To achieve effective tissue regeneration, biodegradable chitosan-poly(L-lactic acid)(PLLA) barrier membrane and nonwoven chitosan fibrous matrix was developed. The use of growth factors with stimulatory effects on cell migration and proliferation and synthesis of matrix components by the cells has been proposed ot promote tissue regeneration. Chitosan-PLLA membranes were prepared by casting PLLA-methylene chloride-ethyl acetate solution on between two chitosan fibrous meshes. Nonwoven chitosan fibrous matrix was highly porous polymer 3-D scaffold using fiber bonding technique. The morphological cahracteristic of chitosan fiber based devices and mechanical property of the chitosan-PLLA membrane were evaluated. And its stability of membrane and degradability of matrix in phosphate buffer medium were evaluated.
Moreover, to shorten the bone-forming period and to enhance the bone-forming efficacy, the feasivility of using the devices as controlled release delivery vehicles of PDGF-BB was monitored. Finally, osteoblast attachment and proliferation test and guided bone regeneration potential were evaluated as wegg. The results showed that PLLA membrane was located between two chitosan fibrous fivrous meshes.
