This study was designed to test the effect of dandelion extracts on the metabolism of the streptozotocin - induced diabetic rats. Male Wistar rats were divided into normal, diabetic control and dandelion extract groups. The extracts were prepared by water, ethylacetate and ether from leaf and root, respectively.
Diabetes was induced by injecting streptozotocin(55mg / kg BW, i.p.) in citrate buffer. The extract was supplemented in 11.45 g of raw dandelion /kg diet for 7weeks.
The results were summarized as follows ;
1. Net weight gain and feed efficiency ratio significantly decreased in the diabetic rats. Weight decrease was observed in dandelion extract groups.
2. Relative weight of liver was significantly decreased and those of kidney and spleen were significantly increased in the diabetic control group compared with the normal groups. Decreased weights of liver were improved in the dandelion extract groups.
3. The diabetic rats showed increase in blood glucose level and decrease in plasma insulin level by STZ. Blood glucose level in water extract groups of dandelion leaf and root was significantly lower than in the diabetic control group. HbA1c content was slightly increased in the diabetic control group compared with the normal group and decreased in dandelion extract groups.
4. Plasma albumin content was significantly lowered in the diabetic control group when compared with the normal group and increased in the ethylacetate extract group of dandelion root. In plasma, bilirubin, uric acid and creatinine contents increased in the diabetic control group and dandelion extracts are capable of ameliorating the increased these contents.
5. SOD, CAT and GSH-Px activities in erythrocyte were significantly increased by injecting STZ, which were restored the altered enzyme activities to almost the normal group by dandelion extracts supplementation. The results indicate dandelion was capable of scavenging of reative oxygen species in erythrocyte.
6. In erythrocyte, GSH content decreased in the diabetic control group compared with the normal group. Whereas LPO content was significantly increased versus the diabetic control group and significantly decreased dandelion extract groups.
7. Hepatic cytochrome P-450 content and XO activity were significantly higher in dandelion extract groups than the diabetic control group. This results showed that dandelion extract attenuated changes of reactive oxygen species generating enzyme activities in liver. The activities of SOD, CAT, GSH-Px, GR, G6PD and GST were significantly increased in the diabetic liver. Supplementation of dandelion water and ethylacetate extract was lowerd their activities in the diabetic rats. This results showed that dandelion extracts exerted antioxidant defense system.
8. Hepatic GSH content was significantly decreased in the diabetic control group compared with the normal group, whereas LPO content was increased. In the animals supplemented with dandelion extract, LPO content was significantly lower and GSH content was markedly elevated in liver.
9. In plasma and liver, triglyceride, total cholesterol and free cholesterol contents were higher the diabetic control group than the normal group. Plasma HDL cholesterol content was increased in dandelion extract groups, whereas VLDL cholesterol and LDL cholesterol contents as well as AI were decreased.
10. Light microscopy showed large areas of fatty liver in the diabetic control group. The fatty liver changes in the liver were reduced in the supplementation dandelion extract. In kidney, tuble epithelial cell atrophy and mesangial expansion were observed in the glomerular area of the diabetic control group. These interstitial and glomerular alterations were significantly attenuated in rats from dandelion extracts groups. These results showed that dandelion extract supplementation attenuated histological changes of tissues.
11. Counts of Lactobacillus was increased in dandelion extract groups compared with the diabetic control group, whereas E. coli were decreased. These results showed intestinal microorganism ameliorate intestinal environment.
