Ⅴ. 연구개발결과
1. 김치류 중 아플라톡신 분석법 확립
- 김치 및 원부재료 중 아플라톡신 분석을 위한 선별방법으로 현재 시판중인 ELISA kit 적용가능성을 검토하였음(정성 : RevealⓇ, Agri-ScreenⓇ/정량 : VeratoxⓇ)
- 시료 전처리 조건을 확립하기 위하여 AOAC법 및 식품공전법에 준하여 추출용매, 추출방법을 검토하였으며, 김치 원부재료 및 제품 중 아플라톡신을 추출하기 위한 전처리 방법으로 추출용매는 김치 원부재료의 경우 1
Ⅴ. 연구개발결과
1. 김치류 중 아플라톡신 분석법 확립
- 김치 및 원부재료 중 아플라톡신 분석을 위한 선별방법으로 현재 시판중인 ELISA kit 적용가능성을 검토하였음(정성 : RevealⓇ, Agri-ScreenⓇ/정량 : VeratoxⓇ)
- 시료 전처리 조건을 확립하기 위하여 AOAC법 및 식품공전법에 준하여 추출용매, 추출방법을 검토하였으며, 김치 원부재료 및 제품 중 아플라톡신을 추출하기 위한 전처리 방법으로 추출용매는 김치 원부재료의 경우 1% NaCl이 첨가된 70% methanol 용액으로, 김치 제품은 70% methanol 용액을 사용하여 추출방법으로는 초음파추출기를 사용한 방법을 선정하였음
- 아플라톡신류를 신속하게 분석하기 위해 u-HPLC/FLD 기기분석 조건을 확립하였음
- 확립된 분석법은 FDA guideline에 따라 검량선의 직선성, 검출한계(LOD), 정량한계 (LOQ), 재현성 및 회수율을 산출하여 유효성 검증을 실시하였음
2. 수출용 김치의 원부재료 및 완제품 중 아플라톡신의 계절별 오염 현황 조사
- 수출용 김치의 원부재료 및 제품 중 아플라톡신 오염 현황 조사
∙수출용 김치 생산업체 현황은 식품의약품 통계자료를 활용하여 확인하였으며, 종류별 수출 현황 및 사용 원료의 원산지를 확인하였음
∙수출용 김치 원부재료 및 제품 51종(김치 16종, 원부재료 35종)을 수집하여 이화학적,미생물학적 품질 특성을 분석하고 아플라톡신 오염도를 분석하였음
∙시판 김치 및 원부재료는 총 149종(김치 21종, 고춧가루 103종, 기타원부재료 25종)을 수집하여 이화학적, 미생물학적 품질 특성을 확인하고 아플라톡신 오염도를 분석하였음
- 수출용 김치의 원부재료 및 제품 중 아플라톡신의 계절별 발생현황 조사
∙계절별 김치 원부재료 총 195종(건고추 및 고춧가루 138종, 기타 부재료 57종)을 수집하여 품질 특성(수분, 일반세균, 젖산균, 효모/곰팡이)을 확인하고 아플라톡신 오염도를 분석하였음. 그 결과 총 195종 원부재료 중 에서 32종(전체의 16.4%)의 원부재료에서 아플라톡신 B1이 검출되었으며, 26종이 건고추 및 고춧가루에서 검출되었음. 검출된 원부재료 중 고춧가루 12종에서 0.09 ∼ 2.56 μg/kg 농도로 오염된 것으로 나타났으며, 모두 식품공전 기준(장류 및 고춧가루 및 카레분, 아플라톡신 B1 10 μg/kg 이하) 이하로 확인되었음
∙계절별로 판매되는 김치 제품 총 87종을 수집하여 이화학적(수분, pH, 산도, 염도), 미생물학적(일반세균, 젖산균, 효모/곰팡이)품질 특성을 확인하고 아플라톡신 오염도를 분석하였음. 그 결과, 87종 중 8종에서 아플라톡신 B1이 검출되었으며, 모두 LOD이하로 확인되었음
