2. 골형성 평가지표 및 동물모델 개발: 인공뼈 개발을 위한 편평골 골결손 동물모델을 개발하였고, external fixator와 kirschnwe wire를 이용하여 장골 골결손 동물 모델도 확립함. 또한 conventional X-ray, micro-CT, 병리 조직검사와 같은 뼈 형성과정 평가 방법도 확립하였다.
3. 중간엽줄기세포의 골모세포로의 분화: 1)골수유래 중간엽줄기세포의 증식 및 분화지표 확립, 2)유래조직에 따른 줄기세포의 분화도 측정, 3)중간엽줄기세포 분화의 신호전달계 분석, 4)성체줄기세포주 확립, 5)백서 성체줄기세포의 확립
4. 3차원지지체에서 중간엽줄기세포의 생장 및 분화: 나노파이버 전기방사법이나 염출법으로 제작된 실크 3차원 지지체의 구조 및 세포 생장을 관찰하였고, 2차원 실크박막, 실크 3차 원지지체, 실크분말에서 줄기세포의 생장과 분화를 유도하였다.
5. 줄기세포 접종 실크 3차원지지체의 in vivo 골형성능: 확립한 장골 골결손 모델을 이용하여 줄기세포가 포함된 실크 3차원지지체를 백서동물의 골결손 부위에 이식한 뒤 x-ray, micro-CT, 조직병리 검사를 통해 골형성능을 평가함.
6. 2차원 실크박막의 병원성프리온 제거 효과: 실크박막의 표면에 항 프리온항체를 처리하여 병원성프리온의 제거를 시도하였으나 프리온 제거효과가 미미한 것으로 관찰됨.
7. 실크 단백의 gel-sol transition과 고형화 기술 개발: 1) 젤 형태 실크단백의 고형화 실험 및 결합 유도를 위한 첨가제 스크리닝 실험, 2) PMMA 고분자 원료와 실크 피브로인 복합재료에 대한 물성 파악, 3) PMMA/silk 인공시멘트의 미세구조 측정, 4) 생체적합성 동물모델 시험을 위한 PMMA/silk 인공시멘트 칩의 제조, 5) 실크 피브로인 복합재료에 대한 물성 파악, 6) 인공시멘트 제조 국제규격 시험방법의 도입, 7) 실크 단백과 PMMA 복합재료의 수술 적합성을 위한 실험동물모델 제작
8. 실크 단백 방수성 박막 제작: 개발된 실크 피브로인 인공뇌막의 생체적합성 평가와 실험 동물모델을 이용한 방수성 평가를 함.
9. 인공시멘트 및 인공뇌경막 시술 후 염증제어를 위한 단백질 치료법 개발 연구: 염증제어 효능이 있는 생체 내 단백질(AMPK 효소)과 세포내 침투를 용이하게 하는 펩타이드(PEP-1PTD)를 융합한 형태의 재조합 단백질을 제조함.
10. 실크 피브로인 자체의 뼈 생성 가능성 탐색: 여러 종류의 실크 피브로인을 스크린닝 하던 중 실크 자체만으로 뼈 생성 분화를 촉진하는 실크 피브로인을 찾음. 정확한 분석을 위해 ALP staining과 Von Kossa staining 수행하여 실크 피브로인 자체의 뼈 생성가능성을 탐색함.
11. 뼈 생성을 촉진하는 침투성 단백질 MSX2 보조제 개발: 뼈생성 기능이 있는 MSX2 효소를 Tat PTD와 재조합 융합단백질로 제조함.
12. 쥐 모델을 이용한 실크 인공 고막 패치의 안정성, 생체적합성 입증: 실크 인공 고막 패치의 생체적합성 및 고막 천공 패치로써의 적합성을 확인하기 위해 동물 실험을 수행한 결과, 실크 패치의 고막천공 치료 시간의 단축 효과 및 고막 재생의 질적 차이가 있는 것을 확인. 실크 인공 고막 패치의 투명성 및 세포친화성, 우수한 생체적합성을 통해 손상된 고막을 효율적으로 재생시킬 수 있음.
13. 실크 생체막의 생분해 현상 관찰: 실크 패치의 생분해도를 관찰하기 위해 rat 피하에 삽입 후에 기간별로 19개월까지 생분해 현상을 SEM, TEM 및 조직학적 염색을 통해 확인함. 실크의 뛰어난 생체적합성과 느린 분해속도, 나노 입자 형태로 분해되어 주변조직으로 흡수되는 특징을 이용하여 조직 공학에서 실크를 적절하게 이용할 수 있는 지식을 제공할 수 있을 것이라 생각됨.
