Bioremediation, which is accomplished by adding exogenous microbial populations or stimulating indigenous ones, attempts raise the rates of degradation found naturally to significantly higher rates. Biodegradation rates of diesel by a selected diesel- degrader, Pseudomonas stutzeri strain Y2G1, and microbial consortia composed of 5 selected diesel-degrading bacteria, were determinated in liquid and soil system. The strain Y2G1 degraded diesel at a concentration as high as 12% (v/v) without any inhibition in its diesel-degrading activity. Diesel degradation rates were greatly affected by environmental factors. Optimal conditions for pH, temperature, and moisture content were pH 8, 25℃, 10%, respectively. In soil system experiments, either inoculation, especially in the form of a consortium, or nutrient addition greatly enhanced diesel degradation (1.5 times). Aeration of the soil in column reactors accelerated diesel degradation rate the most effective, from 63.4 mg/kg · day to 162.6 mg/kg · day. In soil system, experiments were conducted for the investigation of bioremediation using Pseudomonas sp. Y2G1, Pseudomonas sp. Y5K2, Sphingomonas sp. D3K1, and Pseudomonas aeruginosa D2D2 in diesel-contaminated soil. Seeding with diesel-degrader has been demonstrated to be effective. When fertilizer (C:N:P=100:10:3) added to soil, diesel degrading rate was improved from 1.2 to 2 times. About the question of any solid peroxide which would promote the aerobic bioremediation of soil, as the solid peroxide was added to the soil, the soil pH was increased. And then these were inhibited diesel degradation by diesel-degrading bacteria. Mixed culture of the four strains was degraded better than single strains in diesel contaminated soil. Higher degradation rate of diesel by consortia was 68% by Y5K2 & D2D2 and 61% by Y2G1 & D2D2. Aeration and fertilizer of the soil in column reactors accelerated the most effective diesel degradation rate from 665 mg/kg · day to 1069 mg/kg · day. From these results, they were concluded that the major rate-limiting factors in the tested diesel-contaminated soil were diesel-degrading microorganisms, inorganic nutrients and aeration. This study clearly implies that the highest bioremediation efficiency can be achieved by mitigating rate-limiting factors through a designed treatment strategy effectively.