3. 수출용 김치의 원부재료 및 완제품 중 아플라톡신 오염 원인 규명
- 수출용 김치의 제조공정 및 환경 중 아플라톡신 발생 현황 조사
∙수출용 김치를 생산하는 업체 6곳을 선정하여 제조공정별(3단계) 시료 18종을 수집하였음
∙공정별 시료의 이화학적, 미생물학적 품질 특성을 분석하고, 아플라톡신 함량을 분석하여 오염여부를 확인하였음. 제조공정별 시료의 아플라톡신 함량을 분석한 결과, 18종 모두에서 아플라톡신이 확인되지 않아 미생물 특성 결과와 일치하는 것으로 나타났다. 수출을 전문으로 하는 제조업체에서 생산된 제조공정별 시료에서 아플라톡신 오염은 없는 것으로 확인되었음
- 김치의 저장·유통 중 아플라톡신 발생 현황 조사
∙아플라톡신 생산 균주(Aspergillus flavus 및 A. parasiticus 혼합)를 고춧가루에 접종하여 저장조건(25°C, 35°C, 37°C, 동일 RH 75%) 중 아플라톡신의 생산 유무 및 함량을분석하였음. 저장기간 동안 저장온도별(25°C, 35°C 및 37°C)로 생성된 총 아플라톡신류 의 생성량은 각각 평균 6.2, 7.6 및 3.5 μg/kg으로 35°C 저장 시 아플라톡신 생성량이많은 것으로 확인되었음
∙김치는 포장형태(용량, 소재) 및 유통형태(온라인, 오프라인)으로 구분하여 아플라톡신 오염 여부를 확인하였음. 포장형태별 김치 제품의 아플라톡신 오염 현황을 확인한 결과, 아플라톡신이 검출된 제품 8종 중 PET 포장제품 2종(- 김치 중 아플라톡신 오염원인 규명
∙김치의 원부재료 및 제품, 제조공정별 시료 중 아플라톡신 함량을 모니터링 한 결과 부재료 중 건고추 및 고춧가루에서 아플라톡신이 오염되는 것으로 확인되었음. 아플라톡신 생성균주를 처리한 후 저장 조건별로 아플라톡신 생성유무 및 생성량을 확인한 결과, RH 75% 시 생성량의 차이가 발생하지만 25°C, 35°C 및 37°C 조건에서 아플라톡신을 생산하는 것으로 나타났음
4. 수출용 김치의 원부재료 및 완제품 중 아플라톡신 오염 방지 기술 개발
- 아플라톡신 생성 억제를 위한 김치 원부재료 저장조건 설정
∙아플라톡신 생성 억제를 위한 김치 원부재료 저장조건 설정을 위해 아플라톡신 오염에 취약한 고춧가루를 대상으로 아플라톡신 생성균주를 오염시킨 후 온도 및 습도조건별로 저장하면서 아플라톡신 함량 변화를 확인하였음
∙A. flavus를 오염시켜 온도(10, 25 및 35°C) 및 습도(RH 35, 55 및 75%) 저장조건별로 아플라톡신 함량을 확인한 결과, 아플라톡신 B1이 검출되었음. 동일 온도(25°C)에서 습도가 낮을수록 아플라톡신 B1이 저장기간이 경과함에 따라 급격히 감소하였으며, 저장7일째는 모두 불검출 되었음. 동일 습도(RH 75%)에서 온도가 높아질수록 아플라톡신생성량이 높아졌으며, 35°C에서 생성량이 높은 것으로 확인되었음
∙A. parasitivus를 오염시켜 온도(10, 25 및 35°C) 및 습도(RH 35, 55 및 75%) 저장조건별로 아플라톡신 함량을 확인한 결과, 아플라톡신 B1 B2, G1 및 G2가 모두 검출되었음.
