1. 연구개발 결과
가. 프로바이오틱스 효능균주의 선발 및 발효제품 개발 (제1연구분야)
○ 전통발효식품 유래 미생물의 기능성 균주 선발
- 젓갈(22종), 청국장(13종), 김치(30종)에서 SAM, GABA, Folic acid 생성하는 균주를 623종을 분리하였음
- SAM 생성량이 높은 균주는 젓갈유래 10종, 청국장유래 7종, 김치유래 8종이 선발되었음.

1. 연구개발 결과
가. 프로바이오틱스 효능균주의 선발 및 발효제품 개발 (제1연구분야)
○ 전통발효식품 유래 미생물의 기능성 균주 선발
- 젓갈(22종), 청국장(13종), 김치(30종)에서 SAM, GABA, Folic acid 생성하는 균주를 623종을 분리하였음
- SAM 생성량이 높은 균주는 젓갈유래 10종, 청국장유래 7종, 김치유래 8종이 선발되었음.
- GABA 생성량이 높은 균주는 김치유래 8종이 선발되었음.
- Folic acid 생성량이 높은 균주는 젓갈유래 2종, 청국장유래 4종이 선발되었음.
- 선발된 38종의 균주 중 Lactobacilli가 12종, Bacilli가 18종, Enterococci가 4종 및 Leuconostoc이 5종이었음.
○ 우수균주의 특성조사
- SAM 생성량이 높은 균주는 대부분 약산성의 pH에 대한 내성을 가지고, 모두 담즙내성이 있었으며, 5종 병원균 중 4종 이상 억제하는 균주는 6종이었음.
- 선발된 8종 중 GABA 생성량이 높은 2종의 균주 특성은 34℃가 최적온도이고, 내산성, 내담즙성, 항생제 내성 및 효소활성이 있는 반면 항균력은 비교적 낮은 편이었음.
- folic acid 생산량이 많은 균주는 대부분 37℃가 최적온도이고, 내산성, 내담즙성이 있었고, 특히 L. plantarum JA71 균주는 항균력, 항생제 내성 및 효소활성이 높았음.
○ 기억력 증진물질 생성 미생물을 이용한 발효조건 확립 및 제품개발
- SAM 생성량이 1.41mM을 생산하는 Enterococcus durans J6-9 균주, 1% MSG 첨가한 MRS배지에서 GABA 생성량이 386.37μg/mL를 생산하는 Lactobacillus plantarum K255 균주, Folic acid 생성량이 4.04μg/mL를 생산하는Lactobacillus plantarum JA71 균주를 각각 선발하였음.
- 두유 및 쌀음료 생산에 적합한 최적배양조건 설정하기 위하여 RSM(반응표면분석법)으로 실시한 결과는 다음과 같음.
· SAM 생성 발효두유 : 두유농도 7.37%에 Enterococcus durans J6-9 균주 1% 첨가 후 40℃에서 14.73시간 배양했을 때, pH 4.9, SAM 함량은 1.68mM이었음.
· SAM 생성 발효쌀음료 : 쌀음료농도 10%에 Enterococcus durans J6-9 균주 1% 첨가 후 30℃에서 15 시간 배양했을 때, pH 4.81, SAM 함량은 1.3mM이었음.
· GABA 생성 발효두유 : 두유농도 8%, 녹두 1%, MSG농도 0.56%, Yeast Extract 농도 0.85%에 Lactobacillus plantarum K255 균주 1% 첨가 후 40.2℃에서 36 시간 배양했을 때, GABA 함량은 322.06 μg/m, 기호도 점수 5.72이었음.
· GABA 생성 발효쌀음료 : 쌀음료농도 10%, MSG농도 0.42%, Yeast Extract농도 0.7%에 Lactobacillus plantarum K255 균주 1% 첨가 후 33℃에서 28 시간 배양했을 때, GABA 함량은 184.92 μg/mL, 기호도 점수 6.13이었음.
· Folic acid 생성 발효두유 : 두유 고형분 함량 8%에 Lactobacillus plantarum JA71 균주 1% 첨가 후 37℃에서 16 시간 배양했을 때, Folic acid 함량은 11.83 μg/mL, 기호도 점수 6.39 이었음.
