인삼(Panax ginseng C. A. Meyer)의 알콜 추출 공정 후 발생하는 인삼박으로부터 다당체와 올리고당을 분리하여 각각 복강 대식세포(peritoneal macrophage)의 면역활성 증가 효과와 장내미생물 생육 효과에 대하여 연구하였다. 인삼박에 상업용 식물세포벽 가수분해효소 처리(EP3)와 압출성형 후 열수추출법(EGP1)에 의하여 다당체를 분리한 후 Balb/c mice의 복강 대식세포의 활성을 in vitro에서 검색한 결과 EP3와 EGP1이 농도 50 ㎍/㎖에서 lysosomal phosphatase activity가 각각 174.7%와 153.1%로 유의적인 활성을 나타내었으며, NO 및 H_(2)O_(2) 생성에서도 농도 의존적인 활성을 보였다(P<0.05). 또한 in vivo에서도 이들 다당류는 자극이 없는 기저상태 뿐만 아니라, mitogen(LPS)에 의해 자극된 상태에서도 NO 생성량이 대조군에 비해 유의적으로 높은 활성을 보였다(P<0.05). EP3와 EGP1의 조성은 각각 33.9%와 32.7%의 uronic acid, 65.6%와 65.1%의 중성당(galactose, arabinose, rhamnose, glucose), 0.5%와 2.2%의 단백질로 구성되었다. 인삼박 올리고당의 최적 효소로서 cellulase와 pectinase를 복합적으로 사용였고, Recycling Preparative HPLC로 올리고당을 분리하였다. 최적 효소 조건에서의 올리고당 수율은 17.3%이었으며, 78.8%의 glucose로 구성되었다. 인삼박 올리고당의 장내미생물에 대한 효과를 in vitro로서 확인한 결과 Bifidobacterium species는 인삼박 올리고당을 선택적으로 이용하였고, Cl. perfringens는 전혀 이용하지 못하였다. 위와 같은 인삼박의 면역계 및 장내미생물에 대한 생물활성은 앞으로 의약 및 식품 등의 이용분야의 연구를 통하여 새로운 기능성 소재로서 산업적으로 활용성이 높을 것으로 기대된다.

The biological activities of ginseng marc polysaccharides (GMPs) and oligosaccharides (GOs) isolated from ginseng marc were investigated in terms of macrophage-immunostimulating activities and the growth effect of intestinal microorganism, respectively. Two polysaccharides were isolated by commercial enzyme hydrolysis (EP3) and hot water extract of extruded ginseng marc (EGP1), which were used to measure macrophage activities in Balb/c mice under in vitro and in vivo conditions. EP3 and EGP1 increased lysosomal phosphatase activity, NO and H_(2)O_(2) production in a dose dependant manner in vitro (P<0.05). Under in vivo condition, EP3 and EGP1 showed 130% NO production when the macrophage was triggered with 20 ㎍/㎖lipopolysaccharide (LPS) as a mitogen. EP3 and EGP1 were composed of 33.9%, 32.7% uronic acid, 65.6%, 65.1% neutral sugar and 0.5%, 2.2% protein, respectively. Optimal enzyme for the isolation of GOs from ginsneg marc was found to be the mixture of Econase CE and Pectinex Ultra SP-L (6:4, w/w). GOs were effectively separated from enzyme hydrolysates using the recycling preparative HPLC. Under the optimal condition, the yield of GOs were 17.3% and it was composed of 78.8% glucose. GOs promoted selectively the growth of Bifidobacterium species, but not that of Cl. perfringens under in vitro conditions. The observed immune activity of GMPs and the growth effect for intestinal microorganism of GOs suggest that GMPs and GOs have high potentials as functional food ingredients or nutraceuticals.
