연구의 목적 및 내용
세계의 다양한 국가에서 어패류의 양식이 성행되고 있으나, 질병 발생건수가 현저하게 증가하여 많은 피해를 입힘으로서 선결해야할 중요한 과제가 되고 있다. 특히, 바이러스 병은 약제에 의한 치료가 곤란하므로 적극적인 예방대책인 백신의개발이 절실히 요망되고 있다. 지금까지, 어류바이러스백신으로 불활화 및 subunit 백신의 개발을 위한 연구가 행하여 졌지만, 효과는 기대수준에 미치지 못하는 것으로 판단되어진다. 한편, 바이러스의 생백신은 효과를 기대할 수 있을 것으로 기대됨에도 불구하고 양식 환경에의 오염 혹

세계의 다양한 국가에서 어패류의 양식이 성행되고 있으나, 질병 발생건수가 현저하게 증가하여 많은 피해를 입힘으로서 선결해야할 중요한 과제가 되고 있다. 특히, 바이러스 병은 약제에 의한 치료가 곤란하므로 적극적인 예방대책인 백신의개발이 절실히 요망되고 있다. 지금까지, 어류바이러스백신으로 불활화 및 subunit 백신의 개발을 위한 연구가 행하여 졌지만, 효과는 기대수준에 미치지 못하는 것으로 판단되어진다. 한편, 바이러스의 생백신은 효과를 기대할 수 있을 것으로 기대됨에도 불구하고 양식 환경에의 오염 혹은 바이러스병독성의 제어 등의 문제로 세계적으로 연구되고 있지 못하는 것이 현실이다. 본 연구에서는 능성어의 NNV, 넙치의 VHSV 및 돌돔의 RSIV 를 모델로 하여, 어류의 사육온도와 어류바이러스의 병독성의 관계를 상세하게 검토하여, 어류바이러스가 감염 어체내에서 증식은 가능하지만 병독성을 나타내지 않는 온도를 찾아내고, 이들 온도 조건에서 사육한 어체에 바이러스를 감염시켜 바이러스에 대한 특이면역이 유도되는 것을 증명함으로서, 어류바이러스병의 생백신법을 확립하고자 하였다.
연구결과
본 연구는 넙치의 VHSV 감염증, 능성어의 NNV 감염증 및 돌돔의 RSIV 감염증을 모델로 병원 바이러스를 사용한 사육온도 관리형의 어류 면역법 (병원성 바이러스 생백신법)의 개발과 그 유효성에 대한 연구로서 본 연구를 통하여 이하의 성과를 얻었음.
1. 넙치의 바이러스성출혈성패혈증 (VHSV 감염증) : VHSV의 넙치에 대한 병원성은 사육온도의 상승과 동반하여 높아지며 21°C 이상의 사육수온의 경우 VHSV 감염에 따른 사망은 일어나지 않음. 또한, 21°C에서 사육한 넙치의 반수 치사량(LD50)의 100배가 되는 농도로 VHSV를 근육주사법 또는 침지법으로 투여하는 방법을 통하여 VHSV에 대하여 절대적인 감염방어능력이 유도되어짐이 확인되어짐. 나아가 넙치 체내의 VHSV는 넙치를 25°C로 사육하는 것으로 신속히 체내에서 배제되어지는 것이 확인됨.
2. 능성어의 바이러스성신경괴사증 (NNV 감염증) :NNV의 능성어에 대한 병원성은 사육온도가 하강되어짐에 따라 낮아지며 17°C 이하에서 사육함에 따라 NNV 감염에 따른 폐사는 확인되지 않음. 또한 17°C에서 사육하는 능성어에게 LD50의 100배량이 되는 농도로 NNV를 침지감염 또는 근육내접종법을 적용하여도 NNV에 대하여 매우 강한 감염방어능이 유도되어짐이 확인됨. 또한, 17°C로 사육한 능성어에 NNV를 접종하면, NNV는 감염방어능을 획득하기에 필요한 임계레벨 (threshold level, 104TCID50/g) 이상까지 증식하지만, 치사에 필요한 감염가 까지는 증식하지 않음이 확인됨.