14. 실크 나노 섬유막의 생분해 속도를 조절, 이를 이용한 생분해성 지지체 개발: 결정화 방법이 다른 3가지 type 실크 나노섬유막을 실험동물의 피하에 삽입하여 기간별로 생분해성 및 생체적합성을 SEM 관찰 및 조직학적 염색을 통해 확인함.
15. 실크 피브로인을 이용한 인공 진피 대체물 개발: 본 연구실에서 제작한 10-50 마이크로미터 공극을 가진 실크 나노섬유막을 동물 모델에 이식하여 생체적합성 및 창상 치유 효과를 실험한 결과, 염증 반응은 초기를 제외하고는 거의 나타나지 않는 것으로 확인되었고, 창상 치유 효과도 기존 2차원 나노섬유막에 비해 훨씬 뛰어나고 분해 현상도 빠르게 진행되는 것이 관찰됨.
16. 흡수성 실크 뼈 고정판의 생체적합성 확립: 실크 피브로인/박테리아 셀룰로오스 복합 플레이트가 뼈의 재생 촉진과 재생 시간 단축에 효과가 있다는 것을 확인함.
17. 실크 인공 근막 제조 기술 개발: 1마이크로 미만의 공극을 가진 실크 나노섬유막을 제작하여 human condrocyte 배양 시 세포 접착 및 증식이 잘 일어나는 것을 확인함.
18. 실크단백질을 조골세포 및 치주인대세포에 주고 유전자 발현 분석: Microarray 결과 세포의 종류에 따라서 발현이 증감되는 유전자들이 다소 다르게 나타남. 특히 조골세포에서 alkaline phosphatase (ALP)와 교원섬유의 합성이 증가되는 것은 뼈의 형성에 유리하게 작용할 수 있을 것으로 사료됨. BMP-2의 경우 특이하게도 조골세포에서는 실크 단백질에 의하여 그발현이 영향이 없었으나 치주 인대 세포에서는 발현이 오히려 감소하는 것을 볼 수 있음. 실크 단백질에 대한 조골세포의 반응은 좀더 깊이 있게 연구할 필요가 있어서 real-time RT-PCR 및 ELISA, alkaline phosphatase assay로 분석을 진행함.
19. 실크 단백질을 다양한 세포에 주고 세포 증식에 미치는 영향 분석: 조골양 세포와 치주인대 세포에서 물에 쉽게 용해되는 형태의 분말이 용해도가 떨어지는 분말에 비하여 세포 증식을 억제하는 능력이 더 높은 것으로 나타남. 또한 실크 단백질을 가하는 경우 세포 부착을 촉진하는 단백질인 fibronectin과 염증을 매개하고 혈관 재생에 관여하는 것으로 알려진 TGF-β1의 발현이 증가됨.
20. 실크단백질과 수산화 인회석을 혼합하는 경우 물질 안정성 분석: 실크 단백질과 수산화 인화석을 혼합한 이식재를 가토의 두정골 결손부 모델에 적용한 결과, 이식재가 다수 잔존하고 이를 흡수하기 위하여 다핵거대세포가 출현하며, 실크 단백질 사이로 신생골의 형성은 거의 일어나지 않는 것을 알 수 있음.
21. 동물모형에서 실크단백질 PRF 혼합체와 GBR 개발: 실크 단백질 PRF 복합체의 신생골 형성 능력과 치과용 임플란트 주위 골 결손부 회복 능력을 확인함.
22. 실크 단백질의 분자량 차이에 따른 세포 반응 분석: 실크단백질의 분자량의 차이에 따른 세포 반응의 차이 분석한 결과 분자량이 높은 실크 분말 (30kDa)의 경우 분자량이 낮은 실크분말 (<1kDa)에 비하여 세포 증식은 더 많이 시키지만 microarray 분석 상에서 염증을 매개하는 유전자의 발현이 분자량이 낮은 실크 분말에 비하여 더 많이 나타나는 것으로 관찰됨.
23. 실크 단백질과 삼인산 칼슘 복합체에 의한 인공뼈 개발: 실크단백질과 삼인산 칼슘 복합체를 이용하여 가토의 두개골 결손 부위에서 골 형성 능력을 분석한 결과 실크 단백질의 제조 방법에 따라서 생체 내 분해 속도나 골 재생 능력에서 많은 차이를 보임.
24. 실크 단백질을 이용한 골 유도 재생막의 물성 및 골 유도 능력 평가: 실크단백질을 이용하여 제조한 골 유도 재생막은 제조 방법에 따라 물에 대한 용해도가 차이가 남. 제조한 골 유도 재생막의 골 재생 능력을 평가한 결과 대조군에 비하여 양질의 골이 통계적으로 유의할 만큼 실크 박막을 이식한 군에서 많이 형성됨.
