DNA genome을 갖는 바이러스이지만 HBV의 genome 복제과정은 retrovirus와 유사하게 RNA를 주형으로 역전사반응을 통하여 DNA genome을 복제한다. 이런 복제과정을 catalysis를 하는 HBV DNA 중합효소는 terminal protein, spacer, reverse transcriptase, Rnase H, 네 개의 domain으로 구성되어 있으며, HBV DNA중합 효소는 역전사 효소 활성을 가지고 있다. 바이러스 감염 후 host RNA polymerase II에 의해 전사된 pregenomic(pg) RNA가 core, polymerase와 함께 encapsidation이란 과정을 거쳐 core particle을 이룬 후에 이 core particle 안에서 역전사 반응에 의해 바이러스 유전자 합성이 이루어진다. 이런 역전사 반응은 다음 4가지 단계, protein-priming, minus strand DNA 합성, plus strand DNA 합성, circularization의 과정으로 구분할 수 있으며, template switching에 의해 linear RNA 주형을 circular duplex genome으로 전환된다. 이들 과정에서 duck hepatitis B virus(DHBV) 중합효소에 대한 연구는 비교적 많이 알려져 있으나, 사람이 숙주인 HBV 중합효소의 경우는 아직 이런 encapsidation과 역 전사과정에 작용하는 domain들이 규명되지 않았다. 본 연구에서는 HBV polymerase가 encapsidation과 역 전사 반응에 있어 필수 단백질로서 encapsidation과 역 전사 반응에 어떻게 작용하는지 알아보기 위하여 charge-to-alanine (CA) mutagenesis를 통해 연구하였다. 구체적으로, CA mutagenesis는 다섯 개의 연속된 amino acid residues 중 2개 이상이 전하를 갖는 residue (Arg, Lys, Asp, Glu)가 있는 경우 이를 alanine으로 치환하는 것으로 단백질 표면에 위치하는 전하를 갖는 residue가 protein-protein interaction 혹은 protein-nucleic acid interaction에 주로 작용하는데 착안한 기법이다. 연구 결과 HBV polymerase의 TP(terminal protein) 및 RT(reverse transcriptase) domain은 encapsidation과정에 필요한 domains 임을 밝힐 수 있었으며, Rnase H domain의 경우 encapsidation 과정에서는 큰 영향을 주지 않으나, DNA 복제과정 초기에 작용한다는 사실을 규명할 수 있었다. 기존에 보고된 연구 결과는 주로 Rnase H domain의 catalytic site인 DED residues의 mutagenesis를 통해서 그 작용기전을 살펴 보았다. 그 결과, Rnase H domain의 주요 기능인 pgRNA의 제거가 제대로 일어나지 못해 minus strand DNA 합성 시 elongation이 제대로 일어나지 못 하는 것으로 보고되었다. 또한 Rnase H domain의 sulfur group amino acids을 tyrosine으로 치환시켜 작용기전을 살펴 본 결과 encapsidation과정에 필수적인 것으로 보고되어 왔다. 즉 encapsidation 과 minus strand DNA elongation에 작용한다는 기능만 보고되어 왔었다. 하지만, 이러한 보고들의 mutagenesis는 단백질의 구조와 folding에 영향을 주기 때문에 실험시 그 기능을 발휘할 수 없을 수 있다. 따라서 본 연구에서는 구조와 folding 에 거의 영향을 주지 않는 것으로 알려진 CA mutagenesis를 통해 연구를 수행하였다. 그 결과 encapsidation에 필수적이지는 않지만 minus strand DNA 합성이 전혀 이루어지지 않으므로 복제 초기 과정 중 primer translocation과정에 주요하게 작용하는 것으로 추정된다. 이는 단백질 표면의 전하를 띤 residues이 proteinpriming을 통해 4개의 oligonucleotides을 합성한 후 pgRNA의 3’ 쪽 DR1으로 primer translocation 시 DR1이 아닌 다른 특정 염기서열을 인식하는 것으로 추정된다. 즉, 전하를 띤 residues이 pg RNA와 interaction을 통해 primer translocation을 수행함으로써 minus strand DNA 합성이 일어나도록 그 기능을 발휘함을 새롭게 규명할 수 있었다. 이러한 기능이 어떠한 구조를 통해 이루어지는지를 알아보기 위해서 HBV polymerase의 Rnase H domain을 염기서열의 상동성을 이용한 구조화과정을 통해 model structure를 얻었다. 이러한 model에서 전하를 띤 아미노산들이 Rnase H domain의 구조상에서 표면에 위치하고 있다는 것을 확인할 수 있었으며 이는 전하를 띤 residues가 구조적으로 단백질 표면에 위치하여 pgRNA의 특정 염기서열을 인식, 복제 초기 과정인 primer translocation을 정확하게 수행하는데 그 기능을 수행하고 있다는 가설과 일치한다.

Hepatitis B virus synthesizes its DNA genome using pregenomic RNA (pgRNA) as a template by reverse transcription. The first step of HBV genome replication is the encapsidation of the pgRNA, 3.5 kb, into core particles. Hepatitis B virus (HBV) synthesizes its DNA genome through reverse transcription of an RNA intermediate by HBV DNA polymerase (P). Clustered charged-to-alanine scanning (CA) mutagenesis has been developed to study protein functions. This method is designed to alter the surface of functional domains with minimal misfolding or structural alterations at a distance from the altered site. In this study, we used this powerful technique for systematic mutagenesis of HBV polymerase. Beside of its catalytic role, P protein has the ability to recognizes 5' epsilon element, an encapsidation signal, for encapsidation to occur. P protein, composed of 832 amino acids, contains four subdomains : TP (terminal protein) domain, SP (spacer) domain, RT (reverse transcription) domain, and RH (Rnase H) domain. Specific residues or subdomains of the Pprotein that recognize the RNA template have not determined. To address this, we employed CA mutagenensis of the P protein. Analysis of these CA mutants revealed that TP as well as RT domain is involved in encapsidation being consistent with previous finding in DHBV polymrase. Interestingly, two mutants in RH domains were not defective in encapsidation, but failed to synthesize minus-strand DNA at all, suggesting a role of RH domain in reverse transcription. The result indicated that RH domain influences the earlier steps in the minus-strand DNA synthesis, which is the pimer translocation of HBV polymerase.
