연구의 목적 및 내용
도파민 D2 수용체 유전자 결손 쥐를 이용하여 신경정신질환 기전 규명 및 감정조절인자를 발굴하고자 한다. 이를 위하여 본 연구실에서 확인한 D2R-EGFR-ERK 신호전달체계에 초점을 맞추어 도파민 D2R 수용체에 의한 EGFR 신호전달체계가 어떻게 상호작용하는지 분석한다. 또한 뇌의 여러부위에서의 도파민 D2 수용체, EGFR의 발현양상을 이해하고, 특히 편도체에서의 정상쥐와 도파민D2 수용체 유전자 결손 쥐와의 EGFR 발현의 양적 비교 및 ERK 신호조절 등이 특정 행동조절에 어떻게 영향을 미치는 지
연구의 목적 및 내용
도파민 D2 수용체 유전자 결손 쥐를 이용하여 신경정신질환 기전 규명 및 감정조절인자를 발굴하고자 한다. 이를 위하여 본 연구실에서 확인한 D2R-EGFR-ERK 신호전달체계에 초점을 맞추어 도파민 D2R 수용체에 의한 EGFR 신호전달체계가 어떻게 상호작용하는지 분석한다. 또한 뇌의 여러부위에서의 도파민 D2 수용체, EGFR의 발현양상을 이해하고, 특히 편도체에서의 정상쥐와 도파민D2 수용체 유전자 결손 쥐와의 EGFR 발현의 양적 비교 및 ERK 신호조절 등이 특정 행동조절에 어떻게 영향을 미치는 지 분석하여, 스트레스 및 우울증 반응, 공포반응, 등의 행동 변화들을 분석함으로써 도파민성 신경정신 질환에서 나타나는 행동 및 감정조절의 표적인자를 발굴하며 도파민성 신경정신질환 기전 규명과 치료개발에 응용.
연구결과
도파민 D2 수용체 유전자 결손 쥐 (D2R-/- mice)를 통해 D2R-EGFR-ERK 신호전달 기전을 확인한 결과,
‣도파민 D2 수용체를 통한 EGFR의 활성이 metallorptease (MMP) 중 특정 ADAM10과 ADAM17(A Disintegrin And Metalloprotease)에 의해 조절되며, 이를 통하여 ERK의 활성화가 유도되고 있음을 확인하였다.
‣또한, 성인시기 D2R-eGFP 쥐에서의 EGFR의 발현패턴을 분석한 결과,편도체와 복측피개영역에서 발현되고 있는 D2R과 EGFR이 서로 공동발현되고 있음을 확인하였지만, 성인시기 정상 쥐와 D2R-/- 쥐의 편도체에서의 EGFR의 발현을 양적으로 비교한 결과, 큰 차이를 관찰할 수는 없었다.
‣성인시기 정상 쥐와 D2R-/- mice의 공포반응, 불안증세, 인지능력 및 충동성 등의 행동 반응 및 편도체에서의 ERK의 인산화를 관찰한 결과, 정상 쥐와 비교하여 D2R-/- mice는 높은 공포반응 및 불안증세를 보였으며, 공포자극에 대하여 편도체에서의 강한 ERK 인산화가 관찰되었다. 보상기전과 관련된 실험에서는 낮은 집중력 및 보상 획득을 보였고 높은 충동성을 보여주었으며, 이에 대하여 편도체에서는 낮은 ERK의 인산화가 관찰되었다.
‣보다 효과적으로 D2R-EGFR-ERK 신호전달 기전을 효과적으로 차단하기 위하여 세계최초로 D2R-Cre;EGFRlox/lox mice 제작, 이를 통해 보다 확고한 D2R-EGFR 신호전달기전에의해 조절되는 행동양상을 분석하였으며 D2R-Cre;EGFRlox/lox mice 는 보상기전에서 정상 쥐보다 낮은 보상 획득이 관찰되었다. 또한, MMP 억제제인 GM6001 처리시 이러한 보상 획득이 확연히 낮아짐과 함께, 편도체에서의 ERK 인산화 역시 관찰되고 있음이 관찰되었다.
