CaSI1은 wouding, drought, salt, ethylene, pathogen 등과 같은 다양한 스트레스 및 신호 전달 물질에 의하여 그 전사량이 증가하였으며 이는 많은 AP2/EREBP 유전자에서 보여졌던 양상과 일치하는데, 실제로 이를 통하여 CaSI1 이 에틸렌 뿐만 아니라 다양한 신호 전달 체계에서 그 역할을 할 수 있을 것으로 생각되어진다. 하위 단계 유전자의 발현 조절과 다양한 스트레스에 대한 저항성조사를 위해 현재 담배 및 Arabidopsis에서 CaSI1 과다 발현 형질 전환 식물체를 제작, 확보 중이며 이러한 형질 전환 식물체와 야생종에서의 유전자 발현 양상의 비교 및 형태적 차이에 대한 고찰은 보다 특정한 신호 전달 체계와 CaSI1의 연관성을 밝히는데 도움을 줄 것이라 사료된다.

In order to adapt itself to various environmental and developmental cues, plant regulates the expression of genes associated to these stimuli. Transcription factors play an important role in this regulation by mediating the expression of target genes by their upstream signal. As a result, the production of proper mRNA and protein enables the plants to effectively adapt to various stresses from environment. To investigate the signal transduction pathway of various signaling molecules, including ethylene, and to study the expression pattern of related genes, we isolated two full-length mung bean cDNA clones (pVR-EIL1 and pVR-EIL2) encoding EIN3 homologs and the hot pepper cDNA clones (pCaSI1) encoding AP2/EREBP domain containing protein. VR-EILs are the homologs of Arabidopsis EIN3, which plays an important role as a transcription factor in ethylene signal transduction pathway. The mRNA accumulations of VR-EILs were not changed by exogenous ethylene treatment, showing that VR-EILs activities are regulated at the post-transcriptional or post-translational level. Genomic Southern blot data indicate that the additional existence of 2 or 3 EIN3 homologous genes excluding VR-EIL1 and 2. In the case of the transgenic tobacco plants that overexpressed the VR-EIL genes, the seedlings displayed a slightly increased sensitivity to exogenous ethylene and a markedly enhanced proliferation of root hairs, one of the typical ethylene response phenotypes. These data indicate that, indeed, VR-EILs play a role as transcrption factors in ethylene signaling. The PR genes encoding β-1,3-glucanase, osmotin and PR1 were constitutively expressed in VR-EIL-overexpressing lines without added ethylene, indicating that VR-EILs are functional in tobacco cells, thereby effectively transactivating the GCC-box-containing PR genes. In contrast, only the low, basal level of tobacco ACC oxidase (NG-ACO1) mRNA was found in all transgenic seedlings examined. These results raise the possibility that the ethylene-induced activation of ACC oxidase gene might not be linked to the activities of VR-EILs, and led us to investigate the effect of ethylene on the level of ACC oxidase transcript in ethylene-related mutants of Arabidopsis. Consistent with their ethylene-insensitive phenotypes, we could not observe any detectable ethylene-induction of ACC oxidase genes (EAT1 and AtACO2) in either etr1-1 or ein2-1 mutants in light condition. However, the amount of both ACC oxidase mRNAs was clearly upregulated following ethylene treatment in ein3-1 green seedlings, suggesting that ACC oxidase could be induced by ethylene whose signaling pathway is not dependent on the function of EIN3 in the existence of light. AP2/EREBP domain-containing protein, a widely known plant-specific transcription factor superfamily, is involved in various stress signaling pathways such as these triggered by biotic and abiotic stress and exists in plant genome as a multi-gene family. From Xag-treated hot pepper cDNA library, we isolated the cDNA clones (pCaSI1) encoding AP2/EREBP domain containing protein. Database analysis revealed that the deduced amino acid sequence of CaSI1 was highly homologous to that of ERF subfamily. In accordance with these result, CaSI1 was localized to the nucleus of the onion epidermal cell, and its binding capacity with GCC box was stronger than with DRE/CRT. Morever, transactivation assay of CaSI1 using yeast one hybrid system showed that 35 N-terminal and 70 C-terminal amino acids of CaSI1 play important roles in its ability as a potential transcriptional activator. The gene expression level of CaSI1 was increased by various stresses and signaling molecules such as wounding, drought, salt, pathogen and ethylene. Through these data, it is thought that CaSI1 play an important role in various signal transduction pathways including ethylene signaling. To identify the regulation mechanism of the downstream gene of CaSI1 in various signal transduction pathways, we constructed transgenic tobacco and Arabidopsis plants that overexpressed the CaSI1 gene. The investigation of the difference between wild type and 35S::CaSI1 transgenic plants focusing on phenotypes and gene expression patterns will elucidate the biological role of CaSI1 in plant defense and stress responses.
