밝힐 수 있음
-직접 역분화 줄기세포의 척수손상모델 효과도 아직 밝혀진 바 없으며 이에 대한 연구를 진행하여 직접 역분화 줄기세포의 임상적 활용도에 관한 연구를 진행할 수 있음
-일정한 방향성을 지닌 내부구조의 스캐폴드 제작을 통해 신경재생에 최적의 조건을 가지는 스캐폴드 개발을 계속적으로 진행할 예정임
-줄기세포 및 스캐폴드에 관한 연구는 척수손상 뿐 아니라 말초신경손상 및 뇌질환 연구전반으로 확대될 수 있으며 이들의 연구 진행을 위해서 말초신경손상모델 및 뇌졸중모델의 수립을 마치고 후속 연구를 계획 중임

The aim of this study is to develop the most effective method for spinal cord restoration using stem cells, carbon-nanotube (CNT) coated scaffold, neurotrophic factors and rehabilitation. For this purpose, the research was divided into four parts;stem cell, scaffold, delivery system and animal model among initial plan. The stem cell part focused on the development of induced pluripotent stem cells (iPSCs) and differentiation into neural precursor cells, the scaffold part developed a scaffold which is effective for spinal cord regeneration, delivery system part used mesoporous nanoparticles (MSNs) to deliver neural regeneration-associated drugs effectively and the animal model part focused on the establishment of spinal cord injury models and various evaluation methods to detect the effect of treatments clearly.
Result
We could obtain the results and outcomes as following;
-From stem cell part, we had developed a method for development of neural crest cells from human iPSCs, and applied for a patent. And directly reprogrammed neural stem cells and oligodendrocyte precursor cells were supplied and performed experiments for spinal cord regeneration using animal models.
-From scaffold part, we found that developed three dimensional scaffold containing CNT-coated phosphate glass fibers was effective for spinal cord restoration histologically, functionally and electrophysiologically. And two manuscripts including a original article and a review article were published and applied for three patents.For next step, we continue to develop a new microchannel containing scaffold instead of microfiber-filled scaffold.
-From delivery system part, PTEN inhibitor-containing MSNs were applied to neurons and animal models and found effective and a manuscript was submitted.
-From animal model part, we had assessed the effects of stem cell transplantation and scaffold implantation into injured spinal cord using histology, locomotor function, urodynamics and electrophysiology, and related manuscripts and patents were published or applied.
-We did not perform combined treatments of stem cells, neurotrophic factors, and scaffolds yet, and we have a plan for proceeding this experiments soon.
Expected Contribution
-We will reveal the effect of neural crest cells, directly reprogrammed neural stem cells and oligodendrocyte precursor cells to spinal cord injury models, which does not come out into the open.
-We will develop the ideal scaffold which has aligned intraluminal structures which guide outgrowing axons in vivo.
-After successful experiments, stem cells and scaffolds in our study will have wide applications to peripheral nerve injury and brain lesions.