총 아플라톡신 생성량은 A. flavus를 오염시킨 경우 보다 A. parasiticus에서 더 높게 확인되었으며, 아플라톡신 B1의 경우 10배 이상 높게 확인되기도 하였음. 동일 온도(25°C)에서 습도의 영향을 확인한 결과, RH 55%에서 가장 높은 생성량을 나타내었으며, 저장기간에 따라 감소하는 경향은 확인되지 않았음. 동일 습도(RH 75%)에서 온도 에 따른 생성량을 확인한 결과 25°C에서 높게 확인되었으며, 10°C 및 35°C 저장 시료에서는 유사한 함량으로 나타났음
∙결과적으로, A. flavus가 오염된 경우 10°C이하에서 저장할 경우 아플라톡신의 오염을 예방할 수 있었으며, A. parasiticus가 오염된 경우에는 생성균수에 따라 아플라톡신 함량이 증가하는 경향을 나타내어 초기 오염을 방지하고, 저온 및 RH 50% 이하에서 저장하는 것이 바람직 한 것으로 나타났음
- 김치 원부재료 및 제품 중 아플라톡신 생성 억제 및 저감효과 분석
∙김치 원부재료 및 제품 중 아플라톡신의 생성 억제 및 저감효과 분석을 위해 아플라톡신 오염에 취약한 고춧가루, 고춧가루가 첨가된 김치 양념 및 김치제품을 대상으로 아플라톡신 생성량이 높은 A. parasiticus를 오염시켜 물리적(적외선, UV, 감마선), 화학적(유기산, 자몽종자추출물) 및 미생물학적(젖산균) 처리 후 A. parasiticus 수 및 아플라톡신 함량 변화를 분석하였음
∙아플라톡신 생성 균주인 A. parasiticus의 포자 현탁액을 멸균된 고춧가루에 접종하여 30°C에서 16시간 및 96시간 배양 후 적외선(230V, 2000W)을 1, 2, 3분 동안 각각 처리하여 A. parasiticus 수 및 아플라톡신 함량의 변화를 확인하였음. 배양시간에 따라 4 및 8 log CFU/g 수준으로 나타난 A. parasiticus수는 적외선으로 1분 조사 후 2 log CFU/g 수준까지 감소하였으며, 2분 이상 조사 시 검출되지 않았음. 아플라톡신 함량은 초기 오염농도 수준에 따라 감소율이 다르게 나타났으며, 적외선 조사 시간에 따라 아플라톡신 B1과 G1의 감소로 총 아플라톡신 함량이 낮아지는 것으로 확인되었음. 적외선으로 3분 조사 시 전체 감소율은 저농도(4.41 μg/kg)에서는 약 7.7%, 고농도(111.2 μg/kg)에서는 약 11.5%이었음.
∙아플라톡신 생성 억제 및 저감효과 분석을 위해 UV-LED(275 nm)장치를 제작하였으며, 멸균 고춧가루에 A. parasiticus 포자 현탁액을 접종하여 배양한 시료를 장치에 넣고 1, 2 및 3시간 동안 UV를 조사하였음. A. parasiticus 수는 UV 조사 후 1 log 감소하였으며, 조사시간에 따른 차이는 확인되지 않았음. 아플라톡신 함량의 경우, UV 조사후 아플라톡신 B1이 약 26%, G1이 약 22% 감소하여 총 아플라톡신 함량은 평균 25%수준 감소하였으며 처리시간에 따른 차이는 나타나지 않았음
∙감마선 조사에 의한 아플라톡신 생성균의 생육억제 및 아플라톡신 함량 저감 효과를 확인하기 위하여 멸균된 김치 및 양념에 A. parasiticus 포자 현탁액을 오염시켜 배양한 후 5, 10 및 20 kGy로 감마선을 조사하였음. 김치 및 양념에서 A. parasiticus 수(조사 전 4.6 log CFU/g 수준)는 5 및 10 kGy 감마선 조사 후 약 2 log 수준이 감소하였으며, 20 kGy로 조사된 시료에서는 모두 불검출 되었음. 총 아플라톡신 함량은 김치및 양념 모두에서 10 kGy 이상 조사 시 유의적으로 감소하는 것으로 나타났으며 (p<0.05), 20 kGy 조사 처리구에서 감소율은 김치 및 양념에서 각각 8.4 및 11.5%이었음.
∙화학적 처리에 의한 아플라톡신 생성균의 생육억제 및 아플라톡신 저감 효과를 확인하기 위하여 유기산(acetic acid(AA) 1.0%, lactic acid(LA) 1.0%) 및 자몽종자추출물(0.3,0.5 및 1.0%)을 A. parasiticus가 오염된 양념에 처리하여 배양(30°C, 42시간) 후 균수 및 아플라톡신 함량을 측정하였음. A. parasiticus 수(무첨가구 6.4 log CFU/g)는 유기산 첨가구에서 약 2-3 log 감소한 것으로 나타났으며, 자몽종자추출물은 무첨가구와 유사하게 나타나 아플라톡신 생성균의 생육억제 효과가 없는 것으로 확인되었음. 총 아플라톡신 함량은 무첨가구와 비교하여 AA 첨가구에서 약 5.1%로 감소한 것으로 나타났으며, LA 처리구 및 자몽종자추출물 처리구에서는 평균 6.5% 증가하는 것으로 나타나아플라톡신 저감효과가 확인되지 않았음