연구결과의 활용계획
‣본 연구과제의 성공적 수행으로 성인시기 D2R-EGFR-ERK 신호전달기전이 보상회로를 조절하는 역할을 하고 있음을 확인하였고, 특히 뇌의 편도체와 복측피개영역이 이러한 조절을 담당하는 핵심영역임을 알 수 있었다. 최근 중요한 사회적 문제가 되고 있는 중독 (약물, 음식, 게임) 문제에 대하여 신경질환 기전 모델을 제시하고자 한다.
‣D2R과 EGFR의 상호조절에 ADAM10과 ADAM17이 특이적으로 관여하고 있음을 확인함으로써,뇌의 서로 다른 회로의 소통에 대하여 분자적, 세포적 수준에서의 시스템적 조절원리에 대하여 이해하여 관련 질환의 치료 약제 개발에 있어 차별화된 표적 인자를 제시할 수 있게 되었다.
‣도파민성 신경질환과 관련된 주의력결핍 과다행동장애, 사회공포증, 충동성에 대한 행동들에 대하여 D2R이 관여하고 있음을 확인함으로써, 사회적 정신질환에 대한 분자기전 이해가 가능하게 되었다.

Dopamine is the main catecholamine in the brain and serves important regulatory role in diverse neural functions and is involved in a variety of psychiatric disorders including drug abuse, schizophrenia, depression. To identify the specific behavioral and physiological target of this crucial player of dopaminergic system, we propose here to examine the role of dopamine D2R-mediated ERK activation and the regulation of EGFR signaling on D2R-mediated ERK activation in amygdala for these behavioral regulation using D2R knockout mice (D2R-/- mice). Understanding how these dopamine system and EGFR system crosstalk to regulate specific dopaminergic disorder will shed light on the elucidation of molecular basis of compulsive, reward related and also impulsive behaviors.
Result
‣We investigated the cross-talk between D2R and EGFR to clarify the role of EGFR signaling in the regulation of ERK activation. Our findings show that D2R-mediated EGF receptor transactivation regulates ERK activation via ADAM10/17.
‣Immunohistochemical staining image with EGFR shows that D2R and EGFR co-express in the central amygdala (CeA) and the ventral tegmental area (VTA) of an adult D2R-eGFP mice brain. However, in D2R-/- mice, stereological cell counting analysis showed that the number of EGFR positive neurons were not different beween WT and D2R-/- mice.
‣The absence of D2R in the brain decreases the level of attention and increases the level of anxiety and impulsive behaviors. After cued conditioning test, D2R-/- mice showed higher ERK phosphorylation in the CeA than WT mice. Oppositely, after 5-CSRTT, ERK phosphorylation in the CeA of D2R-/- mice was lower than WT mice.
‣To investigate D2R-EGFR signaling more accurately in the adult mice, we generated D2RΔEGFR mice. Reward earning during the 5-CSRTT decreased in D2RΔEGFR mice, but any other behaviors such as anxiety, impulsivity, locomotor activity were not altered. Treatment with GM6001 before the 5-CSRTT reduced reward earning and ERK phosphorylation in the CeA.
Expected Contribution
‣From our recent study, dopaminergic system leading to alterations in EGFR signaling via ADAMs in the adult brain could be associated with neurological and psychiatric disorders. Our understanding of EGFR related-DA signaling in the reward circuit could suggest a new molecular and cellular model of reward-associated behaviors in the mesolimbic system.
‣D2R-ADAM-EGFR signaling proposes D2R-dependent dopaminergic pathologies to develop novel therapeutic strategies for neuropsychiatric diseases.
‣By understanding D2R and EGFR expression patterns in the brain, it could show a model of neuronal circuit in the brain and allow to approcach neuropsychiatric diorders newly such as ADHD, social phobia, impulsivity.
