Cell viability was assessed using a MTT assay. When B103 cells were treated in the presence of A(325_35 and A(31-42, AD reduced the survivability of B103 cells. To examine A(3 uptake, we used western blotting and immunocytochemistry methods. They revealed that extracellular AD peptides are taken up B103 cells. Fluorescent intensity of GFP by Fluorescence microscopy campared proteasome activity when cells were treated with proteasome inhibitor and A(3. Fluorescence intensity was increased by proteasome inhibitors and A(3. To verify this, total protein from cell treated with proteasome inhibitors and AD was analyzed by western blotting for the detection of GFP. The level of GFP was increased. We determined the effect of AP on the proteolytic activity of 26S proteasome. We shown that A(3 inhibited the chymotrypsin-like activity of the purified proteasome fraction. The inhibition of the chymotrypsin-like activity by A(3 was dose-dependent. We investigated the proteasome activity and API-42 level in animal model of AD(Tg2576). When API-42 level in one quarter brain was detected by API-4z immunoassay kits, API-42 level was increased with aging. The chymotrypsin-like activity of the purified proteasome fraction from brain(cortex, hippocampus) was decreased with aging. Also, degradation of [3 - catenin as a proteasome substrate was decreased with aging.