∙미생물 처리에 의한 아플라톡신 생성균 생육 억제 및 아플라톡신 저감효과를 평가하기 위하여 멸균된 김치에 A. parasiticus 포자 현탁액을 접종하고 김치 발효에 관여하는 주요 젖산균 중 Lactobacillus plantarum (LP)과 Leuconostoc mesenteroides (LM)를 각각 접종시켜 30°C에서 배양하며 배양시간(0, 18 및 42시간)에 따른 화학적 및 미생물학적 특성, 아플라톡신 함량을 분석하였음. 배양시간이 경과함에 따라 시료의 pH는 낮아지고, 산도는 증가하는 것을 확인하였으며 젖산균 수도 증가하는 것으로 나타났음. A. parasiticus 수는 젖산균 무처리구 및 LM 처리구에서 배양시간 동안 5 log CFU/g을 유지하는 것으로 나타났으며, LP 처리구에서는 발효 진행에 따른 화학적 변화에 따라 pH가 3.4까지 내려간 시점에서 1 log 감소한 수준으로 확인되었음. 총 아플라톡신 함량의 경우 배양시간이 증가함에 따라 젖산균 무처리구에서는 초기 농도보다 약 2배이상 증가한 것으로 나타났으나, 젖산균 처리구에서는 초기 오염농도와 유사하거나 낮은 수준으로 확인되었음. 결과적으로 김치 유래젖산균이 A. parasiticus의 생육억제 효과는 낮지만, 아플라톡신의 생성량을 50%이상 저감시키는 것으로 확인되었음
- 김치 발효조절에 의한 아플라톡신 생성 억제 효과분석
∙김치 발효 중 농도별 아플라톡신 오염도 저감 효과를 확인하기 위하여 아플라톡신 농도(10 및 20 μg/kg)를 달리하여 김치에 오염시킨 뒤 저장 중 이화학적 및 미생물학적 품질특성을 분석하고, 아플라톡신 함량을 분석하였음. 아플라톡신 B1, B2, G1. G2 및 총아플라톡신 함량은 저장기간이 증가하면서 점차 감소하는 경향을 나타내었으며, 초기 오염농도에 비해 평균적으로 약 49%의 감소율을 나타냄. 특히 아플라톡신 G1의 경우저장 0주차에 10 μg/kg 오염구에서 7.79 μg/kg, 20 μg/kg 오염구에서 17.85 μg/kg이 검출되었으나 저장 8주 후 각각 2.83, 8.55 μg/kg으로 감소하여 60% 이상의 감소율을 나타냄
∙김치 발효 중 젖산균이 아플라톡신 오염도 저감화에 미치는 영향을 평가하기 위하여 아플라톡신 기준농도인 10 μg/kg를 김치에 오염시킨 뒤 김치 발효에 대표적으로 관여하는 젖산균인 Lactobacillus plantrum(AFs+LP)과 Leuconostoc mesenteeriods (AFs+LM)를 각각 접종시켜 4°C에서 8주간 저장하며 이화학적(수분, pH, 산도, 염도) 및 미생물학적 특성(일반세균, 젖산균, 효모 및 곰팡이, 대장균군 및 대장균), 아플라톡신 함량을 분석하였음. 저장 8주 후 아플라톡신 오염 및 젖산균을 첨가한 김치의 아플라톡신 평균 감소율은 아플라톡신 오염구(AFs)에서 7.1%, AFs+LP에서 21.5%, AFs+LM에서 24.1%로 나타나 젖산균을 첨가한 시료에서 감소율이 더 크게 나타남
∙발효조건에 따른 아플라톡신 생성균의 생육억제 및 아플라톡신 오염도 저감 효과를 확인하기 위하여 A. parasiticus 포자 현탁액을 김치에 오염시킨 후 저장온도(4°C, 10°C, 10°C→4°C)를 달리하여 저장 중 이화학적 및 미생물학적 품질 특성을 분석하고, 아플라톡신 함량을 분석하였음. 효모 및 곰팡이 수는 저장 1주차에 10°C→4°C 저장조건에서 2.77 log CFU/mL로 검출되어 약 2.71 log CFU/mL 수준이 감소하였음. 저장 2주차에는 효모 및 곰팡이 수가 2.48∼3.58 log CFU/mL 검출되어 저장 초기에 비해 약 0.30∼1.43 log CFU/mL 감소하였으며 저장 3주차 이후 모든 저장조건에서 효모 및 곰팡이수가 불검출 되었음. 저장조건에 따른 총 아플라톡신 함량은 저장 초기 약 20 μg/kg 수준으로 검출되었으나 저장 2~3주차까지 증가하여 약 25 μg/kg 까지 검출되었고, 이후다시 감소하는 경향을 나타냄. 4℃ 저장구의 경우 저장기간에 따라 아플라톡신 B1에서 유의적으로 감소하여 저장 3주차에 약 14%, 저장 4주차에 약 4%의 감소율을 나타냄
- 아플라톡신 등 곰팡이독소 생성균과 기타 미생물 제어를 위한 방법으로 식품의 재료를 혼합하면서 유해 미생물을 효과적으로 살균, 제거할 수 있는 살균기능을 갖는 식품혼합장치를 고안하였음
5. 수출용 김치의 안전성 확보를 위한 아플라톡신 안전관리 매뉴얼 개발
- 원료 구입, 검수, 보관 등 김치 원료의 관리 및 정선, 탈수, 양념제조 등 배추김치 제조단계별 관리항목을 제시하고, 일반위생관리 항목으로 작업장 관리, 제조환경 관리, 작업자위생 관리 등 아플라톡신이 오염될 수 있는 경로차단에 중점을 둔 지침서를 제안함
- 매뉴얼에는 김치의 안전생산을 위한 농산물 품질관리를 위해 우수농산물 관리제도(GAP) 실천, 농산물 이력추적관리에 대한 내용을 포함하여, 김치 가공이력에 대한 기록 관리를 수행하되 주원료 및 부재료의 생산 및 유통단계 이력이 연계될 수 있도록 전후방 단계(one-step forward, one-step backward)에 대한 이력기록관리 체계 구축에 활용할 수 있도록 함
- 본 매뉴얼은 책자로 발간하여 중소기업에서 활용할 수 있도록 보급할 계획임
6. 수출용 김치의 품질 보증을 위한 국가 품질안전인증제(안) 마련
- 시스템 인증을 위한 평가기준의 경우 식품위생법에서 정하는 최소기준(제조업체 시설기준, 영업자 준수사항 등)을 충족시키고, 전통식품 품질인증규격의 시스템운영 관련 기준과 소규모업체를 위한 HACCP관리기준 중 선행요건관리 항목(15개 항목)을 포괄적으로 수용하며, 여기에 HACCP 개념을 포함하는 수준에서 시스템 인증기준의 제정이 필요함
