연구의 목적 및 내용
종양조직의 미세환경은 다양한 종류의 비종양성 세포들로 조성되며, 정상세포가 생존하기에 불리한 저산소증, 포도당부족 및 산화적 스트레스 상태 등이 특징적임. 본 연구팀은 종양세포는 독특한 종류의 신호전달경로 및 microRNAs 발현을 조절함으로써 정상세포가 생존하기 어려운 미세환경하에서 증식하며 고도의 악성도를 획득한다는 가설을 검정하고자함. 사람 중피세포종 세포주와 종양조직을 이용하여 다양한 미세환경에 대응한 종양 세포의 생존 및 악성도획득과 관련된 생물학적 특성 및 분자생물학적 기작을 규명함으로써, 종
연구의 목적 및 내용
종양조직의 미세환경은 다양한 종류의 비종양성 세포들로 조성되며, 정상세포가 생존하기에 불리한 저산소증, 포도당부족 및 산화적 스트레스 상태 등이 특징적임. 본 연구팀은 종양세포는 독특한 종류의 신호전달경로 및 microRNAs 발현을 조절함으로써 정상세포가 생존하기 어려운 미세환경하에서 증식하며 고도의 악성도를 획득한다는 가설을 검정하고자함. 사람 중피세포종 세포주와 종양조직을 이용하여 다양한 미세환경에 대응한 종양 세포의 생존 및 악성도획득과 관련된 생물학적 특성 및 분자생물학적 기작을 규명함으로써, 종양에 대한 새로운 치료법 및 예후 판정 등에 활용 가능한 생체지표물질 후보를 발굴하기 위한 기초 자료를 확보하고자 함.
본 연구팀은 중피세포종 세포주를 저산소증, 포도당 부족, 산화적 스트레스의 다양한 미세 환경에 노출시켜 미세 환경에 따른 공격적인 표현형의 출현을 확인하였으며, 관련된 분자생물학적 기작과 신호 전달계를 규명하였음. 또한, microarray 기법을 이용하여 저산소증과 산화적 스트레스에 의해 차별적으로 발현하는 miRNAs를 발굴하여 적대적 미세 환경에서의 miRNAs 기능을 평가하였음.
연구결과
□ 저산소증으로 유도된 공격적인 세포 표현형과 관련 분자생물학적 기작 규명
- 중피세포종 세포주를 저산소 환경에 노출시키면 (1) 세포의 생존 증가, (2) 세포 사멸 감소, (3) 세포의 이동성 증가, 그리고 (4) 약물에 대한 저항성 증가 표현형이 나타남.
- 또한, 저산소증에 노출된 세포들은 (1) 저산소 유도 인자 (HIF1/2-α)의 활성과 미토콘드리아 Bcl-2 family 등의 신호 전달계 조절을 통해 적대적인 미세 환경에 적응하여 생존을 하였으며, (2) 상피간엽이행의 활성을 통해서 세포의 이동성 증가를 유발하였으며, (3) DNA 손상 감소에 따른 세포 사멸의 억제 및 세포 주기 조절을 통하여 항암제에 대한 저항성을 획득함.
□ 저산소증에 차별적인 발현을 보이는 miRNA 분석
- 중피세포종 세포주에 저산소를 노출하는 경우, 총 14개 (MS1)와 6개(H513)의 miRNAs가 차별적인 발현을 하였으며, 다양한 표적 유전자들의 발현을 조절함으로써 세포의 증식, 이동, 세포 사멸 조절 등 적대적 미세 환경에서 적응하는 것으로 평가됨.
□ 산화적 스트레스로 유도된 공격적인 세포 표현형 관련 분자생물학적 기작 규명
- 중피세포종 세포주에 산화적 스트레스를 가할 경우, (1) 세포의 생존 증가, (2) 세포사멸 감소, (3) 세포의 이동성 증가, 그리고 (4) 약물에 대한 저항성 증가 표현형이 나타남.
- 또한, 산화적 스트레스에 노출된 세포들은 (1) 저산소 유도 인자 (HIF1-α)의 활성과
(2) PGC1-α의 발현 억제를 통하여 적대적인 미세 환경에 적응하여 생존을 함.
□ 산화적 스트레스에 차별적인 발현을 보이는 miRNA 분석
- 중피세포종 세포주에 산화적 스트레스에 노출 하는 경우, 총 6개 (MS1)의 miRNAs가 차별적인 발현을 보였으며, 다양한 표적 유전자들의 발현을 조절함으로써 세포의 증식, 이동, 세포 사멸 조절 등 적대적 미세 환경에서 적응하는 것으로 평가됨.