25. 실크 단백질과 전분 복합체에 의한 골 재생 능력 평가: silk fibroin/nano hydroxyapatite(nHA)/corn starch 복합체가 nHA/corn starch 복합체에 비하여 통계적으로 유의한 양의 골형성 증대를 야기하지 못함을 확인함.
26. 다공성 실크 단백질 소재의 골지지체와 substance P 복합체에 의한 골 재생 능력 평가: 실크 단백질 지지체와 substance P를 혼합하여 줄기세포를 이식부위에 유도하여도 신생골의 재생이 대조군에 비해 우수하지 않았고 실크 지지체의 생분해성만 빨라짐.
27. 실크 소재 인공뼈 재료의 첨가물로서 Resorcinolic lipid의 효능 분석: 4-HR은 조골양 세포에서 alkaline phosphatase activity와 osteocalcin의 발현을 모두 증가시킴. 4-HR을 함유한 실크 박막의 골 재생 효과가 대조군에 비하여 탁월함. 4-HR은 NF-kB pathway를 억제함. 4-HR을 함유한 실크 소재의 치과용 인공뼈의 골 재생 효과가 대조군에 비하여 우수함.

Health-related industry, especially a market of biomaterials grows rapidly. As longevity of human increases, demand for bone has been increased, too. Despite this global increase of bone or artificial bone demand, supply of bone tissue which is either derived from cadaver or is made from artificial
Health-related industry, especially a market of biomaterials grows rapidly. As longevity of human increases, demand for bone has been increased, too. Despite this global increase of bone or artificial bone demand, supply of bone tissue which is either derived from cadaver or is made from artificial biomaterials is limited. Therefore, demand for artificial bone is expected to grow rapidly in the near future.
Silk is a natural product derived from silkworm and has been used as surgical suture materials for a long time. The biggest advantage of silk fibroin over other biomaterials is biocompatibility. Recently, effort has been exerted on developing new silk fibroin-derived biomaterials in order to apply them in tendon, bone tissue or teeth bone with diverse different physical properties. By combining cutting edge biotechnology such as stem cells technology with silk fibroin-derived biomaterials, they expect to develop artificial bone made of silk.
Currently, most popular biomaterials applied in human prosthesis are polylactide, polyglycolide, and polycaprolactone. Collagen and chitinate are also used. On the top of biocompatibility, silk as a biomaterial has another advantage which is cheap and can be produced from silkworms in large quantities. Moreover, consuming silk fibroin as a biomaterial will contribute to economy of agricultural industry.
To develop artificial bone from silk fibroin, we have produced a variety of silk fibroin-derived biomaterials with different physical properties and applied them on disease models in vitro and in vivo.
Our ultimate goal is to develop artificial bone from silk fibroin derived biomaterials. While pursuing this, we were supposed to control diverse physical properties of silk fibroin. Such by-products of silk fibroin were applied on a number of disease models.
Specific aims of our projects are as follows.
1. Evalutation of biological safety based on ISO10993.
2. Generation of critical-sized “long” bone defect model in rats.
3. Three-dimensional culture of mesenchymal stem cells (MSC) in silk fibroin scaffold.
4. Differentiation of cultured MSC in silk fibroin scaffold into osteoblasts.
5. Artificial bone cement (or filler) for compressive fracture of spine.
6. Absorbable silk fibroin plate for fixation of facial bone fracture.
7. Artificial tympanic membrane and dermis derived from silk fibroin.
8. Artificial dura mater using silk fibroin membrane.
9. Biodegradable silk fibroin plate
10. Silk fibroin membrane for guided-bone healing after tooth extraction
We have accomplished the following results:
1. To establish procedures for toxicity testing on the developed silk materials: Among the biological toxicity testings based on ISO10993, we have established the following tests ①the implantation test ② cytotoxicity test, ③Ames test (genotoxic test), ④ in vivo micronuclei test (genotoxic test), ⑤ in vitro micronuclei test (genotoxic test), ⑥ chromosomal aberration test (genotoxic test). All these tests were performed on the materials used in this study and we confirmed that the all the implants developed from the silk fibroin did not show toxicity.
2. To establish animals model and evaluation criteria for bone formation: We set up the critical sized bone defect rat models not only for flat bone but also for long bone. For a long bone defect model, we used a external fixator and kirschnwe wires. We also established the evaluation criteria for bone formation using conventional X-ray, micro-CT, and histology.
3. Differentiation of mesenchymal stem cell to osteoblast cells: We established a method to quantitatively test proliferation and differentiation status of bone marrow derived mesenchymal stem cells and tested the differentiation capacity of stem cell based on the origin of the stem cells. We analyzed the signal transduction pathway involved in the differentiation of the mesenchymal stem cells and established the adult stem cell lines from rat.
