연구의 목적 및 내용
가시광선에 의해 광경화시켜 키토산을 하이드로겔화시킴으로써 성장인자의 방출제어를 통해 창상부위를 효과적으로 치료할 수 있는 획기적인 스프레이형의 습윤드레싱제 개발

연구의 목적 및 내용
가시광선에 의해 광경화시켜 키토산을 하이드로겔화시킴으로써 성장인자의 방출제어를 통해 창상부위를 효과적으로 치료할 수 있는 획기적인 스프레이형의 습윤드레싱제 개발
○ 가시광선 광경화 가능 수용성 키토산 유도체 합성 및 재료 특성 평가

○ 마우스를 이용한 동물 실험
-키토산 하이드로겔의 상처난 피부의 치료 효능을 확인하기 위하여 Balb C 마우스(수컷, 평균 몸무게: 20 g)를 사용
연구결과
- 1H NMR 분석을 이용하여 가시광선 광경화 가능 수용성 키토산 유도체 합성을 확인함.
- 수용성 키토산 유도체 함량 및 가시광선 광개시제 리보플라빈 몰농도에 따른 하이드로겔 형성 유무를 확인함.
- 수용성 키토산 유도체 IV의 함량과 리보플라빈의 몰농도가 증가할수록, G′와 G″는 증가하였고, G′와 G″의 차이도 현저히 증가하였음. 또한, 하이드로겔의 stffiness는 IV의 함량과 리보플라빈의 몰농도가 증가할수록 높아짐을 확인하였음.
-하이드로겔의 swelling ratio는 수용성 키토산 유도체 IV의 함량이 증가할수록, 리보플라빈의 몰농도가 감소할수록 커짐을 확인하였음.
-수용성 키토산 유도체 IV의 함량이 증가할수록, 리보플라빈의 몰농도가 감소할수록 각 하이드로겔의 분해거동이 빨리 일어남을 확인하였음.
-1일동안의 cell culture에서는 수용성 키토산 유도체 IV의 함량과 리보플라빈의 몰농도 변화에 따른 cell viability의 차이가 거의 없었으나, 7일 이후부터는 IV의 함량이 증가할수록, 리보플라빈의 몰농도가 작을수록 cell viability는 증가함을 확인하였음.
-PDGF-BB, VEGF, PDGF-BB/VEGF, EGF 및 curcumin 방출이 서방형 거동을 보여줌을 확인하였음.
-동물실험을 통해 PDGF-BB, VEGF, PDGF-BB/VEGF, EGF 및 curcumin을 함유한 하이드로겔에서 상처 치유가 촉짐됨을 확인하였음.

Result
- The successful preparation of chitosan derivative for visible photo-curing was confirmed by 1H NMR spectroscopy.
- Hydrogel formation was affected by the amount of chitosan derivative and the concentration of riboflavin.
- The larger the amount of chitosan derivative and the concentration of riboflavin, the larger the storage (G′)/loss (G″) modulus and the higher stiffness.
- The larger the amount of chitosan derivative and the lower the concentration of riboflavin, the higher the swelling ratio.
- The larger the amount of chitosan derivative and the lower the concentration of riboflavin, the quicker the degradation behavior.
- At day 1, little difference in the cell viability was observed on all samples. At day 7, the cell viability improved by the increased amount of chitosan derivative and the decreased concentration of riboflavin.
- Grwoth factors and/or curcumin-loaded hydrogels showed controlled releases of PDGF-BB, VEGF, PDGF-BB/VEGF, EGF and curcumin in a sustained manner.
