연구의 목적 및 내용
유해성 저분자 브롬 및 할로겐 화합물에 대하여 분자수준의 면역독성, 근골격계 장해 및 암발생 기작과 체내대사 과정을 이해하고 새로운 독성 타겟을 탐색하고자, 다음의 연구를 수행하고자 하였음.
1. 다양한 용도에 비하여 선행연구가 별로 없는 브롬 및 할로겐 화합물류의 면역독성, 근골격계 독성 및 암발생 과정에 미치는 영향을 세포 및 분자수준에서 최초로 평가하고자 하였음.
2. 분자수준의 독성기작을 규명하기 위하여 면역반응 관련 신호전달 체계에 대한 연구, 골대사 과정의 신호전달 과정에 미치는 영향
연구의 목적 및 내용
유해성 저분자 브롬 및 할로겐 화합물에 대하여 분자수준의 면역독성, 근골격계 장해 및 암발생 기작과 체내대사 과정을 이해하고 새로운 독성 타겟을 탐색하고자, 다음의 연구를 수행하고자 하였음.
1. 다양한 용도에 비하여 선행연구가 별로 없는 브롬 및 할로겐 화합물류의 면역독성, 근골격계 독성 및 암발생 과정에 미치는 영향을 세포 및 분자수준에서 최초로 평가하고자 하였음.
2. 분자수준의 독성기작을 규명하기 위하여 면역반응 관련 신호전달 체계에 대한 연구, 골대사 과정의 신호전달 과정에 미치는 영향 연구 및 체내 대사 및 대사체 규명연구를 수행하고자 하였음.
3. 독성물질이 작용하는 분자수준의 새로운 독성 타겟을 발굴하고 그 기능을 평가하여 독성에 민감한 생체 지표를 발굴함으로써 독성 연구에 필요한 기반을 조성하고자 하였음.
4. 이를 통하여 저분자량 브롬화합물의 면역독성, 근골격계 장해 및 암발생의 유발기작을 분자수준에서 규명함으로써, 국내 독성학 분야의 선도적인 발전을 이룩하고자 하였음.
연구결과
1. 저분자량 유해화합물의 근골격계 독성 평가: 조골세포의 분화를 촉진하는 Pin1의 영향을 확인하였고, Pin1에 의한 Runx2 및 osterix의 안정성 변화와 아울러 osterix 발현 조절자인 c-Cbl과 Cbl-b 등의 전사조절인자에 대한 역할을 발견하였고, 수 종의 브롬화합물이 특이적으로 특정 부분에 독성 영향을 미침을 발견하여 골격계도 중요한 독성타겟임을 밝혔음. 3T3-L1 전지방세포주의 지방세포 분화조건과 다양한 신호조절인자 중 YY1이 C/EBP beta에 결합하여 전사활성을 감소시킴을 최초로 발견하였으며, C2C12 근육모세포를 이용한 근육세포 분화모델을 정립하여 유해화학물질의 근골격계 및 지방세포 분화에 미치는 독성평가 시스템을 구축하였음.
2. 면역반응 관련 신호전달 체계에 대한 연구: TBBPA, bisphenol A 및 o,p’DDT 등의 유해 할로겐 화합물을 대상으로 알러지 관련 면역기능에 미치는 영향과 호흡기계 면역기능에 미치는 영향을 분자수준에서 규명하였으며, 특히 호흡기 면역반응과 관련하여 독성작용 기전을 MUC5AC 유전자 발현 및 관련 신호전달인자들을 대상으로 살펴 PKCδ에 의한 Akt, ERK1/2, JNK1/2 및 p38 MAPK의 인산화 과정을 밝힐 수 있었음.
3. 체내 대사계에 미치는 영향 연구: 발암물질 활성화와 관련된 CYP 1A1, 1B1 등에 대하여 estrogen 대사와 발암관련 유전자 발현간의 관계를 규명하고자, metformin을 대상으로 연구를 진행하였음. Metformin은 AhR 유전자 발현을 조절하여 발암물질대사효소인 CYP1A1과 1B1을 억제하며, AhR의 발현 억제는 전사조절인자인 Sp1 조절에 기인함을 최초로 발견하였음. 또한 metformin은 tryptophan 2,3-dioxygenase 조절을 통하여 CYP1A1, 1B1을 조절하는 사실을 규명하였음. 이 시험계는 각종 유해화학물질의 대사계 영향 평가 시에 활용이 가능함.
4. 체내 glutathione 포합체/핵산 adduct 규명 연구: LC-Mass 분석법을 활용하여 도출하며, 세포내 glutathione 포합체 및 DNA adduct 생성 가능성을 평가하였음. 특히 1-bromopropnae을 대상으로 in vivo 조건에서 guanine 및 adenine과의 adduct 생성을 각 장기별로 정량 분석하였으며, 각 장기별 adduct 생성특성을 비교하여 독성영향과의 상관성 연구를 위한 자료를 축적할 수 있었음. 또한, glutathione adduct 생성과 DNA adduct 생성 시 대사활성화 과정의 필요 여부를 평가하여 브롬화합물은 대사과정 없이도 포합체나 adduct 형성이 가능함을 최초로 밝혔음.
