Caspase-3 protease의 효소적 활성은 삼산화비소가 처리된 HeLa 세포에서 caspase-3 protease의 분절과 PARP의 분절에 의해 확인되었다. 게다가 삼산화비소는 미토콘드리아에서 세포질 내로 cytocrome c를 방출과 미토콘드리아의 막전위의 변화를 유도하였다. 항산화제인 GSH는 삼산화비소가 처리된 HeLa 세포에서 caspase-3 protease의 효소적 활성과 PARP 분절을 차단하여 세포고사를 억제하였다. 따라서, 이들 결과는 삼산화비소가 HeLa 세포의 세포고사시에 항산화제로 작용하며, 미토콘드리아 기능변화 뿐만 아니라 일련의 caspase들의 활성을 통해 세포고사를 유도한다는 것을 나타낸다.

Aresenic trioxide (As_(2)O_(3)) has been used as an anti-cancer agent in traditional Chinese medicine. As_(2)O_(3), at a low dose, induces clinical remission in patients with acute promyelocytic leukemia (APL) through induction of apoptosis. Despite extensive research, however, the exact mechanism behind the As_(2)O_(3)-induced apoptosis of solid tumor cells still remains unanswered. To investigate the medical insights of As_(2)O_(3) in cervical cancer cells, human cervical cancer cell line HeLa was employed. Herein, the data demonstrated that treatment of As_(2)O_(3) decreased the viability of HeLa cells in a time and dose-dependent manner. As_(2)O_(3)-induced cell death was confirmed as apoptosis characterized by chromatin condensation and ladder-pattern fragmentation of genomic DNA. Also, As_(2)O_(3) increased the catalytic activity of caspase-3, -6, -8 and -9 protease and the expression of Fas and Fas ligand. Activation of caspase-3 protease was further confirmed by degradation of procaspase-3 protease and poly (ADP-ribose) polymerase (PARP) by As_(2)O_(3) in HeLa cells. Furthermore, As_(2)O_(3) induced the changes of mitochondrial transmembrane potential (MTP) and the cytosolic release of cytochrome c from mitochondria. Antioxidant, including GSH, markedly inhibited the apoptosis of HeLa cells with consistent suppression of the enzymatic activity of caspase-3 protease and PARP cleavage in As_(2)O_(3)-treated cells. Taken together, these results suggest that As_(2)O_(3) functions as an oxidant in apoptosis of HeLa cells via activation of intrinsic caspase cascades as well as mitochondrial dysfunction.
