Capsaicin is capable of releasing various neuropeptides from A delta and C type nerve fibers. Among sensory neuropeptides, the tachykinin substance P(SP) shows the best correlation with capsaicin sensitivity. Capsaicin induces the release of SP, and depletes and prevents reaccumulation of SP in C type nerve fibers. SP is mainly involved in pain transmission and in the process of tissue repair. SP is also believed to play an important role in hair growth and cycle, because of the prominence of SP-immunoreactive nerve fibers around hair follicles. Though some indications point to a role for SP in growth control of the hair follicle, a possible trophic role of SP has not yet been studied and the neural mechanisms of hair growth control remain obscure. Minoxidil is widely used as a topical agent for hair loss. Although the mechanism of hair growth by minoxidil is not fully known, the hypothesis includes increasing blood flow in the skin by peripheral vasodilation, stimulating the follicular cell proliferation, opening of intracelluar potassium channels, and modulating immune responses. In this paper, the effects of coapplication of capsaicin and minoxidil on hair growth and cycle, compared with application of capsaicin or minoxidil alone, was evaluated. The results are as follows: 1. On macroscopic examination, hair growth in the depilated area of capsaicin group and minoxidil group began faster than that of coapplied group and control. Thereafter, hair growth of capsaicin group was observed as the fastest, followed by minoxidil group, coapplied group, and control. 2. On Phototrichogram, the percentage of the area of hair regrowth in coapplied group was lower than capsaicin group, similar to minoxidil group, and higher than control at the 15th day after depilation. Though the percentage of the area of hair growth in the coapplied group was highest at the 30th day after depilation, there was no significant difference between coapplied group and capsaicin group. 3. The hair of capsaicin group stopped to grow 20 days after depilation, and that of minoxidil group stopped to grow 25 days after depilation. However, the hair of coapplied group grew steadily up to the 30th day after depilation, growing taller than any other group. 4. On microscopic examination, the anagen stage in capsaicin group and minoxidil group was induced faster than in coapplied group and control. Capsaicin was able to make the hair cycle faster than control, but minoxidil group and coapplied group prolonged the anagen phase of hair cycle. 5. Though mast cell count decreased in all four groups, capsaicin group decreased significantly more than the other group, and maintained the low count onto the 30th day after depilation. Mast cell count of coapplied group was similar to that of minoxidil group. 6. [Methyl-^(3)H] thymidine uptake increased in all four groups until the 5th day after depilation, and then decreased. The uptake of capsaicin group and control were similarly increased on the 5th day, maintained until the 10th day after, then decreased suddenly on the 15th day and persisted to the 30th day after depilation. The uptake of coapplied group was similar to that of minoxidil group, increasing on the 5th days after depilation, and decreasing slowly from the 5th day to the 30th day after depilation. These results suggest that capsaicin can not only induce the anagen phase quickly, but also sustain constant effect on hair growth. Minoxidil can also induce the anagen phase fast, and prolong the anagen phase of the hair cycle. Therefore, it is concluded that the coapplication of capsaicin and minoxidil can grow hair steadily.
