This study was performed to determine the whitening effect of organic solvent extracts from the centipede, Scolopendra subspinipes mutilans. We prepared different concentrations (50%, 70% and 100%) of ethanol, methanol, 100% ethyl acetate and water extracts. We tested melanin inhibitory effect and tyrosinase activity using B16/F10 melanoma cell. As a result, treatment of organic solvent extracts is decreased the biosynthesis of melanin and tyrosinase activity to 36∼86%. Especially the 70% ethanol extracts was the most effective in B16/F10 melanoma cells. In the study on melanogenic protein expression, 70% ethanol extracts of Scolopendra subspinipes mutilans blocked glycosylation of tyrosinase. Therefore this result suggests that 70% ethanol extracts could be developed as skin whitening agents.
The purpose of this study was to investigate antimicrobial, antioxidant, DPPH radical scavenging and SOD-like activity effect of centipede peptides(CP1∼6) from Scolopendra subspinipes mutilans using RNA sequencing. All peptides from Jubak(AFC) had antimicrobial activities in the Candida arbicans, Staphylococcus aureus and Escherichia coli. And the antioxidant was practically effective when concentration of the CP6 was 100 ug/ml. Scavenging effect of DPPH radical of CP6 peptide showed inhibition effect was above 80%. and SOD activity of CP6 peptide was confirmed as a dose-dependent manner.
Inflammation is a important event in the development of chronic inflammatory diseases including arthritis, atherosclerosis, diabetes, and hypertension. The aim of this study was to determine whether it is possible to use Drosophila as a screening tool for anti-inflammatory function. First, we investigated the anti-inflammatory effect of 16 species of insects and one myriapoda ethanol extract on RAW264.7 mouse macrophage and S2 Drosophila cells treated with lipopolysaccharide (LPS) for induction of inflammation. As a result, we selected 4 species of insect, Oxya chinesis sinuosa, Teleogryllus emma, Gametis Jucunda, and Gryllotalpa orientalis as extracts with superior anti-inflammatory effect through down-regulation of inflammatory mediators such as inflammatory cytokine, cyclooxygenase-2, inducible nitro oxide synthase, and antimicrobial peptides (AMPs). To confirm the anti-inflammatory effect of above 4 kinds of insects in vivo, we used AMPs associated genes up- or down-regulated flies induced inflammation by oral administration of dextran sulfate sodium (DSS). As a result, we confirmed G. Jucunda ethanol extract has the highest anti-inflammatory effect among them. These results were consistent with the results in vitro using RAW264.7 and S2 cells. Accordingly, we suggest that it is effective to use flies for 1st screening for a substance having an anti-inflammatory effect, because in vivo screening proceeding was generally spent much time and labor.
