흡연은 체내에서 활성산소종(ROS)를 증가시키어 혈액 내 지질과 지단백의 변화, 산화된 LDL에 의한 내피세포의 손상 등을 유발하므로 심혈관질환 발병의 주요 위험인자로 여겨지고 있으며, 과도한 DNA의 산화는 발암원인이 될 수도 있기 때문에 흡연으로 인한 산화적 손상의 방어기전으로는 항산화 영양상태의 개선이 제시되고 있다. 본 연구에서는 TRAP 및 Comet assay를 통해 효과가 좋은 신선초를 선택하였고, 흡연자 54명을 대상으로 신선초 녹즙을 6주간 매일 300ml씩 보충 섭취시킨 후, 항산화 영양상태, 지질양상 및 지질 과산화와 DNA손상에 대한 개선효과를 보고자 하였다. 대상자들의 나이 및 식이섭취 조사, 신체계측 조사를 수행하였고, 녹즙 섭취 전후로 두 번의 채혈을 거쳐 혈장 항산화 비타민, 혈장 TRAP, 적혈구 항산화 효소, 혈장 지질 및 지단백 양상, CD로 본 산화 LDL 수준, Comet assay를 이용한 DNA손상정도, 간기능 정도를 보았는데, 그 결과는 다음과 같다. 1) TRAP으로 항산화 능력이 좋은 신선초를 선택하였고, 세포의 DNA손상보호효과를 Comet assay로 확인하였다. 2) 비타민 C와 TRAP은 녹즙섭취 전후에 차이가 없었으나, 비타민 E, β-carotene이 녹즙 섭취 후에 증가하였다. 3)항산화효소인 catalase, SOD는 녹즙섭취 후에 증가하였으나, GSH-Px는 변함없었다. 5) Comet assay를 통해 본 DNA 손상은 녹즙섭취 후에 감소하였다. 6) 녹즙섭취 후에 TC, LDL-C은 감소하였으나 TG와 HDL-C은 차이가 없었다. 7) LDL의 산화생성물인 CD는 녹즙섭취 후에 감소하였다. 8) 녹즙섭취 후에 GOT는 변함이 없었지만, GPT는 감소하였다. 본 연구 결과 흡연자의 녹즙보충은 항산화 상태와 지질양상을 개선시켰고 DNA손상회복, 지질과산화 정도 및 간기능에 있어서 개선효과를 기대해볼 수 있었다.

Smoking has been well known to be associated with increased indices of free radical-mediated damage of DNA, indicating that smoking may exacerbate the initiation and propagation of oxidative stresses, which are potential underlying processes in the pathogenesis of many diseases. The purpose of this study was to evaluate whether a daily regimen of green vegetable juice supplementation to smokers can be protective against endogenous lymphocytic DNA damage and whether grape juice consumption could enhance other antioxidant status and lipid profiles. Fifty-four smokers aged 31-47 were given 300ml of green vegetable juice every day for 6weeks in addition to their normal diet, and blood samples were drawn before and after the intervention. The 6 weeks of green vegetable juice consumption resulted in a significant decrease(P=0.000, by paired t-test) in lymphocytes DNA damage expressed by both TL (before: 88.61±1.66 vs after: 62.59±1.43) and TM (before: 23.1±1.53 vs after 15.75±0.8). Vitamin C, TRAP level was not signifcantly changed after the supplementation, but, vitamin E, β-carotene level was significantly increased after the intervention. Antioxidant enzymes, catalase, SOD levels were significantly increased after the trials. Plasma TC, LDL-C level were increased, while TG and HDL-C was not affected by grape juice consumption. Oxidized-LDL measured by level of CD was unaffected. Plasma GPT marker of liver function was significantly reduced after the supplementation. In conclusion, these results support the hypothesis that green vegetable juice rich in carotenoids exert a cancer-protective effect partially via a decrease in oxidative damage to DNA and lipid peroxidation accompanied by improving antioxidant system, lipid profiles and liver funciton.
