유해균 특이 항체 개발과제에서는 5종의 유해세균의 surface protein을 대상으로 유해균 특이 monoclonal antibody(MAb; 각 유해세균별 2종 이상)을 개발하고 다양한 특성분석을 진행하였다. 이를 통해 특히 Escherichia coli O157 또는 Staphylococcus aureus와 상대적으로 높은 특이성을 보이는 MAb를 개발하는데 성공하였다.유해균 특이 항체대체제의 개발에서는 Bacillus cereus와 Staphylcoccus aureus를 사멸시키는 박테리오파지를 분리하고, 이들이 암호화하고 있는 endolysin CBD(각 유해균당 3종)을 대량으로 생산하고 다양한 특성을 분석하였다. 종이-미세유체기반 센서의 개발과제에서는 먼저 종이기반 미세유체기술을 개발하고, 이를 활용한 paper-based 센서를 개발하였으며, 더 나아가 5종의 유해미생물의 동시검출이 가능한 multiple paper-based 센서를 개발하였다.
개발된 항체 및 항체대체제를 먼저 immunomagnetic separation(IMS) 기술에 접목하였고, 다양한 유해균이 혼합되어있는 샘플에서도 매우 높은 수준으로 특정 유해균을 특이적으로 분리해 낼 수 있음을 확인하였다.
마지막으로 농산물 종류에 따른 신기술의 현장적용 최적화를 위해 전처리 적용 방법 및 농축방법을 개발하고 이를 작물 및 식중독균에 최적화하였다. 또한 농산물 종류에 따른 신기술의 사용 매뉴얼 작성을 위해 농산물 종류에 따른 최적화된 미생물 검출법을 확립하였고, 현장 적용을 위한 매뉴얼을 작성하였다.

The demand for rapid detection of foodborne pathogen in fresh vegetables is growing. Pathogen detection is required to prevent the outbreak of fatal food poisoning which can potentially take lives of newborns, elders and patients with weakened immune systems. Prevention of outbreak can benefit from a screening system that can be easily used on-site by untrained personnel. Developing such a screening system would allow regular and frequent screening of fresh vegetables before they are distributed to local stores, and can prevent contaminated food from being cooked and ingested by potential victims of pathogen outbreak. This project’s goal is to develop a sensor that can simultaneously detect five foodborne pathogens from fresh vegetables. The sensor uses paper-based microfluidics and is encased in a reusable plastic cartridge, which enables easy operation of highly sensitive detection that require complex and multiple detection steps. We demonstrate simultaneous detection of Escherichia coli O157:H7, Salmonella typhimurium, Listeria monocytogenes, Staphylococcus aureus and Bacillus cereus and show the developed sensor’s application as a screening platform for on-site foodborne pathogen detection.
Novel and specific bioprobes are an essential part in biosensors for detection of pathogenic bacteria. One of most well-known bioprobe is antibody. Considering the amounts of efforts to develop “good” antibody, it is surprising how difficult it is to come by the right one for your purpose. In this study, we used crude surface protein extract to immunize and to develop monoclonal antibodies specific for the antigen. We constructed 2 MAb cell lines for each of 5 foodborne pathogens. Among them, MAbs for E. coli O157 or Staphylcoccus aureus appeared to be particularly useful for detection of the pathogen. We applied them to the immunomagnetic separation technique and it showed high recovery of the specific pathogen among the mixed bacterial culture. In addition, when it was tested on lateral flow assay, it was able to detect as low as 104 CFU/reaction of the pathogen.
Although antibodies are the most commonly used biorecognition elements, they are suffered from cross-reactivity and high production cost. Recently, bacteriophages have gained increasing interest as alternatives to antibodies due to the host specificity and low-cost. However, lysis-mediated phage infection process remains a burden for the reliable detection methods. Here, we isolated several Bacillus cereus and Staphylococcus aureus phages from various environmental samples and analyzed their genomes. Then we produced engineered cell wall binding domains (CBDs) from the isolated phage endolysins in E. coli and tested their feasibility as new probes to detect multiple pathogens. The CBDs provide high specificity and binding capacity to target pathogens. Fluorescent microscopy revealed that the two different pathogens (B. cereus, S. aureus) were clearly identified by its colors and the presence of non-target bacteria did not interfere with the target-specific binding property of each CBD. The magnetic nanoclusters (MNCs) coated with CBD can be used to separate and concentrate the target bacteria from liquid sample. The CBD-coated MNCs achieved 70 to 90% capture of target bacteria with less than 10% non-specific recovery. This CBD-based magnetic assay is useful for pre-analytical sample processing, especially for separating target bacteria from complex food matrices. In addition, CBDs-coated MNCs can be easily incorporated to other rapid bio-sensing platforms and thus offer a great potential in developing CBD-based bio-sensors.
In addition to the development of pathogen-specific bioprobes and multiple paper-based sensor, we optimized the application conditions for on-site multiple paper-based sensor and produced a manual for the application.
