본 연구팀은 한국인 호발암에서 에피지네틱 바이오마커 발굴 및 표적 치료제 개발을 위하여 총 4개의 연구 주제에 대해서 다음과 같은 연구를 진행하였음.
1. 한국 호발암의 하나인 폐암 세포주에서 G단백질 신호전달계가 암세포의 에피제네틱 변형에 미치는 영향과 그 기전을 구명하기 위하여 실험을 진행한 결과, G단백질 신호전달계가 DNA methyl transferse와 hisotone deacetylase의 발현을 조절한다는 것을 밝혀냈다. Gas 단백질은 DNA methyl transferase 중 DNMT-1과 DNMT-
본 연구팀은 한국인 호발암에서 에피지네틱 바이오마커 발굴 및 표적 치료제 개발을 위하여 총 4개의 연구 주제에 대해서 다음과 같은 연구를 진행하였음.
1. 한국 호발암의 하나인 폐암 세포주에서 G단백질 신호전달계가 암세포의 에피제네틱 변형에 미치는 영향과 그 기전을 구명하기 위하여 실험을 진행한 결과, G단백질 신호전달계가 DNA methyl transferse와 hisotone deacetylase의 발현을 조절한다는 것을 밝혀냈다. Gas 단백질은 DNA methyl transferase 중 DNMT-1과 DNMT-3A의 발현을 감소시키며, histone deacethylase인 HDAC6와 8의 발현은 증가, SIRT6의 발현은 감소시켰으며, transcriptional cofactor 인 p300의 발현도 감소시켰다. 또한 Gai 단백질은 DNA methyl transferase인 DNMT-1과 histone deacethylase HDAC8 발현을 증가시킴을 확인하였다. G단백질 신호전달계가 이들 효소의 발현을 조절하는 기전을 분석한 결과, mRNA의 변화나 단백질이 degradation 되는 것을 조절함으로써 이루어지며, 이는 G단백질의 하위 신호인 PKA나 Epac을 거쳐 JNK 나 ERK, p38과 같은 MAPK 또는 Akt 가 관여함을 밝혔다. 이러한 효소의 발현 조절은 암세포의 성장과 이동, 사멸에 영향을 미친다는 사실을 확인하였다.
2. 전사인자(STAT3)에 의한 Histone Modification 기작 규명 및 암 epigenetic 관련 암 표적유전자의 발현을 규명함으로써, 고형암에서 발암기전으로써의 STAT3억제제 및 후보물질들에 의한 유전자 조절기술개발을 통하여 epigenetic에 의한 유전자발현를 조절할 수 있는 효과적인 물질발굴을 목표로 하였음. (1) STAT3에 의한 HIF-1 단백질 안전성에 관한 기작 규명 (2) 유방암에서 tamoxifen 내성기작과 유방암 및 교모세포종에서 STAT3 단백질을 target하는 inhibitor의 효과를 검증하고, 항암제로서의 가능성을 검토 (3) 간암에서의 STAT3 활성화와 간암의 암줄기세포 표지자 CD133 단백질의 발현 기작을 규명 (4) 뇌종양에서의 STAT3단백질을 직접적으로 표적화하는 신규합성물질(#10117)의 항암효과 검증. 그 결과 STAT3에 의한 HIF-1단백질 안전성에 관한 기작규명 하였으며 유방암 약물내성기작과 간암의 암줄기세포 표지자 CD133단백질의 발현 기작을 규명 하였고 STAT3억제 물질 효능 검증을 통하여 STAT3표적 단백질의 발현에 Histone 단백질의 epigenetic변화가 중요하다는 사실을 규명함. 따라서 STAT3단백질을 표적으로 한국인 호발암의 성장억제, 약물내성 극복, 암줄기세포 타겟 항암치료 전략이 새롭게 부가될 수 있다고 사료됨.
3. (1) 위암, 대장암, 폐암, 유방암, 간세포암의 다단계암화과정에서 프로모터 CpG섬 과메틸화와 반복 DNA시퀀스의 저메틸화가 어느 시점에서 일어나는 변화인지, (2) CpG섬 메틸화표현형(CIMP)이 위암이나 대장암에 존재하는지, (3) 대장암에서 CIMP가 가지는 예후적 의미를 밝히고,(4) 위암, 대장암, 폐암, 유방암에서 LINE-1메틸화레벨이 다단계 암화과정에서 어떻게 변화하는지를 밝히고자 하였고, (5) 종양의 LINE-1 저메틸화레벨이 가지는 예후적 의미를 위암에서 폐암에서 밝히고자 하였다. 간세포암이나 위암에 대한 새로운 DNA메틸화마커를 발굴하기 위하여 pharmacological unmasking array methodology를 도입하였고, 기존에 알려진 DNA메틸화마 커들의 메틸화레벨을 평가하기 위하여 MethyLight technology를 이용하였으며. LINE-1 메틸화 레벨은 파이로시퀀싱기법을 이용하였다. 위암, 대장암, 유방암, 폐암에서 LINE-1메틸화레벨이 다단계 암화과정에서 어떻게 변화하는지를 규명할 수 있었고, 암종의 LINE-1메틸화레벨이 환자의 예후와 관련이 있음을 위암과 폐암에서 밝힐 수 있었다. 또한 수술후 보조항암화학치료를 받은 대장암에서 CIMP가 가지는 예후적 의미는 대장암의 암병기에 따라 다를 수 있으며, CIMP상태는 stage IV대장암에서 유의한 예후변수로 작용함을 밝힐 수 있었다.
