본 연구에 사용된 실험동물은 연세대학교 의과대학 임상의학 연구센터 내에 있는 실험동물부에서 분양 받은 특정병원체 부재동물로서 평균체중이 113.0∼117.8g 되는 6주령의 고혈압쥐(SHR) 수컷 24마리와 대조군으로서 평균체중이 116.8g인 6주령의 WKY계 수컷 6마리가 사용되었다.
실험동물은 크게 다섯 그룹으로 나누어, 정상군(WC; WKY Control), 고혈압 통제군(HC; SHR Control), 고혈압 투여군(HA; SHR L-arginine), 고혈압 훈련군(HT; SHR Trained), 그리고 고혈압 투여훈련군(AT; SHR L-arginine Trained)으로 구분하였고, 각 그룹별로 안정시(Resting)와 운동종료시(Exercise)로 세분하였다.
실험동물의 훈련은 총 9주간의 수영훈련으로서 25℃∼27℃의 수온에서 1주간의 환경적응과 1주간의 수영 적응훈련 후, 본 운동시간을 5분간 점진적으로 증가 시켜 60분 시점에서는 훈련 마지막까지 지속하였으며, L-arginine은 음료에 2% 농도로 용해하여 ad-libitum 방법으로 9주간 투여하였다. 채혈 및 조직샘플링은 연세대학교 의과대학 동물수술실험실에서 이루어졌으며, 그룹별 실험처치 후 에틸에테르(ethyl ether)를 이용하여 마취한 후 복부대동맥에서 전혈을 채취하였고, 채혈 후 2g의 간조직을 샘플링 하였다.
혈압 및 심박수의 측정은 일본 Muromachi kikai(社)의 BP Monitor MK-1100을 사용하였고, 지질성분의 측정은 일본 Hitachi 747과 7150을 이용하여 분석하였으며, GPX 활성은 Roche社의 cobas MIRA를 이용한 UV법으로 측정하였고, MDA와 eNOS 활성은 해당 Kit를 사용하여 spectrophotometer로 분석하였다.
고혈압 통제군의 체중은 정상군에 비해서 크게 증가하였으며(p<.05), 유산소 훈련으로 체중 증가폭은 유의하게 감소되었다(p<.05). 그러나, L-arginine 투여로 인한 체중의 변화는 증가하는 경향을 나타내었다. 심박수와 혈압은 고혈압 통제군이 정상군에 비해서 상당히 높았으나(p<.05), L-arginine 투여와 유산소 훈련에 따라 감소되는 효과를 보였으며(p<.05), L-arginine 투여보다는 훈련을 통한 감소 효과가 두드러지게 나타났다.

The purpose of this study is to investigate the effects of 9 weeks swimming training and L-arginine supplementation on heart rate, blood pressure, blood lipid components, GPX activity, MDA level and eNOS activity in SHR (Spontaneously Hypertensive Rat).
Twenty four male SHR (six-week-old) were divided into four groups consisting of the six SHR control (HC), six SHR L-arginine (HA), six SHR trained (HT), and six SHR L-arginine trained (AT). Six age- and weight-matched normal Wistar-Kyoto (WKY) rats were used as normal control (WC). All of the groups divided into resting and exercising again.
Animals were supplied from Department of Laboratory Animal Medicine, Yonsei University College of Medicine. Animals were cared in SPF barrier area. During the 9-week treatment period, trained groups of the present study participated in a regular swimming training program five times per week, and L-arginine groups, 2% L-arginine solution were supplied ad libitum. Whereas, WC and HC groups did not participate in either training or supplementation.
At the end of the trial, animal killed and samples were taken at the Animal Operation Room of Department of Laboratory Animal Medicine, Yonsei University College of Medicine, and measurement of serum lipid components (TG, T-C, LDL-C and HDL-C), erythrocyte antioxidant enzyme (GPX), lipid peroxidation (MDA), and liver endothelial nitric oxide synthetase (eNOS) was carried out well-established methods. Heart rate and blood pressure were measured in all animals, using a standard tailcuff method.
The results obtained were analyzed via one-way ANOVA(analysis of variance), paired t-test, and Pearson's correlation using SAS program. A value of p<.05 was considered statistically significant.
From this study it was found that body weight(WC, 292.8±16.17; HC, 312.5±6.41;p<.05), heart rate(WC, 346.7±78.76; HC, 457.7±12.83;p<.05), and systolic blood pressure(WC, 130.2±4.4; HC, 213.0±4.2;p<.05) were significantly higher in hypertensive than in normal rats. GPX activity was significantly lower in hypertensive than normal rats(WC, 13,877±2,364; HC, 9,098±,362;p<.05). Lipid peroxidation was significantly higher in hypertensive than in normal rats(WC, 1.37±0.21; HC, 1.76±0.53;p<.05). Also, eNOS activity was significantly higher in hypertensive than in normal rats(WC, 5.18±3.16; HC, 12.83±6.96;p<.05).
After the L-arginine supplementation and swimming training trial, it was found that in treated groups rise of body weight, heart rate, and blood pressure were prevented significantly (p<.05) and GPX activity in erythrocyte was significantly improved more than in the SHR controls (p<.05). In contrast, lipid peroxidation level and eNOS activity were reduced more than in the SHR controls. All of the lipid peroxidation levels were increased after highly intensive exercise, on the contrary, the GPX activities were decreased after same exercise only except HC. Although all the three treated groups (HA, HT, AT) and normal group (WC) showed increased or changeless eNOS activities, after exercise only HC group was decreaed.
In the mean blood lipid component levels, there was a significant difference among the normal control, hypertensive control and treated groups (p<.05). Due to L-arginine supplementation and swimming training trial, all of the blood lipid component levels were decreased greatly (p<.05) except for HDL-C and TG.
Correlation studies showed that TG and eNOS were significantly positively correlated with blood pressure in hypertensive rats (r=.966; r=.831). HDL-C and blood pressure showed a negative correlation (r=-.866). In lipid peroxidation, with GPX and eNOS were significantly positively correlated (r=.897; r=.797), however with HDL-C was negatively correlated in hypertensive rats (r=-.756).
In conclusion it was found that regular exercise training and supplement of L-arginine may prevent development of increased heart rate and blood pressure, reduce lipid peroxidation, enhance GPX activity, and stabilize sensitive eNOS activity.
However, many factors other than aerobic training and L-arginine affect cardiovascular reponses, antioxidant system, and NO synthesis mechanism. Additional factors such as various tissue damage bio-indexes, non enzymetic antioxidant matters, or NOS isomers must be considered in order to present a more complete assessment of the factors contributing to cardiovascular disease.
This paper firmly establishes the groundwork for further research to be conducted in this area.