○ 제품의 기억력증진 효능 구명
- 기억력 증진효능 여부를 판단하기 위해 4주 동안 각 군당 10마리의 Sprague-Dawley(5주령, 250g) 수컷을 실험대상으로 300mL/성인 체중 60kg(=1mL/200g rat), 1일 기준으로 배양물을 투여하였음.
- Scopolamine hydrobromide 처리를 하지 않은 그룹의 대조군(Con)은 아무것도 넣지 않고 살균만 한 두유, 처리군 A는 GABA 생성균을 이용한 발효두유, 처리군 B는 SAM 생성균을 이용한 발효두유, 처리군 C는 Folic acid 생성균을 이용한 발효두유, 처리군 D는 A, B, C를 1:1:1의 비율로 섞은 발효두유를 포함한 실험식이를 급여하여 수미로 찾기를 실시 하였음.
- Scopolamine hydrobromide를 처리하지 않은 실험군과 처리한 실험군 모두에서 A, B, C, D 군이 control에 비해 수(水)미로를 빠져나오는 데 적은 시간이 걸리는 유의적 차이를 보였고, 더 적은 시행오차를 보였음.
- 기억력 증진효능이 끝난 렛트의 혈중지질(TG, HDL cholesterol, LDL cholesterol)과 간기능 수치(GPT, GOT) 분석 결과 control과 처리군 별 간의 유의적 차이가 없었으며, 간, 신장, 비장, 고환, 흉선, 폐의 무게를 비교한 결과 역시 control과 처리군 별 간의 유의적 차이는 없었음.
○ 산업화
- SAM 생성 발효두유 : 백태를 세척하고 6 시간 이상 불린다. 불려진 백태의 껍질을 분리하여 버리고 100∼120℃에서 20 분간 가열함. 삶아진 백태를 갈아 착즙하여 두유원액을 만들고 여기에 생수를 첨가해 고형분 함량을 8%로 맞춤. 그 후 90℃에서 5 분간 살균한 뒤 40℃ 정도로 냉각함. 여기에 Enterococcus durans J6-9 균을 1.0%(v/v)로 접종하고 40℃에서 15 시간 배양함. 배양이 끝난 발효액(전체의 78%)에 딸기쨈(45%)14%, 올리고당 6.0%, 액상과당 2.0%을 첨가하고 혼합하여 발효두유를 제조하였음.
- GABA 생성 발효두유 : 두유농도 8%, 녹두 1%, MSG농도 0.56%, Yeast Extract 농도 0.85%에 Lactobacillus plantarum K255 균을 1.0%(v/v)로 접종하고 40℃에서 36 시간 배양한다. 배양이 끝난 발효액(전체의 83%)에 딸기쨈 15%, 올리고당 1%, 액상과당 1%을 첨가하고 혼합하여 발효두유를 제조하였음.
- Folic acid 생성 발효두유 : 대두 고형분 함량이 8%인 두유를 90℃에서 30 분간 살균하여 37℃로 냉각시킨 후 Lactobacillus plantarum JA71 균주를 1%(v/v) 접종하여 37℃에서 16시간 배양하였음. 발효가 완료된 두유 발효액(전체의 86%)에 올리고당 5%, 딸기잼 8%, 정제수 1% 를 첨가하고 교반하여 발효두유를 제조하였고, 오미자 발효두유는 두유 발효액의 냄새와 맛을 보완하기 위해 발효가 완료된 두유 발효액(전체의 86%)과 탈지분유 4%를 혼합한 후에 올리고당 5%, 오미자청 8%, 정제수 3%를 첨가하고 교반하여 오미자 발효두유를 제조하였음. 오미자 발효두유에 첨가한 오미자청은 오미자와 설탕을 1:1 비율로 혼합하여 3개월간 발효시킨 후 발효오미자를 100 mesh 체로 여과하여 제조하였음.