5. 종양줄기세포계 영향 연구: 장기간에 걸펴 유해화학물질에 저농도로 노출될 수 있는 작업장의 특성을 고려하여 비교적 낮은 농도에서 종양줄기세포주에 브롬화합물을 노출시킬 때 종양줄기세포의 특성이 증가되며, 이때 변화하는 각종 marker의 발현 특성을 확인하여 종양세포의 암화와 관련된 결과를 최초로 발견하였음.
6. 비만 및 지방간 발생 연구: 할로겐화합물인 TBBPA와 o,p’-DDT를 대상으로 체내 지방산 대사관련 신호전달 체계에 대한 영향 평가를 통하여 비만과 비알콜성 지방간에 대한 분자수준의 기전을 규명하여 독성타겟을 밝혔고, 이 시험계를 이용하여 유해화학물질의 지방대사 관련 독성 평가 시 분자기전의 규명이 가능해질 것으로 판단됨.
연구결과의 활용계획
1. 분자 수준의 면역독성 지표를 발굴하여 화학물질에 의한 면역독성 기작의 판단에 활용함.
2. 브롬화합물의 면역반응 관련 신호전달체계에 미치는 영향을 규명하므로써 단순한 독성 평가 수준의 기존의 연구수준을 선진국 이상의 수준으로 발전시킴.
3. 저분자 브롬화합물을 모델로 하여 독성물질의 체내 대사 및 대사체 규명, 골대사 항상성 이해를 위한 연구체계를 확립하고, 독성학 관련 전문 연구인력을 양성함.
4. 확립한 연구기법 및 연구결과를 전문학술지에 적극적으로 공개발표하여 화학물질 독성에 대한 국민의 이해를 진작하고 산업독성학의 발전에 기여함.
5. 결과적으로 독성연구를 위한 다학제 간의 공동연구 기반의 구축을 이룩함.

1. Effects of halogenated compounds on immune functions, bone metabolism and carcinogenesis were investigated for the first time in the level of cells and/or molecules.
2. Effects of halogenated compounds on signal transductions in immune cells and bone cells were investigated in association with their glutathione conjugation.
3. New molecular targets for toxic action were explored to screen the possibly sensitive toxic biomarkers for certain industrial chemicals like brominated compounds.
4. By understanding the toxic mechanism of low molecular weight halogenated compounds, the safety of certain industrial chemicals could be secured.
Result
1. Evaluation of muscular and bone toxicity induced by small mw compounds: Effects of Pin1, a differentiation factor for osteoblasts, on the stability of osterix and RUNX2 were studied, with relations to c-Cbl and Cbl-b. Studies with several bromocompounds revelaed that bone metabolism might be an important toxic target. In 3T3-L1 cells, the role of YY1 in C/EBP beta activity was investigated. Likewise, a toxicity evaluation system with C2C12 muscular cells were established.
2. Signal transduction study in immune response: Effects of TBBPA, bisphenol A and o,p’DDT on allergy-related or respiratory immune functions were investigated in molecular levels. Particularly, toxic mechanism on the expression of MUC5AC and its related signal transduction factors were unveiled, where the role of PKCδ on the phosphorylation of Akt, ERK1/2, JNK1/2 and p38 MAPK were solved.
3. Study on the drug-metabolizing systems: Role of metformin on the expression of carcinogen-activating CYP 1A1 and 1B1. Metformin regulated AhR gene expression through the regulation of transcription factor Sp1, by which the expression of CYP1A1 and 1B1 was suppressed. In addition, metformin regulated tryptophan 2,3-dioxygenase, by which the expression of CYP1A1 and 1B1 was controlled. The established system would be useful to investigate the effects of toxic chemicals in the future.
4. Glutathione conjugation and nucleic acid adduct formation by toxic chemicals: By using LC-Mass analysis, the formation of intracellular GSH conjugates and DNA adduct by 1-bromopropane was evaluated. Particularly, adduct formation of 1-bromopropnae with guanine and adenine was studied in vivo. In addition, the possible role of metabolic activation in the formation of glutathione conjugated and DNA adduct was studied for the first time.
5. Cancer stem cells: In the consideration of work place where the toxic compounds are exposed in low concentration for a long time, effects of low concentrations of certain bromocompounds on stemness of cancer stem cells were studied. In the study, the changes of many markers related with cancer cells were investigated.
6. Effects of xenobiotics on fatty liver: Effects of TBBPA and o,p’-DDT on fatty acid synthesis were studied with relations to the changes in signal transduction molecules. From the study, the molecular mechanism of toxicity could be elucidated. In addition, the established system would be used for understanding the effects of xenobiotics on obesity and fatty liver disease in the molecular level.
Expected Contribution
1. The molecular mekers studied for muscular, bone metabolism and immunotoxicity would be used in understanding the action mechanism of certain toxic compounds.
2. By unveiling the toxic mechanisms of halogen compounds in regulating immune response and bone metabolism, the level of toxicology studies would be upgraded.
3. The established study systems for drug metabolism, adduct analyses and target organ toxicity can be used further to evaluate and understand toxicity of certain toxicants. Human resources involved in the study are to be roled for the toxicology area in the future.
4. The results obtained were published in 15 SCI journals, which would be a concete sign of our improvement in the knowledge of industrial toxicology.
5. Conclusively, the multi-discipline study was successful in upgrading the toxicology area.