4. 암 에피지네틱스 표적 치료기술개발을 통하여 에피지네틱스에 의한 유전자발현 감소를 조절할수 있는 효과적인 기술을 개발하기 위하여, (1) DNA methyltransferase 표적치료 기술을 개발하고, (2) Histone deacetylase isotype 특이적 억제 기술을 개발하며, (3) DNA methylation 및 histone deacetylation의 효과적인 임상 모니터링법을 개발하고, (4) 세포독성 항암제와의 병합요법 개발을 목표로 하였음. 특히, (5) 에피지네틱 변이 유전자 및 광범위한 변이 유전자군(LRES)의 에피지네틱 상태 변이에 따른 항암제 약제 감수성 변화에 대한 연구, (6) 암세포 특이적 에피지네틱 상태 변이 유도에 의한 세포 사멸 메커니즘의 연구, (7) 에피지네틱 상태 변이 유도를 통한 항암제 내성 극복 방법에 대한 연구를 통하여, 다양한 에피지네틱 마커를 개발하고 그 효능을 검증하였음.
이러한 일련의 연구를 통하여, 본 연구팀은 한국인 호발암을 대상으로 하여 암 에피지네틱스 바이오마커 발굴 및 표적치료의 성공적인 개발을 위한 기반 지식을 획득하였음.

1. Lung cancer is one of the major cancer in Korea. We hypothesized that G protein signaling might affect epigenetic changes in cancer cells and investigate thed underlying mechanism. We found that G protein signaling pathway regulates the expression of DNA methyl transferses and hisotone deacetylases. Gas protein decreased DNA methyl transferase DNMT-1 and DNMT-3A, and increased histone deacethylase HDAC6 and HDAC8, and decreased SIRT6 expression. Also, Gas decreased transcriptional cofactor, p300 expression. Otherwise, Gai protein increased DNA methyl transferase DNMT-1 and histone deacethylase HDAC8. We also found that G protein signaling altered mRNA expression or protein degradation to regulate those molecules, and the changes in expression were regulated though JNK, ERK and p38 or Akt via G protein down stream PKA and Epac. Finally, changes in histone deacethylases expression by G proteins affected cancer cell growth, migration and apoptosis.
2. To determin roles of STAT3 and Histone Modification in the solid tumor progression and identification of STAT3 inhibitors via gene modulation to provide therapeutic information for solid tumors.
(1) To analyze STAT3/HIF-1a signaling network and identify novel STAT3 targets
(2) To evaluate the effect of STAT3 target inhibitors on anti-tumor activity specific for breast and GBM cancer and its possible application for anti-cancer drug development.
(3) To assess a role of Stat3 activation in HCC for CD133 expression
(4) To evaluate the effect of candidates targeted to STAT3 on anti-tumor activity specific for solid tumors.
We have reported that STAT3 binds to HIF-1α, thereby up-regulates HIF-1-mediated VEGF expression in hypoxic solid tumor cells, which was inhibited by CADPE, a potential STAT3 inhibitors. Therefore, our data reveaeled an increase STAT3 activation in hypoxia and STAT3 activation-dependent HIF-1a stability and STAT3 activation in Tamoxifen-resistant breast cancer cells. STAT3 inhibitor, #ODZ10117 induced cell death in solid tumors and STAT3 targeting drug using small molecules(#ODZ10117) to evaluate anti-cancer and anti-metastatic activities in-vivo tumor model.
3. We aimed to 1) elucidate DNA methylation changes, both promoter CpG island hypermethylation and repetitive DNA element hypomethylation, which occur along multistep carcinogenesis of gastric cancer, colorectal cancer, lung cancer, breast cancer, and hepatocellular carcinoma, 2) identify whether CpG island methylator phenotype (CIMP) exists in gastric cancer, lung cancer and colorectal cancer, 3) elucidate prognostic implications of CIMP in colorectal cancers, 4) determine the timing of LINE-1 hypomethylation during multistep carcinogenesis, and 5) elucidate whether tumoral LINE-1 hypomethylation carries prognostic implications. To develop novel DNA methylation markers for gastric cancer and liver cancer, we used pharmacological unmasking array methodology. For the extensive candidate DNA methylation marker study, we used the MethyLight technology. LINE-1 methylation levels were analyzed in formalin-fixed tissue samples using pyrosequencing assay. We charaterized curves of methylation changes of LINE-1 during multistep carcinogenesis of gastric cancer, colorectal cancer, breast cancer and lung cancer. And tumoral LINE-1 hypomethylation was found to be an independent prognostic paramter heralding poor survival. And CIMP status was found to be closely associated with survival in adjuvant FOLFOX-treated patients with stage IV colorectal cancer.
4. In order to develop an effective cancer epigenetics targeted therapy, which can control aberrant epigenetic modification leading to transcriptional repression during tumorigenesis, the following aims were established; 1) Development of DNA methyltransferase targeted therapy, 2) Development of histone deacetylase isotype-specific suppression technology, 3) Development of effective clinical monitoring system of DNA methylation & histone deacetylation, 4) Development of combination therapy with cytotoxic anti- cancer drugs. Especially, through 1) Research on changes in anti-cancer drug sensitivity by epigenetically modified genes & long range epigenetic modification, 2) Research on cell apoptosis mechanism through induction of cancer cell specific epigenetic modification, 3) Research on anti-cancer drug resistance-overcoming method through induction of epigenetic modification, various epigenetic markers were found and the effects were validated. Through above researches, our team acquired fundamental knowledge for successful development of cancer epigenetics targeted therapy in major cancer types among Koreans
