연구의 목적 및 내용
본 연구는 유전자 재조합 기술을 이용하여 세포 및 피부 조직으로의 침투성이 우수한 저분자량 프로타민을 성장인자의 N-말단에 융합시킨 3종 이상의 피부흡수 증진 신규 융합 성장인자를 제조한 후 이들 융합 성장인자 복합체의 피부세포 활성 및 상처치유 시너지효능을 나타낼 수 있는 최적의 농도 조합비를 도출하고 코어-쉘 형태의 나노 캡슐 및 양이온성 탄성 리포좀과 같은 피부 전달체 내부에 융합 성장인자 복합체를 포함시킨 외용제를 개발하여 급, 만성창상 부위에 도포 시 성장인자의 신속한 피부흡수와 성장인자 복합체의

본 연구는 유전자 재조합 기술을 이용하여 세포 및 피부 조직으로의 침투성이 우수한 저분자량 프로타민을 성장인자의 N-말단에 융합시킨 3종 이상의 피부흡수 증진 신규 융합 성장인자를 제조한 후 이들 융합 성장인자 복합체의 피부세포 활성 및 상처치유 시너지효능을 나타낼 수 있는 최적의 농도 조합비를 도출하고 코어-쉘 형태의 나노 캡슐 및 양이온성 탄성 리포좀과 같은 피부 전달체 내부에 융합 성장인자 복합체를 포함시킨 외용제를 개발하여 급, 만성창상 부위에 도포 시 성장인자의 신속한 피부흡수와 성장인자 복합체의 상승 작용을 통해 신속한 상치회복과 조직재생의 시너지 효과를 나타낼 수 있는 상처치유 바이오 의약품을 연구, 개발하는 것을 목표로 함.

연구결과
○ 피부흡수 증진 신규 융합 성장인자 3종 (rLMWP-EGF, rLMWP-IGF-I, rLMWP-PDGF-A)의 제조 시스템 구축 및 순도/분자량/아미노산 서열을 확인함.
○ 융합 성장인자의 피부투과도 증진 검증 (24 시간 후 헤어리스 마우스 피부를 통한 투과도): rLMWP-EGF는 0.03±0.01 μg/㎠/h 투과됨 (rhEGF 투과 안 됨), rLMWP-IGF-I 및 rLMWP-PDGF-A는 일반 성장인자 대비 각각 124%, 164% 증가
○ LMWP-융합 성장인자는 일반 성장인자 대비 우수한 피부 세포막 투과도를 나타냄.

Exogenous administration of growth factors has potential benefits in wound healing; however, limited percutaneous absorption, inconsistent efficacy, and high doses have hampered successful clinical use. To overcome these restrictions, we focused on the development of a topical formulation composed of highly skin-permeable multimeric nano-complex of growth factors. In the present study, we fused low-molecular-weight protamine (LMWP) with epidermal growth factor, insulin-like growth factor-I, and platelet-derived growth factor-A (rLMWP-EGF, rLMWP-IGF-I, and rLMWP-PDGF-A) via genetic modification. Then, we used in vitro cell proliferation studies to assess the biological activity as well as the benefits of the combination. After confirming the enhanced skin permeability, the fused growth factor complexes were nano-encapsulated by a dual-axial ultrasonic nebulizer or loaded into the cationic elastic nano-liposomes using double emulsion with microfluidization. Finally, therapeutic effects of the fused growth factor complex-loaded topical formulation on wound healing and tissue repair were assessed in acute and diabetic mouse models.
Result
○ The actual molecular weights and N-terminal amino acid sequences of the LMWP-fused EGF, IGF-I, and PDGF-A were consistent with the theoretical values and sequences.
○ rLMWP-EGF showed significantly enhanced skin permeation, and rLMWP-IGF-I and rLMWP-PDGF-A across excised mouse skin were 124% and 164% higher, respectively, than the native forms.
○ Combinations of native or LMWP-fused growth factors significantly promoted fibroblast proliferation and the synergy of LMWP-EGF, LMWP-IGF-I and LMWP-PDGF-A was optimal at a ratio of 100:100:10 by concentration.
○ The growth factor complex-loaded nano-emulsion appeared to be spherical and the average droplet diameter was 127±4.30 nm.
○ The LMWP-fused growth factor complex and hyaluronic acid (HA) combination induced greater fibroblast proliferation and collagen synthesis, and the optimal proportions of LMWP-fused growth factors and HA were 1, 1, 0.02, and 200, respectively.
○ The LMWP-fused growth factor-HA complexes were incorporated into cationic elastic liposomes (ELs) of 107±0.757 nm in diameter.
○ Topical delivery of the LMWP-fused growth factor complexes accelerated wound re-epithelialization significantly, accompanied by the formation of healthy granulation tissue, within 9 days compared with vehicle.
○ The cationic ELs containing the LMWP-fused growth factors-HA complex also significantly accelerated the wound closure rate in a diabetic mouse model.
Expected Contribution
○ The LMWP-fused growth factor complex delivery system might be a candidate for wound-healing therapy.
○ Additional studies will be carried out to examine the enhanced healing effects on various types of wound, including chronic ulcers and burns.