나. 발효식품용 종균 및 표준 발효공정 개발 (제2연구분야)
○ 전통발효식품 전체 균총 및 메타지놈 분석
- 장류(된장 11종, 고추장11종, 청국장11종, 연변된장 9종, 중국된장7종), 전통주(누룩 30종, 막걸리7종), 젓갈(멸치 5종, 멍게 5종, 새우 6종)의 microbial community 분석하여 전통 발효식품 토착 미생물 지도 및 전통발효식품 토착 미생물 D/B를 구축함.
○ 기능성 발효식품 제조용 유용 발효미생물의 분리, 동정, 기능성 평가 및 유용 유전자원 확보
- 식품연 유전자은행으로부터 분양받은 1,000여종 균주를 재생하여 활성화가 가능한 683종 균주의 16S rDNA 염기서열을 분석하여 미동정 및 동정오류 균주에 대한 정보를 update 하였으며, 이 과정 중 3종, 5균주의 신종 미생물을 확보하였음.
- 확보한 683종 균주에 대한 효소적(amylase, protease), 기능적(항당뇨, 간기능개선, 항알러지) 특성 분석하는 1, 2, 3차 screening 통해 최종적으로 16종의 기능성 발효 종균을 확보하였으며, 이들 균주에 대한 database 구축 및 특허 출원을 완료함.
- 전통발효식품미생물의 genomic DNA sequencing 분석 결과를 바탕으로 gene-based screening을 통해 DNJ, GABA 및 SAM 생합성 유용 유전자군을 함유한 균주(B. amyloliquefaciens 26, 140N ,Lactobacillus brevis 877G, Lactobacillus paraplantarum 72815)를 확보함. 이들 균주에 대한 특성 및 기능성을 평가하여 발효식품 제조에 적용하였으며, 이에 대한 특허 출원을 완료함.
○ 기능성 발효식품이 장내 미생물에 미치는 영향 분석
- 정상마우스에서는 기능성 발효식품식이에 따른 장내 미생물 균총 변화가 관찰되지 않았으나, 질환모델 마우스에서는 기능성 발효식품 식이에 따른 장내 미생물 균총 변화가 관찰됨. 특히, 항당뇨활성이 있는 Bacillus amyloliquefaciens 26 및 140N 균주를 이용하여 제조한 콩발효 식품을 식이한 당뇨마우스에서는 당뇨증상이 완화되었을 뿐만 아니라, 장내에 butyrate를 생산하는 미생물과 Ruminococcus 및 Lactobacillus속 미생물이 증가됨을 확인함.
○ 기능성 종균의 산업적 활용을 위한 표준 발효공정 개발
- 종균을 발효식품제조에 산업적으로 활용하기 위해서는 안전성이 확보되어야 함으로 최종 선발 균주들에 대한 genome 분석 및 gene annotation 을 수행함. 분석결과 이들 균주에는 병원성 유전자가 deletion 되어 있음을 확인함으로써 선발된 균주에 대한 산업적 활용 기반을 마련함.
- 발효식품 유래 유용 균주의 산업적 활용이 용이하도록 균주의 유전적, 효소학적, 기능적 특성을 바탕으로 database를 구축하였음.
- 선별된 기능성 균주 16종 중 발효식품제조 가장 적합한 균주인 26, 140, 369 를 종균화하고 산업적으로 활용하기 위해 casein 등의 부형제를 이용한 균주 제형화 기술을 확립함.
- 선별된 종균인 Bacillus amyloliquefaciens 26(항당뇨), 249(항알러지, 항당뇨), 369(간기능, 항당뇨) 균주를 이용하여 청국장을 제조하는 표준공정을 개발하고 이를 통해 시제품을 제조함.
다. 전통발효식품 미생물 및 생체 대사체 분석 (제3연구분야)
○ 선발 미생물 및 이를 이용한 발효식품의 대사물질 분석
- 전통발효식품 유래 유용미생물 100균주(세균, 효모, 곰팡이) 대사체 분석
- 유용미생물 이용 막걸리, 장류의 대사물질 분석 및 대사체 프로파일링 구축
○ 막걸리의 대사체 모니터링에 의한 발효지표 설정
- 효모 99-7과 누룩 152-1을 사용한 막걸리의 지표물질로 발효기간에 따라 증가하는 아미노산 phenylalanine, arginine, 1-(4-aminobutyl)guanidine을 선정하였고, 막걸리의 과일향과 꽃 향 특성을 나타내는 isoamyl alcohol과 phenethyl alcohol을 지표물질로 선정하고 발효 8일 최대함량을 보여 본 막걸리의 발효기간을 8일로 설정함.
