한국 근해와 열대 태평양 등 다양한 해양환경에 서식하는 265 여 종, 2,400 kg의 저서군체동물 (해면, 원색동물 등)을 체계적으로 채집, 분류하고 세포독성, 항미생물, 효소활성저해 등의 생리활성검색에 의하여 50 종의 유용시료를 파악. 이들 중 일차적으로 15 종의 해면 및 군체멍게 시료로부터 202 종의 천연물을 분리, 정제하였고 이들 중 130 종의 신물질의 구조를 분광학과 화학적 방법으로 규명하였음. 다양한 해양동식물과 해저 퇴적물로부터 2,689 여종의 방선균과 진균을 분리, 순수배양, 보존하였으며 이 과정에서 심해퇴
한국 근해와 열대 태평양 등 다양한 해양환경에 서식하는 265 여 종, 2,400 kg의 저서군체동물 (해면, 원색동물 등)을 체계적으로 채집, 분류하고 세포독성, 항미생물, 효소활성저해 등의 생리활성검색에 의하여 50 종의 유용시료를 파악. 이들 중 일차적으로 15 종의 해면 및 군체멍게 시료로부터 202 종의 천연물을 분리, 정제하였고 이들 중 130 종의 신물질의 구조를 분광학과 화학적 방법으로 규명하였음. 다양한 해양동식물과 해저 퇴적물로부터 2,689 여종의 방선균과 진균을 분리, 순수배양, 보존하였으며 이 과정에서 심해퇴적물 채집 장치를 고안, 제작하였음. 추출물에 대한 화학적, 생화학적 분석을 종합하여 다수의 고역가 세포독성 및 항미생물 활성균주를 파악하고 대량배양된 균주로부터 신물질 106 종을 포함하는 312 종의 천연물 분리 완료하고 생리활성을 검색하였음. 이와 같이 천연물 신약 소재의 개발을 위한 다수의 생물시료와 함께 514 종의 화합물을 확보하였고 그중 세포독성, 항미생물 활성이 강한 Hit 화합물 75종을 얻을수 있었음. 종합하여 본 최종 연구결과로 해양생물의 천연물신약 개발가능성을 입증하였으며 해양신물질 개발을 위한 기술적 기반을 확립하고 향후의 산업화를 위한 천연물 라이브러리의 골격을 구축하였고 고역가 천연물의 대량 분리를 완료하였음.

1. Benthic colonial invertebrates have been systematically collected from various locations of Korea, Micronesia and Indonesia. These samples weighing more than 2,400 kg, have been classified into more than 265 species of sponges and tunicates. Fifty specimens have been selected for chemical investigation on the basis of combined results of bioactivity tests and TLC and proton NMR analysis of the crude organic extracts and partitioned fractions.
2. Secondary metabolites have been isolated from selected animals utilizing various laboratory techniques and chromatographic methods. From 15 sponges and colonial tunicates, 202 compounds have been isolated and 130 new ones have been structurally elucidated by combined chemical and spectral methods. These compounds have been derived from various biogenetic origins and have belonged to various structural classes: linear and cyclic di- and sester-terpenes, alkaloids, saponins, fatty acid derivatives, cyclic peptides sphingolipids, heteroaromatics, macrolides, mixed biogenetic products, etc. Several compounds displayed significant cytotoxic, antifungal, and/or enzyme-inhibitory activities.
3. More than 2,689 microbial strains including 1,342 actinomycetes and 1,347 fungi have been isolated from various marine organisms and sediments of diverse locations. For this purpose, a device to collect deep sea sediments has been designed and manufactured. These strains have been categorized and stored for future research. Strains exhibiting unique chemical profile and potent bioactivity have been selected and cultivated for chemical investigation. Up to date 514 compounds have been isolated and 236 new metabolites have been structurally elucidated by combined spectroscopic, chemical and crystallographic methods. The structures of these compounds are diverse and includes those from various biogenetic origins: fatty acid derivatives, polyketides, modified bases, macrolides, polyaromatics, macrolides, cyclic peptides, etc.
4. In short, more than 265 species of sponges and tunicates weighing 2,400 kg, have been systematically collected from various locations of Korea, Micronesia and Indonesia and more than 2,689 microbial strains including 1,342 actinomycetes and 1,347 fungi have been isolated from various marine organisms and sediments of diverse locations. From the selected marine inverterbrates and microbial strains, 514 compounds have been isolated and 236 new ones have been structurally elucidated to culminate a marine natural product library.
5. For the advanced works such as chemical derivatization and bioactive mechanism, psammaplin A, a potent anticancer agent derived from sponge was isolated in large scale (>1 g). Also isolated large scale (>1 g) were anticancer agent halichondramide A and pectenotoxin-2. In addition, large scale isolation of fungi-derived metabolites and ohmyungsamycin A derived from actinomycetes which showed potent cytotoxicity and anti-microbial acitivity was developed and accomplished.
