본 연구의 목적은 천연물 신약개발의 3대 요소인 천연물화학, 합성화학, 생화학을 융합시켜 해양천연물 유래 화합물 라이브러리를 구축하고 이를 기반으로 천연물신약의 소재를 개발하는 것임. 본 연구에서는 천연물 탐색을 위하여 한국 근해와 열대 태평양 등지에서 265 여 종, 2,400 kg의 저서 군체동물을 체계적으로 채집, 동정하고 생리활성검색에 의하여 50 종의 유용시료를 파악, 이들 중 일차적으로 15 종의 해면 및 군체 멍게로부터 202 종의 천연물을 분리, 정제하였고 이들 중 130 종의 신물질의 구조를 분광학과 화학적 방법으로
본 연구의 목적은 천연물 신약개발의 3대 요소인 천연물화학, 합성화학, 생화학을 융합시켜 해양천연물 유래 화합물 라이브러리를 구축하고 이를 기반으로 천연물신약의 소재를 개발하는 것임. 본 연구에서는 천연물 탐색을 위하여 한국 근해와 열대 태평양 등지에서 265 여 종, 2,400 kg의 저서 군체동물을 체계적으로 채집, 동정하고 생리활성검색에 의하여 50 종의 유용시료를 파악, 이들 중 일차적으로 15 종의 해면 및 군체 멍게로부터 202 종의 천연물을 분리, 정제하였고 이들 중 130 종의 신물질의 구조를 분광학과 화학적 방법으로 규명하였음. 다양한 해양 동식물과 해저 퇴적물로부터 2,689 여종의 방선균과 진균을 분리, 보존하였으며 이 과정에서 심해퇴적물 채집 장치를 고안, 제작하였음. 화학적, 생화학적 분석을 종합하여 다수의 고역가 세포독성 및 항 미생물 활성균주를 파악하고 대량 배양된 균주로부터 신물질 106 종을 포함하는 312 종의 천연물 분리정제 완료, 천연물의 구조를 규명하였음. 이와 같이 514 종의 해양천연물을 확보하였음. 또한 합성 화학적 기법을 통하여 13 종의 활성이 규명된 해양천연물의 전합성을 완료하였고 이들 유래 신규 활성유도체 300 종을 제조하였음. 천연물과 합성 유도체에 대해서는 항암, 항 미생물 및 이들과 관련된 다양한 효소저해 활성의 검색결과로 126 종의 Hit물질을 선정하였음. 선정된 고역가 활성물질에 대해서는 in vivo 활성 및 작용기전, 대량생산 등의 후속연구를 통하여 5 종의 선도물질 후보를 도출하였음. 종합하여 본 연구를 통하여 해양생물의 천연물신약 개발가능성을 입증하였으며 해양신물질 유래 신약 선도물질의 개발을 위한 기술적 기반을 확립하고 향후의 산업화를 위한 소재를 도출하였음

1. Benthic colonial invertebrates have been systematically collected from various locations of Korea, Micronesia and Indonesia. These samples weighing more than 2,400 kg, have been classified into more than 265 species of sponges and tunicates. Fifty specimens have been selected for chemical investigation on the basis of combined results of bioactivity tests, TLC and proton NMR analysis of the crude organic extracts. In addition, 2,689 microbial strains of 1,347 actinomycetes and 1,342 fungi have been isolated from diverse locations. For this purpose, a device to collect mid-depth sediments has been designed and prepared. These strains have been categorized and stored for future research. Strains exhibiting unique chemical profile and potent bioactivity have been selected and cultivated for chemical investigation.
2. From 15 selected sponges and colonial tunicates, 202 compounds have been isolated and 130 new ones among these have been structurally elucidated by combined chemical and spectral methods. Similarly 312 compounds have been isolated from the culture broths of selected microbes and 106 new metabolites have been structurally elucidated by combined spectroscopic, chemical and crystallographic methods. The structures of these 514 compounds are very diverse and includes those from all of the major biogenetic origins.
3. Based upon the structural novelty and bioactivity, selected marine natural products have been chemically derivatized to 300 novel compounds with systematic carbon skeletons and diversified functional groups.
4. Based upon the combined results mentioned above, a marine natural products library has been prepared consisting of 514 natural products, 183 previously obtained ones and 300 synthetic derivatives. These compounds are derived from diverse biogenetic origins, thus structurally very diverse: terpenoids, polyketides, alkaloids, peptides, saponins, aromatics, bases, mixed biogenetic products, etc, and possess potent bioactivity.
5. For the natural products, and synthetic derivatives, cytotoxicity has been measured against 6 cancer cell-lines to produce 61 Hit compounds. Those including halichondramides, wondonins, F452-R (asperphenins), and salternamide A were further studied for signal transduction and in vivo animal study for anticancer activity. In addition, antimicrobial activity has been also tested against pathogenic bacterial and fungal strains. Additional tests have been performed for the inhibitory activities against Srt A, ICL, Na+/K+-ATPase as well as bioactive mechanisms including actin depolymerization, anti-angiogenesis, cancer-metastasis, and transcription that has resulted in the selection of 65 Hit compounds. In particular, 2 synthetic compounds from psammaplin A have showed potent inhibition against pathogenic bacteria and MRSA as well as in vivo animal study. Potent compounds were analyzed for HPLC, physico-chemical properties, and pharmacokinetics.
6. For the mass supply of Hit and/or potent compounds, 13 marine natural products such as psammaplin A have been schemed for total syntheses and succeeded. In addition, 4 natural products including F452-R (asperphenin) have been isolated in large-scale.
7. Based upon combined evaluation on synthetic, natural products, bioactivity, and/or pharmaceutical studies on Hit compounds, have selected are 5 candidates of Lead compounds for the advanced study on marine-derived drug development. Those are wondonin, asperphenin, psammaplin (two groups) and ohmyungsamycin.
