제1세부과제 연구결과 감귤 과육부산물을 이용한 대량 생산용 배지를 개발하여 바이오겔 수율을 30% 이상 증대 시켰으며, 감귤 과육부산물을 활용하여 바이오겔 생산 시 배지의 조성인 설탕물의 함량을 줄임으로써 바이오겔 생산 단가 절감 및 생산량 증가 효과를 볼 수 있었다. 또한 안정적인 대량 생산을 위한 균주 개발을 위해 자외선 조사와 MNNG로 개량된 감귤바이오겔 생성 균주 3종(Gluconacetobacter sp. CRSS13-01, CRSK13-11, CRSK13-12)을 확보하였다. 개량 균주의 대량 배양시 기존 균주보다 30% 이상의 바이오겔 생산량이 증가 및 물성의 변화로 다양한 산업 분야에 적용이 가능할것으로 여겨진다. 생산된 감귤 바이오겔은 Human 세포 3종에 대해 세포독성이 없음을 확인하였다. 제1협동과제에서는 감귤 바이오겔을 기반으로 천연고분자 또는 생체적합성 고분자로 제조되는 조직재생용 감귤 바이오 폼(foam) 제조하였고 그 결과 방사선 가교기술을 이용하여 생체적합성 천연고분자를 포함하는 감귤 바이오겔 기반의 기능성 폼인 gelatin/bacterial cellulose foam 1종을 개발 완료하였다. 또한 방사선 이용 표면활성 감귤 바이오 폼(foam)을 제조하였으며, 이는 방사선 그라프트 기술을 이용하여 표면활성기가 도입된 감귤 바이오 폼인 AAc-g-BC, AEMA-g-BC foam 2종을 개발 완료하였다. 제작된 기능성 폼(foam)에서의 세포증식 및 분화를 증가시키기 위해 생체활성물질이 추가로 도입시켜, 월등한 세포증식 및 흡착 성능을 나타내는 감귤 바이오 폼인 Gel-AAc-g-BC, AEMA-g-BC 2종을 개발 완료하였다. 개발된 4종(AAc-g-BC, AEMA-g-BC, Gel-AAc-g-BC, AEMA-g-BC)의 감귤바이오 폼(foam)제는 세포지지체로서 우수한 친수성과 세포친화성, 기계적 물성 및 서로 연결된 다공성의 내부 구조를 가지고 있으며, 이러한 특성으로 세포증식 및 분화도에서도 우수한 결과를 얻었기에, 향후 조직재생용 세포지지체로서 활용이 가능할 것으로 기대된다. 본 연구과제에서 개발된 감귤 바이오 폼(foam)은 아래와 같은 기술의 성과확산에도 기여하여 미래창조과학부에서 주최한 ‘2014년 연구개발특구 창조 특허기술 박람회’ 에 창조특허기술로 선정되어 출품되었으며(2014년 11월 4일~5일, 대전컨벤션센터), 현재 관련기술은 (주)새남소재, (주)아이튠무역 과 기술이전을 협의중에 있다. 제2협동과제에서는 혼합미생물 배양을 통한 생산효율 증대연구를 수행하여 Lactobacillus plantarum KCCM 80077등 3종의 유산균을 분리하였으며, 분리된 유산균을 혼합한 배양액을 이용한 BC 생산성 증대방법 개발(특허출원 및 논문투고)하였다. 바이오겔의 식품소재화 활용연구 및 제품개발 연구 결과 바이오겔의 침전도 실험을 통해 현탁 안정성을 확인하였으며 음료제형에 첨가 가능한 소재 개발하였으며, 이를 이용하여 향료와 설탕을 넣지 않고 감귤 바이오겔과 제주산 한라봉/블루베리를 첨가하여 한천 및 젤라틴으로 겔화시킨 큐브형태의 젤리 2종 개발하였다. 또한 향장품으로의 바이오겔의 소재 활용연구 및 제품개발 연구 결과 8종의 바이오셀룰로오스 모델링팩을 개발(특허출원_피부보습용조성물) 하였으며, 모공수축에 도움을 주는 화산석송이(특허보유)와 피부보습 효과를 보이는 바이오셀룰로오스를 혼합한 모공팩 및 에센스 개발하였다.

