Effects of enalapril and Ginkgo biloba extract on bradykinin-induced cardiovascular responses원문보기

Enalapril, one of ACE inhibitors, resulted dry cough and angio-edema as side effects due to the accumulation of bradykinin. Gingko biloba extract, containing various kinds of flavonoids, was examined whether it could reduce these side effects of enalapril. Bradykinin induced different vascular responses, such as vasocontraction in the artery through B_(1) receptor and vasodilation in the arteriole through B_(2) receptor. Bradykinin (10^(-9)-10^(-7)g/ml) resulted dose-dependent contractile response in the isolated and perfused ring form of thoracic aorta of the rat. But the contractile response was showed tachyphylaxis and did not induced in the endothelium cell damaged vessels. Pretreatment of indomethacin 10^(-5)M completely inhibited the contraction by bradykinin, while pretreatment of L-NAME 10^(-5)M did not affect at all. These results would be suggested that bradykinin induced the contraction of arota by the interaction with B_(1) receptor located in endothelial cells through the production of prostaglandins. Pretreatment of enalapril 10^(-5)g/ml alone or combination with Gingko biloba extract 10^(-5)g/ml increased the sensitivity of contraction by bradykinin, but pretreatment of Gingko biloba extract alone did not affect. Bradykinin 10㎍/kg, i.v. resulted the decrease of systolic and diastolic blood pressures such as -23±4mmHg and -38±6mmHg, respectively in urethane and ketamine induced anesthetized rat. The decrease of diastolic blood pressure by bradykinin was recovered to the normal level within 1 minute and showed repeatedly. Pretreatment of indomethacin or L-NAME did not affect at all. These data indicated that bradykinin resulted the arteriolar vasodilation by B_(2) receptor, but neither through prostaglandins nor NO. Pretreatment of enalapril 1mg/kg resulted the sustained decrease of diastolic blood pressure by bradykinin such as more than 30minutes. Pretreatment of Gingko biloba extract 2mg/kg alone did not affect it, but coadministration of enalapril and Gingko biloba extract lasted it only for 3 min and recover to the normal level. These results indicated that Gingko biloba extract could inhibit the increased response of bradykinin B_(2) receptor induced by enalapril, so Gingko biloba extract could be reduce the side effects of dry cough induced by enalapril.