○ 된장의 기능성 대사체 탐색 및 발효지표 설정
- 시판메주로 제조한 된장과 양산된장의 주요 지표물질은 L-arginine, 4-pyridoxic acid, L-serine, isoglutamine, cytosine, D-threonic acid 성분들로 이들 성분이 시판되는 메주와 된장의 특징적인 성분으로 확인됨.
- Bacillus amyloliquefaciens subsp. plantarum FZB42(T) 균주를 접종한 된장(26)의 발효중기(30일) 기능성 증가는 m/z value 233.09[M+H]+ (C8H15N3O5) 물질과 261.14[M+H]+ (C9H19N5O4) 물질 이외, ornithine, 1-deoxynojirimycin, genistin 과 같은 물질의 증가로 확인되어, 이들 물질을 발효된장의 기능성 향상을 위한 지표물질로 선정함.
○ metabolomics 기술 기반 발효식품이 생체 대사에 미치는 영향 분석
- 발효식품이 생체 대사체에 미치는 영향을 알아보기 위하여 각 정상군과 비만군의 동물모델에서 발효식품을 식이로 급여한 동물의 간조직 대사체를 분석한 결과, 정상군은 각 식이별 간조직 대사체에 큰 변화가 없는 것으로 나타났으나, 비만군의 간 대사체는 급여한 식이에 의해 큰 영향을 받는 것으로 나타남.
- Bacillus amyloliquefaciens (M26, M140)을 이용한 콩발효식품을 급여한 동물모델은 간 대사체 프로파일의 변화를 시판 당뇨개선제제 식이 동물모델과 유사하게 보였으며, 이들 균주를 이용한 청국장 식이는 당뇨개선 효과가 있는 것으로 확인됨.
2. 활용에 대한 건의
가. 프로바이오틱스 효능균주의 선발 및 발효제품 개발 (제1연구분야)
○ 본 연구에서 분리된 SAM, GABA, Folic acid 생산균은 그동안 기능성 있는 외국의 수입종균에 의존하던 국내 발효음료 시장에서 경쟁력을 가질 것이며 동시에 수입대체효과가 있을 것으로 예상됨. 또한 본 연구결과는 두유 및 쌀음료업체에 기술이전을 통하여 실용화할 계획임.
○ 선발된 균을 상업화하기 위해서는 동결건조된 분말로 제조하여야 하며, 동결건조 후에도 활력이 유지되어야 실제 활용이 가능함. 따라서 일련의 단계를 거쳐 만족스러운 결과가 나올 수 있도록 해야 할 것이며, 만족스러운 결과가 나온다면 관련 업체와의 협조를 통하여 제품생산이 될 수 있도록 할 것임
나. 발효식품용 종균 및 표준 발효공정 개발 (제2연구분야)
○ 본 연구 결과는 전통발효미생물과 발효식품의 안전성 및 기능성 측면에서의 우수성을 입증할 수 있는 과학적인 근거를 제시한 것으로, 연구결과를 바탕으로 구축한 전통발효식품 토착 미생물 D/B는 향후 수행될 관련 연구과제의 결과와 연계하여 지속적으로 관리하고 update할 필요가 있음. 이를 통해 구축한 전통발효식품 토착 미생물 지도 및 D/B는 전통식품산업 활성화 및 미생물자원 이용 활성화에 기여할 것임.
○ 전통발효미생물과 장내미생물 균총과의 상관성 규명을 통해 발효식품 식이가 한국인 microbiome에 미치는 영향 규명 연구 및 한국인의 유전 및 생리적 특성에 맞는 probiotics 개발 연구의 기반 자료로 활용 가능함.
