However, specific regulators for the ANP secretion have not yet been defined. Histamine is present in cardiac tisssues in micromolar concentration. Intrinsic role for the regulation of atrial ANP secretion is unknown. The present study was designed to define the effect of exogenous histamine on atrial secretion of ANP. Experiments were performed in beating or non-beating perfused rabbit atria. In non-beating atria, histamine inhibited an accentuation of ANP secretion induced by atrial distension-and-reduction in terms of extracellular fluid (ECF) translocation (rate of atrial myocytic ANP release) in a volume change-dependent manner. Chlorpheniramine, an inhibitor of histamine H_1 receptor failed to modulate histamine-induced inhibition of atrial myocytic ANP release. Ranitidine, an inhibitor of histamine H_2 receptor blocked histamine-induced inhibition of atrial ANP release. Dimaprit, an agonist of histamine H_2 receptor inhibited accentuation of ANP release by atrial volume changes. In beating atria, histamine elicited dual effects in atrial ANP secretion, an increase followed by an inhibition. The increase in ANP secretion was coincident with an increase in ECF translocation, while the decrease was closely related with an inhibition in myocytic ANP release. Chlorpheniramine failed to modulate histamine-induced increase in ANP secretion and partially blocked an inhibition in myocytic ANP release. Ranitidine blocked histamine induced changes in ANP secretion, ECF translocation and myocytic ANP release.