4. Proliferation and differentiation of mesenchymal stem cells on the three dimensional scaffold. We observed proliferation of cells on the three dimensional silk materials. We were able to induce the differentiation of stem cells growing on the two dimensional silk membrane, three dimensional silk materials and silk powders.
5. In vivo bone formation with stem cell-loaded three dimensional silk materials: Three dimensional silk materials injected with stem cells were transplanted to the fractured area of the rat which was induced to have a long bone deficit. From these animals, weevaluated bone formation by X-ray and micro-CT, and biopsy.
6. Removal of pathological prion by two dimensional silk membrane: We tested the silk membrane for the removal of the pathological prions and found that it is not effective.
7. Establishment of methods for gel-sol transition and solidification of silk fibroin powder.
8. Generation of water-repellent membrane with silk protein: We tested for the biological compatibility and water repellency of artificial dura mater developed with silk fibroin.
9. Studies on the protein therapy for controling inflammation after surgery of artificial cement or artificial dura mater: We generated recombinant fusion protein of anti-inflammatory protein, AMPK, and a peptide (PEP-1 PTD) for improving cell permeability.
10. Testing the ability of silk fibroin for bone formation: We found that the silk fibroin can stimulate the differentiation of stem cells into osteoblast which was documented by ALP staining and von Kossa staining.
11. Generation of Tat-MsX2 for stimulating the bone formation : We generated recombinant fusion protein of an enzyme MSX2 which can stimulate bone formation and a peptide (Tat) for improving cell permeability.
12. Test of toxicity and biocompatibility of artificial silk eardrum: Animal model was used to test for toxicity and biocompatibility of artificial silk eardrum and we found that the artificial silk eardrum was not toxic and more biocompatible.
13. Biodegradation of silk membrane: After we implanted silk fibroin patch in subcutaneous tissues of rat for 19 months and observed using electron microscopy and immunohistochemistry, we concluded that silk fibroin was biocompatible because it was absorbed in adjacent tissues without any necrosis or malignant transformation.
14. Different methods of crystalization of silk fibroin ended up with difference of degradability in tissues.
15. Silk fibroin patch with 10-50 micrometer pore size made for artificial dermal tissue was excellent in terms of wound healing, inflammatory reaction and degradability.
16. A composite biomaterial of silk fibroin/bacterial cellulose has osteoconduction effect, which could reduce bone regeneration time.
17. Artificial myofascia : Fibrous nano membrane of silk for fascia was excellent in cell adhesion and proliferation.
18. Microarray analysis of gene expression profile of osteoblast cells or dental ligament cells after treatment of silk fibroin: Alkaline phosphatase gene and collagen genes were upregulated, however, BMP-2 was downregulated.
19. Water soluble fraction of silk fibroin powder has inhibitory effect on cell proliferation. Silk fibroin increased expression of fibronectin and TGF-β1.
20. A composite biomaterial of silk fibroin/hydroxyapaptite inhibit new bone formation.
21. Silk fibroin enriched with PRF was more effective in bone formation in critical-sized skull defect model. We think that silk fibroin enriched with PRF may be used as guided-bone regeneration.
22. Cell proliferative potential of silk fibroin varies depending on molecular weight of silk fibroin. Silk fibroin proteins of higher MW (about 30 kDa) induces more cellproliferation compared to those of lower MW (<1 kDa), however. induces expression of genes related with inflammation.
23. A composite material of silk fibroin/tricalcium phosphate showed different biodegradability and new bone forming capacity depending on manufacturingmethods.
24. Depending of crystalization methods, silk fibroin membrane showed variable water solubility. We found that applying silk fibroin membrane on the top of critical-sized skull defect enhanced new bone formation.
25. Comparing between two composite biomaterials (silk fibroin/nano hydroxyapatite(nHA)/corn starch and nHA/corn starch), we did not found statistically significant difference in osteogenesis.
26. Substance P attached to silk fibroin sponge did not enhance new bone formation in critical-sized bone defect model. Instead, degradation was enchanced after attaching substance P.
27. Membrane made of a composite biomaterial of silk fibroin/4-hexylrecorcinol (4-HR) has outstanding bone regenerative potential compared to controls.
Many results of this project can be applied to clinic (clinical trial) as well as other research fields. Among our accomplishments from this project, artificial tympanic membrane made of silk fibroin has got permission for clinical use from Korean Food & Drug Administration and undergone clinical trial. Artificial bone filler cement with silk fibroin for compressive fracture and artificial silk fibroin dura mater were patented. We also expect developing silk fibroin biomaterial further will be beneficial for agricultural industry.
In conclusion, we have pursued modifying silk fibroin as a biomaterial and applying on diverse disease models to generate artificial bone or other medical prosthesis. We accomplished a number of clinically applicable products made of silk. We expect that silk fibroin could be a good biomaterial applicable for artificial bone and other medical prosthesis.
