생식발생독성시험은 생식주기 전반에 미치는 영향을 평가해야 하므로 동물대체시험법 개발에 한계가 있으며, 아직 연구 개발은 초기 단계에 있다. 따라서 본 연구에서는 수컷 생식독성을 신속하게 검색할수 있는 동물대체시험법을 마련하기 위해 수컷 생식줄기세포를 이용한 in vitro 검색시험법을 개발 확립하고자 하였다.

수컷 생식줄기세포의 배양은 장기적으로 안정적인 배양을 할 수 있도록 feeder-free system 배양기술의 적용을 통해 배양 시스템을 확립하였고, Oct4, Tex18, Piwil2, Vasa 등 특이적 유전자 발현을 통하여 수컷 생식줄기세포의 특성화를 확인하였다. MTT assay로 ICsub>50값을 확인하여 최고의 농도를 선정하였으며, 수컷 생식독성 평가지표로 OECD TG 489의 in vivo mammalian comet assay를 적용하여 대조물질(양성 3종, 음성 1종) 및 시험물질 12종에 관하여 독성평가를 하였다.
수컷 생식줄기세포를 이용한 comet assay 시험법의 실험실간 전수 가능성 확인을 위해 양성대조물질 1종 및 시험물질 3종에 대하여 comet assay를 수행한 결과, 실험실간 결과가 일치하는 것으로 나타났다.
대조물질(양성 1종, 음성 1종) 및 시험물질 1종에 대하여 수컷 마우스 정자를 이용하여 in vivo comet assay를 한 결과, 양성대조물질 및 시험물질은 정자의 DNA 손상을 일으키는 것으로 나타났으며, 음성대조물질은 정자의 DNA에 영향을 미치지 않는 것으로 나타났다. 수컷 생식줄기세포를 이용한 comet assay 시험법의 타당성을 확인하기 위하여 in vitro comet assay와 in vivo comet assay 간의 상관성 분석 결과, 양성대조물질에서는 Tail DNA%, Tail length 및 Olive Tail Moment의 R2값이 각각 0.95, 0.95, 0.85, 0.91로 나타났으며, 시험물질에서는 Tail DNA%, Tail length 및 Olive Tail Moment의 R2값이 각각 0.96, 0.96, 0.90, 0.91로 나타나 높은 상관관계를 나타냈다.
더 나아가서, 수컷 생식줄기세포의 세포사 기전 확인을 위해 ROS 생성량을 측정하여 DNA 손상과 ROS 생성량 간의 상관성이 있는 것을 확인하였다. 또한 TUNEL assay, Annexin V 및 PI 확인을 통하여 in vitro comet의 독성 경향과 비슷한 것으로 나타났다. 이러한 결과들을 바탕으로 수컷 생식줄기세포의 독성영향은 apoptosis에 기인한 것으로 판단되며, 수컷 생식줄기세포를 이용한 comet assay가 수컷 생식독성을 예측하는데 좋은 대체시험법이 될 수 있다고 사료된다.

Development of alternative test methods is still in its early stage as the effect on the entire reproductive cycle should be assessed in reproductive and developmental toxicity tests. Therefore, this study aims to establish the development of an in vitro screening test method using male germline stem cells to develop a new alternative test method, which can detect male reproductive toxicity rapidly.
We established the cell culture system with the feeder-free system culture method in order to stably culture male germline stem cells. And then, we identified the characterization of male germline stem cells through specific gene expression such as Oct 4, Tex 18, Piwil 2, and Vasa.We calculated IC50 values with the MTT assay to select the highest concentrations, and evaluated toxicity of male germline stem cells using control substances (3 positive toxic substances and 1 negative toxic substance) and 12 test substances, based on OECD TG 489 (In vivo mammalian comet assay).
We evaluated between-laboratory transferability using 1 positive substance and 3 test substances with the in vitro comet assay. As a result, we found out that the results obtained from the 2 laboratories were consistent.
We also performed the in vivo comet assay with male mouse sperms using 1 positive substance,1 negative substance and 1 test substance. The results show that the positive substance and the test substance damaged sperm DNA but the negative substance did not have any impact on it. To demonstrate the validity of the comet assay using male germline stem cells, we analyzed the correlation between in vivo and in vitro comet assays (Head DNA%, Tail DNA%, Tail Length and Olive Tail Moment). As a result of the analysis, we identified a strong correlation between those 2 assays as R2 = 0.95, 0.95, 0.85, 0.91 for the positive substances and R2 = 0.96, 0.96, 0.90, 0.91 for the test substances.
Furthermore, we confirmed the apoptosis mechanism of male germline stem cells to measure ROS production, and identified the correlation between DNA damage and ROS production. We also identified the correlation between in vitro comet toxicity and apoptosis analysis using TUNEL assay, Annexin V and PI staining. Based on those results, toxicity of male germline stem cells was caused by apoptosis. The comet assay using male germline stem cells could be a candidate for alternative test methods to predict male reproductive toxicity.