- 수출경쟁력 강화 또는 수입산 제품의 위생수준 강화를 위해 교역현장에서 문제가 될 수 있는 항목들을 중요관리점(CCP) 항목으로 명시하는 방안을 고려함(예를 들어 고춧가루에서의 아플라톡신 유무, 주요 채소원료의 농약잔류기준 부합 여부 등)
- 제품품질 인증기준은 KS나 전통 규격의 품질기준을 고려하여 설정하되 위생이나(사분,이물질 유무) 및 국제교역(고형량)과 관련된 기준은 반드시 포함시키도록 함

1. Establishment of method of analyzing aflatoxin in kimchi
- The applicability of ELISA kit, which is currently available in the market, was examined as a screening method for the analysis of aflatoxin in kimchi and raw ingredients (qualitative: RevealⓇ, Agri-ScreenⓇ/qu
Ⅴ. Results of Research
1. Establishment of method of analyzing aflatoxin in kimchi
- The applicability of ELISA kit, which is currently available in the market, was examined as a screening method for the analysis of aflatoxin in kimchi and raw ingredients (qualitative: RevealⓇ, Agri-ScreenⓇ/quantitative: VeratoxⓇ).
- Extraction solvents and methods were examined according to AOAC and Korean Food Standards Codex to establish sample pretreatment conditions. For the pretreatment for extracting aflatoxin in kimchi raw ingredients and final products, 70% methanol with 1% NaCl was selected as extraction solvent for raw ingredients and 70% methanol for final kimchi products. Ultrasonic extractor was selected as extraction method.
- The u-HPLC/FLD instrumental analysis conditions were established for the fast analysis of aflatoxins.
- A validation test was performed for the established analytical method by calculating the linearity of calibration curve, limit of detection (LOD), limit of quantitation (LOQ), reproducibility, and yield according to the FDA guidelines.
2. Investigation of the current status of aflatoxin contamination in raw ingredients and kimchi for export by season
- Investigation of the current status of aflatoxin contamination in raw ingredients and exported kimchi as final product
∙The current status of companies manufacturing kimchi for export was confirmed by KFDA statistical data, wherein the current export status by type and origin of raw ingredients used was identified.
∙A total of 51 kinds of raw ingredients and exported kimchi as final product (16 kinds of kimchi, 35 kinds of raw ingredients) were collected and analyzed for physicochemical and microbiological quality characteristics as well as aflatoxin contamination.
∙A total of 149 raw ingredients and final kimchi products in the market (21 kinds of kimchi, 103 kinds of red pepper powder, 25 kinds of other raw ingredients) were collected and analyzed for physicochemical and microbiological quality characteristics as well as aflatoxin contamination.