○ 발효식품 제조에 적용이 가능한 기능성 우수종균은 포화상태의 발효식품시장 활성화를 위한 기능성 장류제품 개발에 활용 가능하며, 기술이전을 통해 영세업체 중심인 장류산업의 경쟁력 확보에 활용 가능함.
다. 전통발효식품 미생물 및 생체 대사체 분석 (제3연구분야)
○ 전통발효식품 유래 유용미생물을 이용한 콩 발효식품이 동물모델에서 간 대사체에 많은 영향을 주는 것을 알 수 있었고 발효식품에 의한 당뇨개선 효과 규명을 위한 기초자료로 활용될 수 있을 것으로 판단됨.

Ⅳ. Result of R&D
1. Selection of probiotics and development of fermened products(Research section 1)
○ Selection of probiotics isolated from traditional fermented food
- 623 strains producing SAM, GABA and Folic acid were isolated from Jeotgal (22 kinds), Cheonggukjang (13 kinds) and Kimchi
Ⅳ. Result of R&D
1. Selection of probiotics and development of fermened products(Research section 1)
○ Selection of probiotics isolated from traditional fermented food
- 623 strains producing SAM, GABA and Folic acid were isolated from Jeotgal (22 kinds), Cheonggukjang (13 kinds) and Kimchi (30 kinds).
- 10 strains isolated from salted seafood, 7 strains isolated from Cheonggukjang and 8 strains isolated from kimchi were selected for the strains producing high concentration of SAM.
- 8 strains isolated from kimchi were selected for the strains producing high concentrations of GABA.
- 2 strains isolated from salted seafood and 4 strains isolated from Cheonggukjang were selected for the strains producing high concentrations of folic acid.
- In 38 kinds of selected strains, 12 strains identified as Lactobacilli; 18 strains identified as Bacilli; 4 strains identified as Enterococci; 5 strains identified as Leuconostoc.
○ Characterization of the selected bacteria
- Almost of the strains producing high concentrations of SAM had tolerance to slightly acid. All of them had bile tolerance and 6 strains showed the antimicrobial activity against more than 4 kinds of pathogens in 5 kinds of pathogens.
- In 8 kinds of selected strains, 2 strains producing high GABA concentrations were investigated their physiological characteristics. The optimal growth temperature was 34℃. While they showed pH tolerance, bile tolerance, enzyme activity and antibiotic tolerance, antibacterial activity was relatively low.
- The optimal growth temperature of the strains producing high concentrations of folic acid was almost 37℃. The strains showed pH tolerance, bile tolerance.
Especially, L. plantarum JA71 showed high value of antibacterial activity, enzyme activity and antibiotic tolerance.
○ Establishment of optimum cultivation condition and development of product using the bacteria producing memory enhancement substances
- Enterococcus durans J6-9 producing 1.41mM of SAM, Lactobacillus plantarum K255 producing 386.37 μg/mL of GABA in an MRS broth containing 1% MSG and Lactobacillus plantarum JA71 producing 4.04 μg/mL of folic acid were respectively selected.
- The result of RSM (response surface methodology) for establishing of optimum cultivation condition for producing soy milk and rice drink
· Fermented soy milk producing SAM: 1% of Enterococcus durans J6-9 strain was inoculated into soy milk (solid content=7.37%); then the soy milk was incubated for 14.73 h at 40℃. After incubation, the concentrations of SAM were 4.68mM and the pH was 4.6.
· Fermented rice drink producing SAM: 1% of Enterococcus durans J6-9 strain was inoculated into rice drink (solid content=10%); then the rice drink was incubated for 15 h at 30℃. After incubation, the concentrations of SAM were 1.3mM and the pH was 4.81.
· Fermented soy milk producing GABA: 1% of Lactobacillus plantarum K255 strain was inoculated into 8% soy milk, 1% mung bean, 0.56% MSG and 0.85% yeast extract; then the soy milk was incubated for 36 h at 40.2℃. After incubation, the concentrations of GABA were 322.06 μg/mL and the preference score was 5.72.
· Fermented rice drink producing GABA: 1% of Lactobacillus plantarum K255 strain was inoculated into 10% rice drink, 0.42% MSG and 0.7% yeast extract; then the rice drink was incubated for 28 h at 33℃. After incubation, the concentrations of GABA were 184.92 μg/mL and the preference score was 6.13.
