본 과제에서는 지질대사 이상에 의해 유발되는 대사질환인 비알콜성 지방간, 인슐린저항증, 비만, 당뇨 등 대사질환의 예방과 치료가 기대되는 의약용 천연 소재를 개발하기 위해서 총 9,700종의 해외식물 추출물 은행으로부터 민속학적 정보와 화학적 정보에 근거하여 선발된 자원에 대한 in vitro 지방생합성 효소(hDGAT1&2,hGPAT1)와 세포 내 지방 생합성 저해 활성 관련 1차, 2차 탐색과 문헌, 특허정보를 조사하여 5종(FBM037-071, FBM054-100, FBM036-015, FBN089-063, FBM010-099
본 과제에서는 지질대사 이상에 의해 유발되는 대사질환인 비알콜성 지방간, 인슐린저항증, 비만, 당뇨 등 대사질환의 예방과 치료가 기대되는 의약용 천연 소재를 개발하기 위해서 총 9,700종의 해외식물 추출물 은행으로부터 민속학적 정보와 화학적 정보에 근거하여 선발된 자원에 대한 in vitro 지방생합성 효소(hDGAT1&2,hGPAT1)와 세포 내 지방 생합성 저해 활성 관련 1차, 2차 탐색과 문헌, 특허정보를 조사하여 5종(FBM037-071, FBM054-100, FBM036-015, FBN089-063, FBM010-099)의 유효자원을 최종적으로 선정하였음.
이들에 대한 지질대사 관련 in vitro 효소 저해 활성과 선택성, 세포 내 지방 생합성 저해, 지방산으로 유도된 세포 내 지질축적 억제, 지질대사 관련 유전자 발현 , in vivo 기능성 평가 (OLTT, OGTT)와 안전성 평가(경구급성독성)를 통하여 전 임상으로 진입 가능한 후보소재 1 종을 발굴하였음.
후보소재인 FBM010-99 추출물은 DGAT와 pancreatic lipase의 활성을 효과적으로 억제하고, HepG2와 십이지장선암세포(HuTu 80)내의 중성지방 생합성을 저해하며, acute oral lipid tolerance test에서 장관에서 중성지방의 흡수를 효과적으로 억제하여 중성지방의 혈중강하 효과를 나타내므로, 비만, 인슐린저항성, 이상지질혈증, 간지방증(hepatic steatosis) 및 NAFLD 등의 대사성 질환의 예방 및 치료의 유용한 의약용 소재로 활용이 가능함.

About 9.700 species of foreign plants, which were selected on the basis of ethno-pharmacological and chemical information from foreign plant library, were screened by in vitro enzyme assays and finally 5 candidates(FBM037-071, FBM054-100,FBM036-015, FBN089-063, FBM010-099)were chosen through solvent fractionation, in vitro enzyme assay, cell-based assay such as cellular TG formation, FFA-induced lipid accumulation, glucose uptake and adipocyte differentiation.
Among them, FBM010-099 was selected as one of pre-clinical candidate based on in vitro, in vivo evaluation, FBM010-099 inhibited DGAT and pancreatic lipase activity. As a result, the IC50 values for DGAT1 and DGAT2 enzymatic activities were 9.0 μ g/ml and 5.0 μg/ml, respectively; however, it showed moderate inhibition activity in pancreatic lipase. In addition, it decreased intracellular biosynthesis in HepG2 and HuTu80 cells. Further oral administration of FBM010-099 significantly reduced postprandial TAG in mice following an acute oral lipid challenge model. Taken together, the current study indicates that it alleviated postprandial hyperlipidemia by inhibiting DGAT activity and therefore may be used as effective lipid-lowering therapeutic agents.
