IV. 연구개발결과
O 모델 : 천연물의 처방, 약재, 성분, 효능 정보를 포함하는 전통 천연물 공개 데이 터베이스 및 표적 단백질 정보를 수집하고, 이를 기반으로 통합 데이터베이스를 구축하였다. 가상 인체 시스템을 위한 인체 모델링 정형언어를 개발하여 조직 특이적,조직 간 유전자 조절 관계, 단백질 상호 작용 관계, 신호 전달 경로 정보, 대사 반응
관계, 다수준간의 관계를 통합한 공통 (generic) 가상 세포 모델을 구축하였다.
O 소재 : 전통 천연물 특화 통합형 초고속 성분/생리활성 분석 기술 (1HTac) 개발하였다. 초고속 분획화 시스템 (Sep-Box) 을 활용하여 소청룡탕, HT071 처방의 분획물을 도출하였다. 소청룡탕, 1HTac 시스템을 활용하여 도출된 HT071 유효/지표성분 분리 및 표준화를 하였다. 그리고 한의학효능, 원물, 추출물, 식물정보, 채집과 같은 정보를 담고 있는 천연물 정보 라이브러리를 구축하였다.
O 표적 : 인간 탈인산화 효소군 확보 및 전통천연물 스크리닝으로 전통천연물의 분자표적 규명 및 다중천연물 (MC) 에 의한 다중표적 (MT) 조절효과를 규명하였다. 유효화합물의 프로브를 사용하여 FITGE 방법을 통해 표적 단백질을 규명하였다.

O 마커 : 비만동물모델에서 얻은 멀티오믹스데이터 분석을 통한 신규 바이오마커를 발굴하였다. 태음조위탕과 그 구성성분 분석 및 지표성분의 항비만/당뇨 in vivo 효능을 검증하였다. 그리고 단백체 분석을 통한 태음조위탕과 그 구성성분 및 지표성분의 항비만/당뇨 작용기전 규명 및 진단/치료용 마커를 발굴하였다.
O 인체 : 멀티오믹스 기술을 통합한 식품기반 MC/MT 전통천연물 고유의 항상성 도전 모델 및 고위험군 확인 인체시험기술을 개발하였다. 임상시험에서 발생하는 임상화학, 전사체, 대사체 등 빅데이터 수집을 위한 통합 storage를 개발하였다. 그리고 국제공동연구를 위한 MOU 체결 및 네트워크를 구축하였다 (네덜란드 TNO, NIZO).

In the coming era, our society needs to be prepared for the challenges posed by aging population and prevalence of infectious diseases caused by globalization. Big pharmas will keep generating novel drugs for various modern diseases including cancers and obesity. However, a large fraction of such dr
In the coming era, our society needs to be prepared for the challenges posed by aging population and prevalence of infectious diseases caused by globalization. Big pharmas will keep generating novel drugs for various modern diseases including cancers and obesity. However, a large fraction of such drugs may cause unexpected side-effects as individuals have distinct genetic make-ups and lifestyles despite those drugs go through a rigorous assessment by authorities (e.g. , FDA). Also, many of the big pharmas are running out of strong candidates of drug leads, which impose healthcare concerns on many nations. In this context, compounds found in traditional oriental medicine that have been clinically used for centuries deserve special attention for systematic development, and are important assets of ours. Their strong efficacies to treat various symptoms have been empirically validated although many of their mechanisms of action still remain to be elucidated. Also, one important feature of traditional oriental medicine is multicomponent-multitarget (MC/MT) principle where multiple compounds get together to show synergistic combinations.
The ultimate objective of Bio-Synergy Research Center is to modernize traditional oriental medicine using state-of-the-art techniques involving modeling, computations, multi-target molecular imaging, multi-omics, molecular and chemical biology and medical sciences for better understanding on the mechanism of action, and to generate new sources for novel drugs. This immense task of the Center will lead Korea as a dominant player not only in a natural product market, but also in markets of various drugs and relevant biotechnology fields. The potential is far beyond our lmagmation.
In order to realize the abovementioned challenges, five major groups within the Bio-Synergy Research Center termed as Modeling, Material, Targeting, Marker and Clinical trial have focused on following research areas:
○ Modeling human biosystem modeling, generic in silico human system development, construction of an efficient platform to process information on materials, targets and clinical trials , data mining analyzing correlations between performance and efficacies, organism - specific ontologies, epigenomic regulation analysis , 3D in silico human body visualization.
○ Material standardization of natural products found in traditional oriental medicine, construction of a natural product library and identification of compounds suitable for MC/MT.
○ Targeting activity of natural products found in traditional oriental medicine, multiple natural probe synthesis and construction of a drug- human molecule network.
○ Marker construction of a clinical database based on multi -omics data, identification of disease/toxicity biomarkers and relevant compounds in traditional oriental medicine, development of diagnosis kits, analysis ofMC/MT principle and multi-omics (i.e ., metabolomics and proteomics) to analyze efficacies of multiple compounds in traditional oriental medicine.
○ Clinical trial safety tests of compounds found in traditional oriental medicine , development of big data integration storage, and development of individual health status index, and studies on clinical effects of compounds found in traditional oriental medicine.
Studies conducted by the five groups come into a synergy by complementing one another. The Modeling group basically supports all the studies pursued by the other four groups by providing information platforms. Upon this support, the Material group identifies bioactive compounds in a high-throughput manner, and their targets are analyzed by the Target group. The Marker group then looks into mechanisms of action associated with the bioactive compounds. Finally, the Clinical trial group examines clinical effects of the candidate compounds from traditional oriental medicine. The five groups have produced some notable outcomes during last 3 years as accurately documented in this evaluation report.