· Fermented soy milk producing folic acid: 1% of Lactobacillus plantarum JA71 strain was inoculated into soy milk (solid content=8%); then the soy milk was incubated for 16 h at 37℃. After incubation, the concentrations of folic acid were 11.83 μ g/mL and the preference score was 6.39.
○ Proof the memory enhancement effect of the product
- To determine the memory enhancement effect, fifty Sprague-Dawley male rats (5 weeks of age; body weight=250g) were arranged into 5 group. They were fed the culture (300mL/ 60kg body weight/day = 1mL/200g rat weight/day)
- Group A was fed with fermented soymilk of Lactobacillus plantarum K255 inoculated, group B was fed with fermented soymilk of Enterococcus durans J6-9 inoculated, group C was fed with fermented soymilk of Lactobacillus plantarum JA71 inoculated, group D was fed with mixure of A, B and C fermented soymilk on the same ratio and control was fed with plain soymilk(90℃, 5mins heating). All the groups fed each soymilk for 4 weeks and then divided 2 groups each. One was orally administered scopolamine hydrobromide and the other was not.
- All the groups which were treated scopolamine hydrobromide and were not, The measuring time to escape water maze of group A, B, C, D were shorter than control.
And the number of hit the water maze wall were fewer on group A, B, C and D than control.
- There was no signigicant differences in TG, HDL, LDL, GOT, GPT, body weight and organ weight between control and fermented soymilk fed groups.
○ Industrialization
- Fermented soy milk producing SAM: Soybeans were washed and soaked for 6 h in enough water; then the peeled soybeans were boiled about 20 minutes at 100 to 120℃. The boiled soybeans were grinded and filtered to extract juice. After water was added for making 8% of solid content, it was sterilized at 90℃ for 5 min. 1% of Enterococcus durans J6-9 strain was inoculated into soy milk; then the soy milk was incubated for 15 h at 40℃. After incubation, 14% strawberry jam, 6% oligosaccharides and 2% high fructose corn syrup were added and mixed to finished fermented soymilk.
- Fermented soy milk producing GABA: 1% of Lactobacillus plantarum K255 strain was inoculated into soy milk (solid content=8%); then the soy milk was incubated for 36 h at 40℃. After incubation, 15% strawberry jam, 1% oligosaccharides and 1% high fructose corn syrup were added and mixed to finished fermented soymilk.
- Fermented soy milk producing Folic acid: Soy milk(solid content=8%) were sterilized for 30 min at 90℃ and cooled to 37℃. After cooling, 1% of Lactobacillus plantarum JA71 strain was inoculated into the soy milk and incubated for 16 h at 37℃. 5% oligosaccharides, 8% strawberry jam and 1% purified water were added to finished fermented soy milk and mixed; 4% skim milk, 5% oligosaccharides, 8% schisandra chinensis baillon, 3% purified water were added to finished fermented soy milk and mixed for improving the flavor.
2. Technology development of starter for industrial use and standardized fermentation process (Research section 2)
○ Analysis of total microbial flora and metagenome in traditional fermented foods
-Microbial communities in Soybean pastes(11 kinds of Doenjang, 11 kinds of Kochujang, 11 kinds of Cheonggukjang, 9 kinds of Yeonbeun Doenjang, 7 kinds of Chinse Doenjang), Traditional wine(30 kinds of Nuruk, 7 kinds of Makgeolli), Jeotgal(5 kinds of sardines, 5 kinds of sea squirts, 6 kinds of shrimp) were analyzed to found map and D/B of native microorganism in traditional fermented foods.
○Separation, identification, functionality evaluation of beneficial fermentation microorganism for production of functional fermented foods and securement of beneficial gene resources
-1,000 species of cultures obtained from gene bank of KFRI were revitalized. 16S rDNA base sequence of each of 683 species was analyzed and information about unidentified or falsely identified cultures was updated. During this process, 5 new cultures of 3 species were secured.