□ 저포도당 및 포도당 결핍으로 유도된 약물 저항성 획득과 분자생물학적 기작 규명
- 포도당이 결여된 중피세포종 세포주는 Metformin에 대한 약물 저항성을 획득하며, 이는 Metformin에 의해 손상 받은 미토콘드리아의 자가 탐식 기전이 활성화되어 세포사멸이 감소되기 때문이며, 이와 함께 MDR1 단백질이 미토콘드리아 내 약물을 유출시키는 능을 하기 때문임.
연구결과의 활용계획
본 연구의 산물인 종양 미세 환경의 신호 전달계와 microRNAs의 발현 양상은 중피세포종의 진단과 예후 판정 인자로 활용될 계획이며, 궁극적으로 중피세포종의 공격성을 낮추며 약물 저항성을 극복할 수 있는 새로운 치료 전략을 수립하는데 기초 자료로 활용될 계획임. 또한, 본 연구에서 확보된 결과와 실험 모델을 활용하여 발암기전에서 다양한 미세환경의 영향력과 상호작용을 조절하는 microRNAs 양상 변화와 관련된 하부 신호전 달계 또는 물질을 동정하는 연구의 기초 자료로 활용될 계획이며, microRNAs외의 후성 유전학적 변형 특히 chromatin modification의 발암기전에서의 역할을 규명하는 연구에 이용될 계획임.

The proposed study tested a hypothesis that cancer cells proliferate and acquire malignancy in hostile microenvironment (hypoxia, glucose deficiency, and oxidative stress) through regulating distinct set of signaling molecules and microRNAs. The present study demonstrated that hypoxia, glucose deficiency, and oxidative stress promoted the acquisition of aggressive phenotypes in human malignant mesothelioma (HMM) cell lines, and a subset of microRNAs responding to hypoxia and oxidative stress were involved in the generation and maintenance of the malignant phenotypes in the hostile tumor microenvironments.
Result
□ Impact of hypoxia on aggressive phenotypes and underlying molecular mechanisms
HMM cells exposed to hypoxia showed enhanced cell clonogenicity, survival, migration, and resistance to cisplatin in vitro. The molecular mechanisms underlying the hypoxia-induced malignant progression were associated with activation of hypoxia inducible factors (HIF1/2-α), mitochondrial Bcl-2 family members, epithelial to mesenchymal transition, cell cycle arrest, and repair of DNA damage.
□ Differentially regulated microRNAs in HMM cells exposed to hypoxia
Under hypoxia, HMM cells showed a total of 14 (MS1 cells) and 6 (H513 cells) differentially expressed microRNAs. By using bioinformatic analysis, predicted target genes of the microRNAs were categorized into various biological processes, including response to hypoxia, cell motion, cell proliferation, and negative regulation of apoptosis.
□ Effects of ROS on aggressive phenotypes and related molecular mechanisms
HMM cells exposed to ROS-induced oxidative stress showed enhanced cell survival, migration, decreased apoptosis, and drug resistance in vitro.
Activation of hypoxia inducible factors (HIF1/2-α) and inhibition of PGC1-α expression were believed to be the molecular mechanisms responsible for the oxidative stress-induced malignant progression.

Under hypoxia, MS1 cells showed a total of 6 differentially expressed microRNAs using miRNA microaaray. By using bioinformatic analysis, predicted target genes of the microRNAs were categorized into various biological processes, including regulation of cell proliferation, migration, and apoptosis.
□ Effects of glucose deprivation on aggressive phenotypes and related molecular mechanisms
HMM cells cultured in glucose-deprived medium were resistant to Metformin. The reduction of cell apoptosis were resulted from activated autophagy engulfing the compromised mitochondria by Metformin and concordant up-regulation of MDR1 that inhibited accumulation of the drug within the mitochondria.
Expected Contribution
A distinct set of microRNAs and signaling molecules may be valuable to develop innovative diagnostic and prognostic tool for HMM patients and contribute to the development of an effective management of the malignant disease. Our data and experimental model could be a valuable tool to investigate interaction of microRNAs and key signaling pathways altered by hostile tumor microenvironments, which ultimately contribute to the elucidation of HMM carcinogensis.
