The cellular and physiological effects of methoprene, juvenile hormone analogue, on developmental stages in the gypsy moth, Lymantria dispar were studied. Methoprene inhibited egg hatching and reduced the wet weights of a hatching larvae. The differences in protein and carbohydrate contents as well as protein patterns between control and methoprene treatment groups were observed during egg development. Chemical compositions of the larval hemolymph were affected by methoprene. Protein, amino acid and lipid contents of the methoprene treatment groups were lower than those of the control groups except for carbohydrate. The activity of the juvenile hormone specific esterase were the lowest in control groups(2.30n mol/min/ml) and increased in the topical treatment groups. Finally it reached the maximum value(3.01n mol/min/ml) in the ingestion group. Treatment with methoprene also prolonged the larval stages, and the male 7th instar larvae were consecutively induced. Pupation was disturbed and the length of ovarioles and number of eggs were decreased. Midgut epithelium was simple column types composed of the columnar, globlet and regeneration cell. In the methoprene treatment groups, morphological changes were observed. The number of lipid droplets and vacuoles of columnar cells were increased. Microvilli of goblet cells became elongated and contained numerous mitochondria. Cytopalsmic extrusion were observed at the early stages and cytoplasm membrane were destroyed at the female 7th instar larvae of the methoprene treatment groups. In larvae, fat bodies of the methoprene treatment groups had inter-cellular spaces and vacuoles. The number of lipid droplets was increased while that of glycogen was decreased. Testes were composed of 4 follicles. In the control groups, normal spermatogenesis were advanced. Lymantria dispar of the methoprene treatment group had small sized testes and spermatogenesis were also progressed, but the maturity were suppressed. Ovaries composed 4 ovarioles which are polytropic ovarioles. In the control group, normal oocytes were matured and yolk yolk protein were formed in cytoplasm. A lot of lipid vesicles and abnormal Oo¨cytes were observed in the methoprene treatment groups. There were processing of yolk formation, numerous glycogen particle and the shape of protein droplets were changed in the methoprene treatment groups. Egg maturation were not completely controlled in the methoprene treated gypsy moth.