-Through the 1st to the 3rd screenings, where enzymatic (amylase, protease) and functional (antidiabete, liver function improvement, antiallergy) properties of 683 secured cultures were analyzed, 16 functional fermentation cultures were finally secured. Database of these cultures was established and patent application was submitted.
-Through gene-based screening based on the analysis result of genomic DNA sequencing of microorganism in traditional fermented foods, cultures containing beneficial DNJ, GABA and SAM biosynthetic gene clusters (B. amyloliquefaciens 26, 140N ,Lactobacillus brevis 877G, Lactobacillus paraplantarum 72815) were secured. Properties and functionality of the cultures were evaluated and applied to production of fermented foods. Patent application was submitted.
○ Analysis of effects of functional fermented foods on intestinal microorganism
-Microbial flora changes in Intestinal of the functional fermented foods diet was not observed in normal mice, however microbial flora changes in Intestinal of the functional fermented foods diet was observed in disease model mice. In particular, soybean fermented food prepared using the anti-diabetic active strains of Bacillus amyloliquefaciens 26 and 140N diet group relieved symptoms of diabetes in diabetic mice, as well as butyrate producing microbes and Ruminococcus and Lactobacillus strains were increased.
○Development of standard fermentation process for industrial utilization of functional starters
- Safety of starter must be ensured in order to use fermented foods production, final candidate strains carried out genome analysis and gene annotation. Genetic analysis of pathogenicity of these strains had been confirmed that all selected starter strains did not have any pathogenic gene.
- In order to use fermented food derived useful strain in food industries, genetic, enzymatic and functional characters of strains were data based.
- Strain #26, #140, #369 out of 16 kinds of previously selected candidate strains were selected for fermented food production and formulation technology was established using additives like casein and oligo-saccharides.
- Develop a standard process for manufacturing Cheonggukjang using selected starter 26 (anti-diabetic), 249 (anti-allergic), 369 (liver function) and prototypes were manufactured according to standard process.
3. Analysis of metabolites in microorganism from traditional fermented foods and animal (Research section 3)
○ Metabolite analysis of selected microorganism and fermented food
- Metabolite analysis of useful 100 strains (bacteria, yeast, and mold) derived from traditional fermented food
- Metabolite analysis and profiling of makgeolli and soybean food using useful microorganism
○ The fermentation biomarker setting up by the metabolite monitoring of makgeolli - Biomarker of makgeolli using yeast 99-7 and nuruk 152-1 was based on measures of the amino acid such as phenylalanine, arginine and 1-(4-aminobutyl)guanidine increased depending on the fermentation period. Isoamyl alcohol and phenethyl alcohol showing the fruit and flower aroma characteristic of the makgeolli was selected as biomarker. The fermentation showed the maximum content on 8 days and the fermentation period of the makgeolli was set to 8 days.
○ Functional metabolite research and fermentation biomarker set up of deonjang - Major biomarker of commercial small producted doenjang vs commercial mass producted doenjang were L-arginine, 4-pyridoxic acid, L-serine, isoglutamine, cytosine and D-threonic acid. These metabolite, which were positively related to the commercial meju and deonjang.
- The funtionality increase of deonjang(26) fermented by Bacillus amyloliquefaciens subsp. plantarum FZB42(T) on fermentation 30 days were related to the m/z value 233.09[M+H]+ (C8H15N3O5), 261.14[M+H]+ (C9H19N5O4), ornithine, 1-deoxynojirimycin, and genistin. These metabolite were selected as biomarker for the funtionality improvement of fermented deonjang
○ Research on the effect of fermented food into biological metabolism through metabolomic technology
- We investigated the soy diet fermented by Bacillus amyloliquefaciens, which produce high content of 1-deoxynojirimycin and have strong α-glucosidase inhibitory activity, showed remarkable changes on liver metabolites compared with normal and commercial soy diets in obese mice group on PLS-DA model.
- This study highlights the insight to the liver metabolites dependence according to different soy diet of control and obesity mice group. The metabolic profiles of mice liver were concerned in each soy diet, especially the soy diets fermented by B. amyloliquefaciens (M26, M140) strongly changed liver metabolic profiles in obesity mice group.