The citrus fruits have long been valued as part of a nutritious and tasty diet. The flavors provided by citrus are among the most preferred in the world, and it is increasingly evident that citrus not only tastes good, but is also good for people. It is well known that the citrus and citrus products are a rich source of vitamins, minerals, flavonoid and dietary fiber (non-starch polysaccharides) which have the effect of anti-cancer, anti-allergy, anti-virus, and anti-inflammatory effect.
Gluconacetobacter sp. KCG326 and Gluconacetobacter sp. SEA623-2 were previously identified as biocellulose producer from combucha and citrus fruit juice. Bacterial cellulose(BC) produced by Gluconacetobacter sp. KCG326 and SEA623-2 have hard and soft physical properties of tensile strength and a high water retention value. The development of high-producing strains is a prerequisite for the efficient biotechnological production of biocellulose. Strain improvement is a powerful tool in commercial development of microbial fermentation processes. The strain of Gluconacetobacter sp. KCG326 and Gluconacetobacter sp. SEA623-2 were subjected to mutation involving treatment of NTG(N-methyl-N-nitro-N-nitrosoguanidine) for 20min followed by UV-irradiation for 60min. The mutant were compared with wild type for biocellulose production. The mutant CRSK13-11 and CRSK13-12 of Gluconacetobacter sp. KCG326 showed higher cellulose yield than the wild strain. And The mutant CRSS13-01 of Gluconacetobacter sp.SEA623-2 showed higher cellulose yield than the wild strain. Highest biocellulose production in stationary culture was obtained by mutant CRSK13-11, CRSK13-12, and CRSS13-01, which were 30% increased in compared to the wild strain, KCG326 and SEA623-2, respectively. After mutation, this newly improved strain showed tremendous indusrial potential for biocellulose production. The cellulose produced by the selected bacteria could be used for the suitable cosmetic and medical materials. The cytotoxicity of the BC was examined to determine the effective concentration for treatment. Exposure to 100 mg/mL of BC for 24hr did not affect the viability of the THP-1, HDFn and HaCaT.
In this project, the high-functional bio-foam based on the citrus bio-gel were developed using by rariation technology for tissue engineering scaffolds. where these functional bio-foam with natural polymers than the functional bio-foam prepared using the synthetic polymer is more biocompatible, it has problem in low mechanical properties and the polymer itself is easily dissolved by the body fluids. Therefore, most of the natural polymer material is generally improved the mechanical properties through chemical treatment, such as cross-linking agent.
In this study, using a radiation instead of cross-linking agent and a chemical treatment to prepare the bio-functional foams and surface activated foams with citrus-gel for tissue engineered scaffolds. These bio-foams were evaluated the physical, chemical and biological properties to confirm the bio-availability for tissue engineering scaffold.
This study was designed to develop a efficient mass production technology of bacterial cellulose and processed food using bacterial cellulose. In this study, Gluconacetobacter sp. gel_SEA623-2 isolated from citrus that produces bacterial cellulose was used to examine the effect of initial concentration of acetic acid and mixed culture inoculated with Lactobacillus plantarum KCCM 80077 on productivity of bacterial cellulose. In single culture(Gluconacetobacter sp. gel_SEA623-2), although bacterial cellulose productivity was igher when the initial concentrations of acetic acid were 0.0% and 0.5%, than when it was 1.0%, there was no significant difference. However, in mixed culture(Gluconacetobacter sp. gel_SEA623-2 and Lactobacillus plantarum KCCM 80077), adding 0.5% acetic acid resulted in dry weight of 37.83±6.81 g/L and thickness of 10.33±0.58 mm, showing a significant difference from that of single culture added with 1% acetic acid, 28.40±1.23 g/L and 7.50±0.50 mm (p<0.05). In addition, We have developed foods and cosmetics using the bacterial cellulose produced as a result of the study.
