Especially, Monascus pigments have been used in various foods as a natural coloring agent for many East Asian countries. Recently, many kinds of synthetic dyestuff have been found to be hazardous for human health. In many countries only limited kinds of such dyestuff are permitted for use in food processing. Therefore, there is a need to develop alternative source of natural food colorants such as Monascus pigment. Monascus pigments are currently accomplished to research and develop commercially in China, Japan and Taiwan as commonly used food colorants. However, because of the insufficiency of industrial technologies, these pigments have been imported entirely despite of the increasing public concern and demand on natural food colorant for safety in our country. Therefore, modern bioprocess technology, which is heavily reliant on fundamental research such as assay of pigment structure, metabolic pathway and so on, needs to be applied to allow the development of more efficiency for large scale bioreactors and operation policies in industrial scale. In these studies, we can contribute to process model for applying industrial bioprocess technology to natural pigment production by cultivation of Monascus sp. under submerged culture. First, using 30g/L glucose and 1.5g/L MSG(monosodium glutamate) as the carbon and nitrogen source respectively, we developed a chemically-defined medium that could support the rapid and extensive production of the biomass and pigments. A high glucose concentration led to low growth rate, pigment synthesis and considerable ethanol production. In addition to either initial suitable glucose concentration or the use of controlled lactose feeding in fed-batch systems, the use of slowly metabolized carbon and energy substrates may be used to minimize ethanol formation. We also found that the time to adding the precursor amino acid and trace metal was optimal at 84 hrs and beginning of fermentation for red pigment formation. Second, after development of the optimal medium composition and cultural condition, batch-culture in jar fermenter were used to determine the optimal culture conditions for the production of the red pigments by controlling pH and DO(dissolved oxygen) in submerged culture. The level of the dissolved oxygen concentration were significantly increased the metabolic production at the DO 80%. And then pH of 5.5 increased the productivity of biomass and pigment by a factor of 1.4 and 1.9 compared with controlling DO 80%. In a pH range of 4.0 to 8.5, neutral pH favored the production of red pigment and pigment excretion into the medium, whereas the yellow pigment production was favored in acidic pH. A controlled pH change from 4.0 to 7.0 resulted in the highest water-soluble red pigment production. these results indicate that water-soluble red pigment is derived from chemical transformation of orange precursors formed at pH 4.0 to red pigment formed at pH 7.0. Third, based on the optimal production conditions by addition of trace metal and nitrogen source in the batch culture process, fed-batch cultures in jar fermenters were carried out in which three kind of nutrient feeding strategies were applied. When trace metal and MSG were added at the beginning and after 60hr of fermentation in fed-batch culture, respectively, cell concentration, red pigments and water-soluble red pigment was 2.5, 2.4 and 7.1 times higher than that of batch culture. A controlling pH change from 4.0 to 7.0 resulted in a very favorable effect on excretion of the pigments in fed-batch culture, whereas water-soluble red pigment was not achieved effectively as previously results.