3. 돌돔의 이리도바이러스감염증 (RSIV 감염증) :RSIV의 돌돔에 대한 감염성은 사육수온이 28- 30°C일 때 가장 강하고, 사육수온이 하강하여 18°C 이하의 조건에서 사육한 돌돔은 RSIV의 감염에 의한 사망이 확인되지 않음. 그러나, 18°C에서 사육한 돌돔의 체내에는 RSIV가 확실히 감염되어 threshold level (105.4genome/mg)을 상회할 정도로 증식하지만, 폐사에 이르는 정도까지는 증식하지 않음이 확인됨. 이상의 결과로부터 어류의 사육온도 관리조건에서 병원 바이러스를 직접 체내에 투여하는 방법에 의해 대상어류를 죽이지 않고도 체내에 해당 병원체에 저항하는 매우 강한 방어면역을 유도 할 수 있음을 본 연구를 통하여 최초로 확임함.
연구결과의 활용계획
본 연구를 통하여 어류의 사육온도관리에 의해 바이러스의 병독성 제어가 가능한 것을 확인시킴과 동시에, 그 성과를 생백신에 응용하여, 앞으로의 어류양식에 있어서 무엇보다도 중요한 바이러스병의 예방 및 피해의 경감에 공헌하고자 함. 본 연구에 의해, 어류 바이러스의 생백신법이 확립되면 어류백신개발의 큰 전환점을 제시하게 됨. 본 연구를 통하여 확인된 생백신은 개발비용이 낮고, 여러가지 어종 및 바이러스에 응용가능하며, 높은 예방 효과를 기대할 수 있는 방법으로, 생백신이 실용화되면 어류양식에 있어서의 바이러스성 질병에 의한 피해가 대폭 경감되어 수산업발전에 크게 공헌할 수 있을 것으로 기대됨.

A goal of this study is to develop effective vaccines for control of fish virus diseases. Although several kinds of vaccines including inactivated and subunit vaccines have been developed for fish virus diseases, almost no vaccine has been marketed because those protection efficacy might be not enough. Another type of vaccine, "live vaccine" has not been developed for fish virus diseases due to difficulty for control of viral virulence. Recently, I and my colleagues developed a novel immunization method for fish virus diseases using live pathogenic virus together with Poly(I:C). In this procedure, Poly(I:C) is administered to induce interferon in fish to control viral virulence. In this study, we would employ "fish rearing temperature" instead of Poly(I:C) administration for control of viral virulence in infected fish for development of simple and novel live vaccines for fish virus diseases.
Result
In this study, we demonstrated potential of fish immunization with live pathogenic viruses.
It was possible to induce specific protective immune response to VHSV in Japanese flounder by rearing fish at 21°C and delivering live VHSV, regardless of the delivery route. VHSV seemingly was naturally inactivated at 25 °C and was quickly eliminated from the immunized fish reared at 25 °C.
In the 1st NNV-infection, mortalities of sevenband grouper were reduced by decreasing fish rearing temperature, and no mortality was observed in the fish reared at 17 °C regardless of the infection method. Moreover, almost all of the survivors were protected from the 2nd NNV-infection, meaning that those survivors mounted a specific protective immune response against NNV. These data strongly suggest the potentiality of a live NNV vaccine for sevenband grouper by rearing fish at 17 °C. In sevenband grouper inoculated with live NNV vaccine at 17°C, NNV multiplied far beyond the threshold level, but did not achieve the critical level. Furthermore, almost all vaccinated fish survived the challenge with homologous NNV, demonstrating that a protective immune response to NNV was mounted in the fish vaccinated with live NNV.
The optimum temperature for the onset of RSIV infection in rock bream was 28 –30°C. However, when rock bream were reared at a low temperature (≤ 18°C) and received RSIV inoculation, RSIV reliably infected and multiplied slowly, beyond the threshold level for rock bream to mount a protective immune response against RSIV infection (≤105.4genome/mg), but not achieve acritical level (109.0genomes/mg). The survivors, that were received RSIV infection at 18°C, we recompletely protected from re-challenge with homologous RSIV, suggesting the potential for alive RSIV vaccine for rock bream by regulating water temperature.
Expected Contribution
The present study is to reveal that control of viral virulence is possible by regulation of fish rearing temperature in order to apply for development of live vaccine for fish virus diseases, and in the future we would contribute to reduction and prevention of fish viral diseases, the important problems in fish culture industries. Establishment of live vaccines for fish virus diseases by this study will make a major impact to fish vaccine development industries in the world. Also, the live vaccines developed in this study are expected to be high protective efficacy and low cost, and can be applied to various fish species and virus diseases, suggesting that utilization of these live vaccines are essential. We believe that the utilization of these live vaccines will contribute to fish aquaculture industries by largely reducing the economic losses due to fish virus diseases.