- Investigation of the current status of aflatoxin contamination in raw ingredients and kimchi for export by season
∙A total of 195 kinds of raw ingredients of kimchi by season (138 kinds of dried red pepper and red pepper powder, 57 kinds of other raw ingredients) were collected and identified for quality characteristics (moisture, general bacteria, lactic acid bacteria, yeast/fungi) and analyzed for aflatoxin contamination. The results showed that aflatoxin B1 was detected in 32 out of 195 raw ingredients (16.4%), among which 26 were dried red pepper and red pepper powder. Among the aflatoxin-contaminated raw ingredients, 12 kinds of red pepper powder were contaminated at concentration of 0.09 ∼2.56 μg/kg; all were identified to fall below the Korean Food Code criteria (fermented soybean products and red pepper powder and curry powder, aflatoxin B1 of less than 10 μg/kg).
∙A total of 87 kimchi products by season were collected and tested for physicochemical (moisture, pH, acidity, salinity) and microbiological (general bacteria, lactic acid bacteria, yeast/fungi) quality characteristics, and then analyzed for aflatoxin contamination. As a result, aflatoxin B1was detected in 8 out of 63 kinds, but all of them were identified to fall out below than LOD.
3. Determination of the causes of aflatoxin contamination in raw ingredients and kimchi for export
- Investigation of the current status of aflatoxin contamination in the manufacturing process and environment of kimchi for export
∙6 companies manufacturing kimchi for export were selected, and 18 kinds of samples by manufacturing process (3 steps) were collected.
∙Samples by process were analyzed for physicochemical and microbiological quality characteristics and for aflatoxin content to determine the presence of contamination. The aflatoxin content analysis in samples by process found no aflatoxin contamination in all 18 samples, which was consistent with the result of microbiological characteristics. Thus, samples by process produced in companies manufacturing kimchi for export only were confirmed to have no aflatoxin contamination.
- Investigation of the current status of aflatoxin contamination during the storage and distribution of kimchi
∙The production of aflatoxin and its contents were analyzed by inoculating aflatoxin-producing strains (Aspergillus flavus and A. parasiticus mixture) onto red pepper powder under storage conditions of 25°C, 35°C, 37°C, and same RH 75%. By storage temperature (25, 35, and 37°C), the average total aflatoxins produced during the storage period were 6.2, 7.6, and 3.5 μg/kg, respectively, showing the highest aflatoxin production at 35°C.

4. Development of technology for preventing aflatoxin contamination in kimchi for export
- Setting up the storage conditions for raw ingredients to inhibit the production of aflatoxin
∙To set up the storage condition for raw ingredients of kimchi to inhibit aflatoxin production, samples of red pepper powder vulnerable to aflatoxin contamination were inoculated with aflatoxin-producing strains and stored at different temperature and humidity conditions and monitored for aflatoxin content change.
∙When contaminated by A. flavus and stored at different temperature (10, 25, and 35°C) and humidity (RH 35, 55, and 75%) conditions, aflatoxin B1 was detected. At the same temperature (25°C) with lower humidity, aflatoxin B1 greatly decreased as the storage period increased; it was not detected on the 7th day of storage. At the same humidity (RH 75%) condition, aflatoxin production increased with higher temperature, and it was highest at 35°C.
∙When contaminated by A. parasiticus and stored at different temperature (10, 25, and 35°C) and humidity (RH 35, 55, and 75%) conditions, aflatoxin B1, B2, G1, and G2 were all detected. Total aflatoxin production was higher in A. parasiticus inoculation compared to A. flavus and was particularly more than 10 times higher in the case of aflatoxin B1. The effect of humidity at the same temperature (25°C) showed that production was highest at RH 55%, and that it didn't decrease as the storage period increased. Production by temperature at the same humidity (RH 75%) was higher at 25°C and similar at 10°C and 35°C.
∙Therefore, aflatoxin contamination could be prevented by storage below 10°C in case of A. flavus contamination. Since aflatoxin content tended to increase by the number of bacteria produced in case of A. parasiticus contamination, initial contamination should be prevented, and storage should be done at lower temperature and below RH 50%.
- Analysis of inhibitory and reducing effects on aflatoxin production in raw ingredients and final kimchi products
∙To analyze the inhibitory and reducing effects on aflatoxin production in raw ingredients and kimchi products, samples of red pepper powder vulnerable to contamination, kimchi seasonings containing red pepper powder, and kimchi products were inoculated with A. parasiticus, which can produce higher amount of aflatoxin; after physical (infrared light, UV, gamma ray), chemical (organic acid, grapefruit seed extract), and microbiological (lactic acid bacteria) treatments, the number of A. parasiticus and aflatoxin content changes were analyzed.
