Rats (Sprague-Dawley, Wistar and spontaneously hypertensive rats) were anesthetized with thiopental (50 mg/kg, IP) and right femoral artery and vein were cannulated to monitor arterial blood pressure and to serve as an intravenous infusion route, respectively. The magnitude of relaxation was not different between Wistar and spontaneously hypertensive rats. In addition, the inhibitory effect of the drugs on the vascular preparations in vitro was not affected by the endothelium removal, suggesting that the relaxation was endothelium-independent. These results suggest thai diazepam and lorazepam have a depressor effect mediated through the central GABAergic system. The absence of a peripheral circulatory effect of these drugs in vivo despite the relaxation effect on the isolated vascular preparations in vitro may be associated with their pharmacokinetic properties in the circulation.