∙The spore suspension of aflatoxin-producing A. parasiticus was inoculated onto sterilized red pepper powder and incubated at 30°C for 16 hours or 96 hours, and then treated with infrared light (230V, 2000W) for 1, 2, or 3 minutes to monitor the changes in the number of A. parasiticus and aflatoxin content. The number of A. parasiticus was 4 or 8 log CFU/g depending on the incubation time, decreasing to 2 log CFU/g after 1-minute irradiation of infrared light; it was not detected after at least 2 minutes of irradiation. The aflatoxin content differed in its decrease rate depending on the degree of initial contamination, and the total aflatoxin content was decreased mainly by the decrease in aflatoxin B1 and G1 depending on the time of infrared light irradiation. The overall reduction rate after 3 minutes of infrared light irradiation was about 7.7% at low concentration (4.41 μg/kg) and 11.5% at high concentration (111.2 μg/kg).
∙For the analysis of inhibitory and reducing effects on aflatoxin production, UV-LED (275 nm) apparatus was manufactured. Samples of sterilized red pepper powder were inoculated with A. parasiticus spore suspension and incubated, and then placed in the aforesaid apparatus and treated with UV irradiation for 1, 2, or 3 hours. The number of A. parasiticus decreased to 1 log after UV irradiation, but it did not differ by irradiation time. In case of aflatoxin content, aflatoxin B1 decreased by about 26%, and aflatoxin G1, by about 22% after UV irradiation, for a 25% average decrease in total aflatoxin content; it did not differ by treatment time, however.
∙To identify the growth inhibition in aflatoxin-producing fungi and the decrease effect in aflatoxin content by gamma irradiation, samples of sterilized kimchi and kimchi paste were inoculated with A. parasiticus spore suspension and incubated, and then irradiated with gamma ray at 5, 10, and 20 kGy The number of A. parasiticus (4.6 log CFU/g before irradiation) decreased by about 2 log in kimchi and seasonings after 5 and 10 kGy gamma irradiation; it was not detected in samples with 20 kGy irradiation. Total aflatoxin content significantly decreased in all samples of kimchi and seasonings when irradiated at 10 kGy and higher levels (p<0.05), with 8.4 and 11.5% reduction rates in the 20 kGy irradiation treatment group for kimchi and seasonings, respectively.
∙To identify the growth inhibition in aflatoxin-producing fungi and the decrease in aflatoxin content by chemical treatment, A. parasiticus-contaminated seasonings were treated with organic acid (acetic acid (AA) 1.0%, lactic acid (LA) 1.0%) and grapefruit seed extract (0.3, 0.5, and 1.0%) and incubated (30°C, 42 hours); the number of bacteria and aflatoxin content were then measured. The number of A. parasiticus (6.4 log CFU/g in the non-treated group) decreased by about 2-3 log in organic acid-treated groups but was similar in grapefruit seed extract-treated groups and non-treated groups, suggesting no growth inhibitory effect on aflatoxin-producing fungi. Total aflatoxin content decreased to 5.1% in the AA groups, compared to the non-treated groups, increasing by 6.5% in LA and grapefruit seed extract treatment groups.
∙To identify the growth inhibition in aflatoxin-producing fungi and the decrease in aflatoxin content by microbiological treatments, samples of sterilized kimchi were inoculated with A. parasiticus spore suspension followed by Lactobacillus plantarum (LP) and Leuconostoc mesenteroides (LM) which are major lactic acid bacteria involved in the fermentation of kimchi and incubated at 30°C. The chemical and microbiological characteristics and aflatoxin content by incubation time (0, 18, and 42 hours) were then analyzed. The pH decreased, and the acidity increased in samples as the incubation time increased; the number of lactic acid bacteria also increased. The number of A. parasiticus was maintained at 5 log CFU/g in the non-treatment group and LM treatment group during the incubation time, decreasing by 1 log in the LP treatment group when the pH was reduced to 3.4 due to chemical changes by fermentation. Total aflatoxin content more than doubled compared to the initial concentration in the non-lactic acid bacteria-treated group as the incubation time increased, but it was similar or lower in the lactic acid bacteria-treated group. In other words, kimchi-originated lactic acid bacteria showed lower inhibitory effect on the growth of A. parasiticus but reduced aflatoxin production by more than 50%.
- Analysis of inhibitory effects on aflatoxin production by controlling kimchi fermentation
∙To identify the reduction effect on aflatoxin contamination by concentration during kimchi fermentation, samples of kimchi were contaminated with aflatoxin at different concentrations (10 and 20 μg/kg); the physicochemical and microbiological quality characteristics and aflatoxin content were then analyzed during the storage period. The amount of aflatoxin B1, B2, G1, and G2 and total aflatoxin gradually decreased as the storage period increased, showing about 49% reduction rate compared to the initial contamination concentration. In the case of aflatoxin G1 inparticular, the content was 7.79 μg/kg in the 10 μg/kg contamination group and 17.85 μg/kg in the 20 μg/kg contamination group at 0 week of storage but decreased to 2.83 and 8.55 μg/kg, respectively, after 8 weeks of storage, recording more than 60% reduction rate.
∙To evaluate the effect of lactic acid bacteria on the reduction of aflatoxin contamination during kimchi fermentation, samples of kimchi were contaminated with aflatoxin at the reference concentration of 10 μg/kg, and then inoculated with Lactobacillus plantarum (AFs+LP) and Leuconostoc mesenteroides (AFs+LM), the major lactic acid bacteria involved in kimchi fermentation, and stored at 4°C for up to 8 weeks, during which the physicochemical (moisture, pH, acidity, salinity) and microbiological characteristics (general bacteria, lactic acid bacteria, yeast and fungi, coliform bacteria group and coliform bacteria) and aflatoxin content were analyzed. After 8 weeks of storage, the average reduction rate of aflatoxin in kimchi following aflatoxin contamination and lactic acid bacteria addition was 7.1% in the aflatoxin-contaminated group (AFs), 21.5% in the AFs+LP group, and 24.1% in the AFs+LM group, showing higher reduction rate in lactic acid bacteria-added samples.
∙To identify the growth inhibition in aflatoxin-producing fungi and the decrease in aflatoxin contamination by fermentation conditions, samples of kimchi were inoculated with A. parasiticus spore suspension and stored at different storage temperatures (4°C, 10°C, 10°C→4°C), during which the physicochemical and microbiological quality characteristics and aflatoxin content were analyzed. The number of yeast and fungi was 2.77 log CFU/mL under the 10°C→4°C storage condition at the first week of storage, showing about 2.71 log CFU/mL decrease. At the 2nd week of storage, the number of yeast and fungi was 2.48∼3.58 log CFU/mL, showing about 0.30∼1.43 log CFU/mL decrease compared to the initial storage; they were undetectable in all storage conditions after 3 weeks of storage. The total aflatoxin content by storage condition was about 20 μg/kg at the initial storage but increased until 2~3 weeks of storage up to about 25 μg/kg before decreasing afterward. In the case of the 4℃ storage group, the amount of aflatoxin B1 significantly decreased depending on the storage period, showing about 14% reduction rate at the 3rd week of storage and about 4% at the 4th week of storage.

5. Development of aflatoxin safety management manual for ensuring the safety of kimchi for export
- The guidelines are proposed to suggest management items at each stage of kimchi manufacturing such as management of kimchi ingredients (raw ingredients purchase, inspection, and storage), careful selection, draining, and making of seasonings and to emphasize blocking the possible paths of aflatoxin contamination such as general sanitary control including working area management, manufacturing environment management, and workers’ hygiene management.
- The manual will include Good Agricultural Practices (GAP) and Agricultural Products Traceability for the quality control of agricultural products for safe kimchi manufacturing, to perform record management of kimchi manufacturing history, and for use in the establishment of traceability management in steps (one-step forward, one-step backward) by connecting the history of production and distribution of raw ingredients.
- This manual will be published as a booklet and distributed for use in small and medium businesses.
6. Preparation of National Quality and Safety Certification for the quality assurance of kimchi for export
- For the evaluation standards for system certification, there is a need to satisfy the minimum criteria defined in the Food Sanitation Act (manufacturing companies’facilities standards, business compliances, etc.) and to accept comprehensively the traditional food quality certification management criteria and prerequisite management items (15 items) among HACCP management criteria for small businesses and to establish the criteria for system certification at such level including HACCP concepts.
- To strengthen the export competitiveness or the sanitary standards of imported products, measures for listing problematic items in the trading field as critical control point (CCP) items should be considered. (For example, presence of aflatoxin in red pepper powder, pesticide residue standards in major vegetable ingredients, etc.)
- The criteria for product quality certification should be established considering KS or conventional standard quality criteria but should include criteria related to hygiene (mineral impurities, foreign substances) and international trade (solid weight).
